저자들은 하지 자색반과 고환 통증으로 내원한 알코올성 간경화증 환자에서 LCV의 원인 감별하는 과정에서 HCC와 하대정맥 막성폐쇄를 진단하고 이에 대한 치료 후 LCV의 호전된 증례를 경험하였기에 문헌고찰과 함께 보고하는 바이다. Vasculitis is a heterogeneous group of diseases that destroy blood vessel walls by inflammation. Approximately half of vasculitis cases are idiopathic, but sometimes associated with genetic factors, medicines, chronic infection, autoimmune diseases, and malignancies. Although the mechanism remains unclear, vasculitis secondary to malignancy, also known as paraneoplastic vasculitis, has been reported. It is generally associated with hematologic malignancies rather than solid malignancies and commonly presents as leukocytoclastic vasculitis or polyarteritis nodosa. We experienced a case of leukocytoclastic vasculitis in a patient with hepatocellular carcinoma and membranous obstruction of the inferior vena cava. 동절기 단열갱폼으로 인한 경제성 효과 및 투입 원가 분석 연구

This paper verifies the superiority of warming work in winter by applying the insulation gang-form to the apartment housing site and analyzes the economic feasibility of the application. According to the experimental results, the actual cost of warming work was about 52 million won less than planned, and 160 million won less than the existing average.(Note - The cost of gang form material increased from 260 million won to 310 million won after the change) As a result, the construction cost could be reduced by about 110 million won. As the costs of warming work can change depending on the number of floors, the building number, and the area of each site, it is deemed necessary to conduct a thorough review in advance at the site where the cost of warming work is to be applied. 본 논문은 단열갱폼의 공동주택현장 적용을 통해 단열갱폼 동절기 보양우수성을 확인하고, 현장 적용에 따른 경제적 타당성을 분석하고 있다. 실험결과에 따르면 실제 보양투입 비용은 사전계획대비 약 5천 2백만 원을 절감하였으며, 기존 일반갱폼대비 약 1억 6천만 원을 절감하였다.(참고 - 변경전 갱폼 자재비는 2억 6천만 원에서 변경 후 3억 1천만 원으로 약 5천만 원 증가함) 결과적으로 단열갱폼으로 변경하여 약 1.1억 원 가량의 공사비를 절감할 수 있었다. 현장마다 층수, 동수, 면적이 달라짐에 따라 보양공사비가 변경될 수 있으니 단열갱폼 적용 예정인 현장에서는 사전에 충분한 검토가 이루어져야 할 것으로 판단된다.
Effect of α- Glycosidase Inhibitor in Multidrug Resistant Cell Lines

The objective of this study was to evaluate the reversal of multidrug resistance of human cell lines by specific inhibitors of α-glucosidases and mannosidases that had been reported to be involved in N-linked oligosaccharide processing of glycoproteins. N-methyldeoxynojirimycin, 1-deoxynojirimycin, and castanospermine, which were known to be potent inhibitors of both α-glucosidase Ⅰ and Ⅱ, showed no activity against the multidrug resistant phenotype of the cell lines of SNU1DOX, KB-V1, and MCF-7/ADR. In contrast, 1-deoxymannojirimycin, an inhibitor of mannosidase Ⅰ, resulted in a slight reversal for the vinblastine resistance of the KB-V1 cell line, but did not show any activity toward the other cell lines. Parallel experiments with tunicamycin, an inhibitor of N-linked glycosylation, also resulted in no significant changes in multidrug resistant (MDR) phenotype of the cell lines tested in this work. These observations suggest that the unglycosylation of P-glycoprotein associated with the inhibitor treatments might not be correlated with the reversal of multidrug resistance of the cell lines tested in this study.

The pharmacokinetics of CKD-732 (6-0-4-[dimethyl-aminoethoxy)cinnamoyl]-fumagillolㆍhemioxalate) was investigated in male SD rats and beagle dogs after bolus intravenous administration. The parent compound and metabolites obtained from in vitro and in vivo samples were determined by LC/MS. The main metabolite was isolated and identified as an N-oxide form of CKD-732 by NMR and LC/MS/MS. CKD-732 was metabolized into either M11 or others by rapid hydroxylation, demethylation, and hydrolysis. The blood level following the intravenous route declined in first-order kinetics with $T_{1}$2/$\beta$ values of 0.72-0.78 h for CKD-732 and 0.92-1.09 h for M11 in rats at a dose of 7.5-30 mg/kg. In dogs, $T_{1}$2/$\beta$ values of CKD-732 and M11 were 1.54 and 1.79 h, respectively. Moreover, AUC values increased dose dependently for CKD-732 and M11 in rats and dogs. The CLtot and Vdss did not change significantly with increasing dose, indicating linear pharmacokinetic patterns. The excretion patterns through the urine, bile, and feces were also examined in the animals. The total amount excreted in urine, bile, and feces was 2.13% for CKD-732 and 1.29% for M11 in rats, and 1.58% for CKD-732 and 2.28% for M11 in dogs.

X-ray crystallographic studies of the deoxy form of human adult hemoglobin (Hb A) have shown that β99Asp is hydrogen bonded to both α42Tyr and α97Asn in the α₁β₂ subunit interface, suggesting that the essential role of β99Asp is to stabilize the deoxy-Hb by creating the intersubunit hydrogen bond. In particular, for Hb Kempsey (β99Asp→Asn), molecular dynamics simulation indicated that a new hydrogen bond involving β99Asn can be induced by replacing α42Tyr with a strong hydrogen-bond acceptor such as Asp. Designed mutant recombinant (r) Hb (β99Asp→Asn, α42Tyr→Asp) have been produced in the Escherichia coli expression system and have shown that functional defects of Hb Kempsey could be compensated by the α42Tyr→Asp substitution. However, as the α42Tyr→Asp mutation has never been reported before, it is still possible that the functional properties of r Hb (β99Asp→Asn, α42Tyr→Asp) may be due to the mutation itself. Thus, it is required to producer Hb (α42Tyr→Asp) and r Hb Kempsey (β99Asp→Asn) as controls, and to compare their properties with those of r Hb (β99Asp→Asn, α42Tyr→Asp). r Hb (α42Tyr→Asp) could not be purified because it is an unstable hemoglobin which forms Heinz bodies. r Hb Kempsey (β99Asp→Asn) exhibits very high oxygen affinity and greatly reduced cooperativity. Thus, r Hb (β99Asp→Asn) and r Hb (α42Tyr→Asp) compensate each other.

It is well documented that tethered cord is considered to be a main cause of developing progressive motor and sensory changes of the legs and incontinence in young children but the question remains as to the cause of tethering induced dysfunction of the spinal cord. The purpose of this study is to elucidate the changes of regional spinal cord blood flow that might he a main pathophysiological factor of progressive neurological deficit after tethering of the cord and to analyse the effect of untethering at various intervals after fixation of the filum terminate in kittens during their growth.

Methyl mercaptan oxidase was successfully induced in Thiobacillus thioparus TK-m using methyl mercaptan gas, and was purified for the detection of mercaptans. The purification procedure Involved a DEAE (diethylaminoethyl) -Sephacel, or Superose 12, column chromatography with recovery yields of 47.5 and 48.5%, and specific activities of 374 and 1240.8 units/mg-protein, respectively, The molecular weight of the purified methyl mercaptan oxidase was 66.1kDa, as determined by SDS-PAGE. The extract, from gel filtration chromatography oxidizes methyl mercaptan, producing formaldehyde, which can be easily detected by the purpald-coloring method. The optimized temperature for activity was found to be at 55$\^{C}$. This enzyme was inhibited by both NH$_4$Cl and (NH$_4$)$_2$SO$_4$, but was unaffected by either KCl or NaCl at less than 200 mM. With K$_2$SO$_4$, the activity decreased at 20 mM, but recovered at 150 mM. In the presence of methanol, full activity was maintained, but decreased in the presence of glycerin, ethanol and acetone 43, 78 and 75%, respectively.

Aminoacyl-tRNA synthetases are essential enzymes catalyzing the attachment of specific amino acids to cognate tRNAs. In the present work, the substrate analogue L-methionine hydroxamate was used to identify functional residues located in the active site of the E. coli methionyl-tRNA synthetase (MetRS). This compound inhibited bacteria, yeast, and human MetRS activities to a similar degree, suggesting a conserved active site structure and mechanism between MetRSs of different phylogenetic domains. Mutants of the E. coli MetRS resistant to methionine hydroxamate were also isolated. These mutants contained a substitution either at T10, Y15, or Y94. These residues are highly conserved among the different MetRSs and the mutants showed decreased aminoacylation activity, suggesting their functional and structural significances. The putative roles of these residues are discussed on a structural basis.
