Objective: Staphylococcus aureus (S. aureus) is one of the major microorganisms responsible for subclinical mastitis in dairy cattle. The present study was designed with the aim to explore the DNA methylation patterns using the Fluorescence-labeled methylation-sensitive amplified polymorphism (F-MSAP) techniques in a S. aureus-infected mouse model. Methods: A total of 12 out-bred Institute of Cancer Research female mice ranging from 12 to 13 weeks-old were selected to construct a mastitis model. F-MSAP analysis was carried out to detect fluctuations of DNA methylation between control group and S. aureus mastitis group. Results: Visible changes were observed in white cell counts in milk, percentage of granulocytes, percentage of lymphocytes, CD<sup>4+</sup>/CD<sup>8+</sup> ratio (CD<sup>4+</sup>/CD<sup>8+</sup>), and histopathology of mice pre- and post-challenge with S. aureus. These findings showed the suitability of the S. aureus-infected mouse model. A total of 369 fragments was amplified from udder tissue samples from the two groups (S. aureus-infected mastitis group and control group) using eight pairs of selective primers. Results indicated that the methylation level of mastitis mouse group was higher than that in the control group. In addition, NCK-associated protein 5 (Nckap5) and transposon MTD were identified to be differentially methylated through secondary polymerase chain reaction and sequencing in the mastitis group. These observations might play an important role in the development of S. aureus mastitis. Conclusion: Collectively, our study suggests that the methylation modification in Nckap5 and transposon MTD might be considered as epigenetic markers in resistance to S. aureus-infected mastitis and provided a new insight into S. aureus mastitis research in dairy industry and public health.

The innate immune system coordinates the inflammatory response to pathogens. To do so, its cells must discriminate self from non-self utilizing receptors that identify molecules synthesized exclusively by microbes. Toll-like receptors have a crucial role in the detection of microbial infection in mammals and insects. In mammals, they have evolved to recognize conserved products unique to microbial metabolism. These include lipopolysaccharide (LPS), lipotechoic acids, and peptidoglycans (PGN). We show here that TLRs, including TLR2, are expressed on the THP-1 human leukemia cell line. Activation of TLR2 signaling in THP-1 by PGN induces the synthesis of various soluble factors and proteins including interleukin-$1{\beta}$, interleukin-8 and TNF-${\alpha}$ and apoptosis of THP-1 with PGN dose and time dependence. Moreover, in this study we show that PGN induces apoptosis of THP-1 cells in a TNF-${\alpha}$-dependent manner. These findings indicate that TLR2 signaling results in a cascade leading to tumor apoptosis and differentiation, which may suggest new clinical prospects using TLR2 agonists as cytotoxic agents in certain cancers.
FEEDING RICE STRAW SUPPLEMENTED WITH UREA-MOLASSES LICK BLOCK TO LACTATING COWS IN BHUTAN

Twelve cross bred cows ($300{\pm}20kg$) were fed a basal ration consisting of 1 kg concentrate and untreated or urea treated rice with or without urea-molasses-wheat bran lick block supplementation. The lick blocks were prepared locally using cement as a binding agent. The experiment lasted for 48 days consisting of a preliminary period of 14 days and a measurement period of 34 days. Daily dry ma tter intake (DMI) of straw and lick block, and daily milk yield were recorded during the measurement period. Intake of lick block when fed with untreated straw (US) was significantly higher (p < 0.01) than with urea treated (TS) straw (397 vs 307g $100kg^{-1}$), but lick block supplementation did not significantly affect the in take of US (1.80 vs 1.83kg $100kg^{-1}day^{-1}$) or TS (2.27 vs 2.17kg $100kg^{-1}day^{-1}$). Both urea treatment and lick block supplementation significantly increased (p < 0.01) the milk yield of cows, and the increase due to urea treatment was higher than that due to lick block supplementation. Benefit/cost ratio obtained for feeding US or TS with 1 kg dairy concentrate was similar (5.4), but the marginal return favours TS (5.4:1.0). The benefit/cost ratio for US and TS supplemented with lick block was 3.4 and 3.7. respectively, but the marginal return with US was higher than with TS (1.6:1.0 and 0.9:1.0, respectively).

Objective: This review consolidates the available information on the socio-economic and ecological significance of Mithun in the lives of ethnic communities in the Eastern Himalaya. Methods: Standard guidelines were followed for the review and data collection was carried out at three stages; literature search, literature screening, and literature review and analysis. Results: Records indicate a long association of Mithun with the ethnic groups. Mithun serves as a symbol of pride and local currency for barter trade in the ethnic society. Its utilities range from being used as a bride price to settling legal disputes. Several cultural festivals and local ceremonies are celebrated around this bovine. Due to its semi-wild nature, this animal also has an ecological role to conserve broad leaf sub-tropical forests. However, it remains neglected and has not received policy attention, leading to a stagnated growth. The institutions for Mithun research and development are also weak. Furthermore, the species is under threat from new diseases and habitat alteration triggered by climate change. Conclusion: Founded on the current state of knowledge, there is a need for institutional development, strengthening institutional linkages, and promoting regional cooperation among Mithun rearing countries for further research and development of this unique cattle.
