        Molecular characterization of chemosensory protein genes in Bactrocera minax (Diptera: Tephritidae)

        Chemoreception is of great importance for survival of insects. Chemosensory proteins (CSPs) are believed to be involved in the perireceptor events of chemosensory system in many insect species, but this has not been clarified in the citrus fruit fly, Bactrocera minax, a serious dipteran pest of citrus crops. Previous studies indicated that four CSP genes were identified in many fly speices in Diptera. In the present study, we also identified four CSP genes in B. minax,namely BminCSP1–4, from a transcriptome database. All CSP proteins encoded by these genes bear the typical hallmarks of the CSP family: an N-terminal signal peptide and the four highly conserved cysteine residues. Phylogenetic analysis comparing with other dipteran CSPs indicated that dipteran CSPs may evolved from three ancestral CSP genes, and revealed the sequence diversities of BminCSPs and showed that BminCSP1–4 are clustered in separate groups, indicating the possibility of their contrasting function in B. minax. Furthermore, the tissue distribution of the four CSP genes in adult B. minax was analyzed by real-time quantitiative PCR. The results demonstrated that BminCSP3 are significantly transcriptionally enriched in antennae; BminCSP4 expressed primarily in heads; BminCSP1 and BminCSP2 showed high expressions in different tissues, such as antennae, abdomens and wings. Based on these findings, the different implications for the functions of BminCSPs are discussed. This study will offer a significant indication for further functional studies of the CSPs in B. minax

        Mechanisms of Extracellular NO and Ca2+ Regulating the Growth of Wheat Seedling Roots

        Our previous studies suggested the cross talk of nitric oxide (NO) with Ca2+ in regulating stomatal movement. However, its mechanism of action is not well defined in plant roots. In this study, sodium nitroprusside (SNP, a NO donor) showed an inhibitory effect on the growth of wheat seedling roots in a dose-dependent manner, which was alleviated through reducing extracellular Ca2+ concentration. Analyzing the content of Ca2+ and K+ in wheat seedling roots showed that SNP significantly promoted Ca2+ accumulation and inhibited K+ accumulation at a higher concentration of extracellular Ca2+, but SNP promoted K+ accumulation in the absence of extracellular Ca2+. To gain further insights into Ca2+ function in the NOregulated growth of wheat seedling roots, we conducted the patch-clamped protoplasts of wheat seedling roots in a whole cell configuration. In the absence of extracellular Ca2+,NO activated inward-rectifying K+ channels, but had little effects on outward-rectifying K+ channels. In the presence of 2 mmol L−1 CaCl2 in the bath solution, NO significantly activated outward-rectifying K+ channels, which was partially alleviated by LaCl3 (a Ca2+ channel inhibitor). In contrast,2 mmol L−1 CaCl2 alone had little effect on inward or outward-rectifying K+ channels. Thus, NO inhibits the growth of wheat seedling roots likely by promoting extracellular Ca2+ influx excessively. The increase in cytosolic Ca2+ appears to inhibit K+ influx, promotes K+outflux across the plasma membrane, and finally reduces the content of K+ in root cells.

      • Measurement of neutron cross sections and resonance parameters of <sup>169</sup>Tm below 100 eV

        <P>The neutron total cross-sections of thulium (<SUP>169</SUP>Tm) were measured in the neutron energy region from 0.01 eV to 100 eV by using the time-of-flight method at the Pohang Neutron Facility, which consists of an electron linac, a water-cooled tantalum target with a water moderator, and a 12 m time of flight path. Two thulium plates with different thicknesses were used for the neutron transmission measurement. The background level was determined by using a notch-filter of Co, In, and Cd sheets. The present measurement was compared with the previous ones, and a new set of resonance parameters of <SUP>169</SUP>Tm isotope was obtained from the transmission rate by using the SAMMY code, with a comparison with the recommended parameters by Mughabghab.</P>

        Effects of chelated Zn/Cu/Mn on redox status, immune responses and hoof health in lactating Holstein cows

        To evaluate the effects of chelated Zn/Cu/Mn on redox status, immune responses and hoof health in lactating Holstein cows, 48 head in early lactation were divided into healthy or lame groups according to their gait score. Cows were fed the same amount of Zn/Cu/Mn as sulfate salts or in chelated forms for 180 days, and foot-and-mouth disease (FMD) vaccine was injected at day 90. The results showed that lame cows had lower antioxidant function, serum Zn/Mn levels, hair Cu levels, and hoof hardness. Moreover, increased antioxidant status, FMD antibody titers, serum and hair levels of Zn/Cu/Mn, and hoof hardness and decreased milk fat percent and arthritis biomarkers were observed in cows fed chelated Zn/Cu/Mn. In summary, supplementation with chelated Zn/Cu/Mn improved antioxidant status and immune responses, reduced arthritis biomarkers, and increased accumulation of Zn/Cu/Mn in the body and hoof hardness in dairy cows.

      • Expression of Connexin 43 and E-cadherin Protein and mRNA in Non-small Cell Lung Cancers in Chinese Patients

        Aim: Connexin 43 (Cx43) and E-cadherin are important biomarkers related with cancer. Their expression at protein and mRNA levels was here investigated in 50 primary lung carcinoma tissues and 20 samples of adjacent normal tissue of Chinese patients with non-small cell lung cancer (NSCLC). Methods: Protein and mRNA expression were evaluated by ABC immunohistochemistry and RT-PCR. Results: (1) The positive expression rates of Cx43 and E-cadherin protein were higher in the adjacent normal tissues than those in the primary lung carcinoma tissues; (2) the positive expression rates of Cx43 and E-cadherin protein decreased with NSCLC progression; (3) the expression of E-cadherin protein was not related with the pathological type of NSCLC; and (4) the relative quantity of the Cx43 or E-cadherin mRNA expression was correlated with the the histological type, clinical stage, cancer cell differentiation and the lymph node metastasis. Conclusion: The data suggested that the Cx43 and E-cadherin are reduced with NSCLC progression, and might be important biomarkers for judging the metastasis and prognosis.

        Gonadotropins Improve Porcine Oocyte Maturation and Embryo Development through Regulation of Maternal Gene Expression

        The present study assessed the effect of FSH and LH on oocyte meiotic, cytoplasmic maturation and on the expression level and polyadenylation status of several maternal genes. Cumulus-oocyte complexes were cultured in the presence of FSH, LH, or the combination of FSH and LH. Significant cumulus expansion and nuclear maturation was observed upon exposure to FSH alone and to the combination of FSH and LH. The combination of FSH and LH during entire IVM increased the mRNA level of four maternal genes, C-mos, Cyclin B1, Gdf9 and Bmp15, at 28 h. Supplemented with FSH or LH significantly enhanced the polyadenylation of Gdf9 and Bmp15; and altered the expression level of Gdf9 and Bmp15. Following parthenogenesis, the exposure of oocytes to combination of FSH and LH during IVM significantly increased cleavage rate, blastocyst formation rate and total cell number, and decreased apoptosis. In addition, FSH and LH down-regulated the autophagy gene Atg6 and upregulated the apoptosis gene Bcl-xL at the mRNA level in blastocysts. These data suggest that the FSH and LH enhance meiotic and cytoplasmic maturation, possibly through the regulation of maternal gene expression and polyadenylation. Overall, we show here that FSH and LH inhibit apoptosis and autophagy and improve parthenogenetic embryo competence and development.

      • Screening of MicroRNA in Patients with Esophageal Cancer at Same Tumor Node Metastasis Stage with Different Prognoses

        Patients at the same pathological stage of esophageal cancer (EC) that received the same surgical therapy by the same surgeon may have distinct prognoses. The current study aimed to explore the possibility of differentially-expressed microRNAs (miRNAs) underlying this phenomenon. Samples were collected from EC patients at the same tumor node metastasis (TNM) stage but with different prognoses. Paracancerous normal tissues were taken as controls. The specimens were histopathologically analyzed. Differentially-expressed miRNAs were analyzed using real-time quantitative reverse transcription polymerase chain reaction. Compared with patients with poor prognosis, those with good prognosis exhibited 88 two-fold or more than two-fold increased miRNA fragments and 4 half-decreased miRNAs. The most noticeably up-regulated miRNAs included hsa-miR-31, hsa-miR-196b, hsa-miR-652, hsa-miR-125a-5p, hsa-miR-146b, hsa-miR-200c, hsa-miR-23b, hsa-miR-29a, hsa-miR-186, hsa-miR-205, hsa-miR-376a, hsa-miR-410, hsa-miR-532-3p, and hsa-miR-598, whereas the most significantly-downregulated miRNAs were hsa-let-7e, hsa-miR-130b, and hsa-miR-103. EC patients at same TNM stage but with different prognoses show differentially-expressed miRNAs.

      • Values of Three Different Preoperative Regimens in Comprehensive Treatment For Young Patients with Stage Ib2 Cervical Cancer

        Objective: To compare the clinical efficacy of concurrent chemoradiotherapy, neoadjuvant chemotherapy, and intracavity brachytherapy in comprehensive treatment for young patients with stage Ib2 cervical cancer. Methods: One hundred and twelve young patients with stage Ib2 cervical cancer were enrolled retrospectively in our hospital from January 2003 to June 2005. They were categorized into three groups according to preoperative regimens, including the concurrent chemoradiotherapy group (Group 1, n=38), the neoadjuvant chemotherapy (Group 2, n=49), and the intracavity brachytherapy group (Group 3, n=25). Radical hysterectomy was performed following these regimens. Chemotherapy and radiotherapy were given according to pelvic lymph node metastasis, deep cervical stromal invasion, intravascular cancer emboli, histological grading, vaginal stump and positive surgical margin. Results: The cancer disappearance and superficial muscle invasion rates were statistically significantly better in the concurrent chemoradiotherapy group than in the other two groups (P<0.01). No statistically significant difference was noted in the deep muscle invasion rate, surgical time and intraoperative blood loss among three groups, but significantly more postoperative complications occurred in the concurrent chemoradiotherapy group. The 2-year pelvic recurrence was statistically significantly lower in the concurrent chemoradiotherapy group compared to other two groups, while the 5-year survival was higher. Conclusion: Concurrent chemoradiotherapy is efficacious for young patients with stage Ib2 cervical cancer.

        The Expression Characterization of Chicken Uncoupling Protein Gene

        The UCPs are members of the mitochondrial inner membrane transporter family, present in the mitochondrial inner membrane. Their main function is increasing the energy expenditure via diminishing the resulting production of ATP from mitochondrial oxidative phosphorylation instead of yielding dissipative heat. They are associated with the metabolism of fat and regulation of energy expenditure. The UCP gene can be viewed as the candidate gene for chicken fatness. In the present study, RT-PCR and Northern Blot methods were developed to investigate the expression of the UCP gene in ten tissues including heart, liver, spleen, lung, kidney, gizzard, intestine, brain, breast muscle and abdominal fat of chicken. The results of both RT-PCR and Northern Blot methods showed that the UCP gene expressed specific in breast muscle. The expression levels of UCP gene in breast muscles from egg-type and meat-type chickens of hatching, 2, 4, 6 and 8 wk of age were detected by RT-PCR assay and results showed that the expression levels of UCP gene were related to breeds. Expression level of UCP gene in layers was higher than that in broilers at various weeks of age except at 6 wk. The UCP gene's expression was higher at 6 wk and had no significant difference among other weeks of age in broilers; in layers the expression level of UCP gene had no significant difference among weeks of age. The experiment results also showed that insulin could increase the expression level of UCP gene by 40% compared with control group.
