Characterization of stem rot disease caused by Botryosphaeria dothidea on plum tree in Korea

Botryosphaeria dothidea(B. dothidea) is a causal agent is commonly associated with stem canker, dieback, fruit and stem rot disease on various hosts worldwide. Typical symptoms of a small blister and brown-stained lesion usually appeared on the braches or stems of infected plants. In Korea, B. dothidea has been reported to causes a significatnt problem to several commercial crops including stone fruits. In early 2020, a symptom that sesembled B. dothidea infection was spotted on a plum tree in the cultivation area around Wanju-gun. Samples of symptomatic plum tissues were collected and causal agent was purified first on water agar followed by potato dextrose agar. Initial identification via morphological attribute shows that 15 fungal isolates show strong resemblances to B. dothidea. Molecular identification based on DNA sequences of three molecular markers i.e. Ribosomal DNA internal transcribed spacer, beta-tubulin, and elogation facter-1 alpha confirms the morphological result above. BLASTN and phylogenetic analysis results show that the fungal pathogen belongs to B. dothidea family group. The pathogenicity test of four selected isolates, which were performed on the detached stems, confirms that these isolates are pathogenic to plum trees. Therefore, in this study, we reported to have characterized B. dothidea as a causal agent to stem rot disease occur in the plum tree in Korea.
양파 마른썩음병 원인균 동정과 약제배지를 균사생장 억제효과 검정

양파는 우리나라에서 재배하는 겨울철 작물 중 하나로 그 재배적 가치가 매우 높다. 양파는 흑색썩음균핵병, 마른썩음병(시들음병), 무름병 등이 발생하면 수확량이 감소하게 된다. 본 연구에서는 양파 마른썩음병의 원인균을 분리하여 병원성 검정, 종 동정을 실시하였다. 그 결과, Fusarium oxysporum과 F. commue균을 분리하였고, 이 중 F. oxysporum이 병원성이 높은 것으로 조사되었다. 양파에서 분리한 마른썩음병균의 균사 생장억제효과를 검정하기 위하여 양파에 사용되는 6종의 살균제로 약제 희석배지를 제조하고 이를 이용하여 균사생장 억제효과를 검정하였다. 그 결과, 프로클로라즈망가니즈 등 2종 이상의 살균제에서 마른썩음병균의 균사생장을 효과적으로 억제하는 것으로 확인되었다. 현재, 양파 마른썩음병 방제용 살균제가 전무한 상태이므로, 향후 포트시험 등을 통한 양파 마른썩음병 방제효과를 검정하고, 포장시험 등을 통해서 양파 마른썩음병 방제용 살균제를 등록이 필요할 것으로 판단된다.

Leaf blight disease caused by Stemphylium species is an important disease in Allium crops, specifically onion, garlic and welsh onion. In 2018, leaf blight symptoms were severe and damaged onion and garlic in Jeonnam province in Korea. In addition, small purple spots on garlic burbs were observed in a post-harvest storage warehouse. Several Stemphylium isolates were isolated from diseased leaves from the field and from garlic bulb samples and were analyzed in terms of homology and the phylogenetic relationship based on the internal transcribed spacer region and calmodulin gene sequence. The results showed that among three Stemphylium species identified, S. vesicarium is most prevalent on onion and garlic. S. eturmiunum was for the first time identified as pathogenic to onion and garlic, whereas S. solani was found in welsh onion crops. Although these isolates grew well at the optimum temperature at 20 - 25℃, they could also grow at low temperatures of 10 - 15℃. A pathogenicity test was conducted using S. vesicarium and S. eturmiunum on onion and garlic respectively. These results showed that two Stemphylium species were highly virulent with cross pathogenicity in onion and garlic. The results of this study can support the biological characterization of Stemphylium species in Korea. Moreover, further research will need to develop fungicide application strategies for onion and garlic crops.

Fusarium wilt caused by Fusarium oxysporum and crown and foot rot caused by F. solani can result in serious economic losses on several cucurbitaceous crops, such as watermelon and cucumber in Korea. These diseases associated with soil-borne pathogens mainly induce wilting symptoms. They are extremely difficult to control and there are few effective management strategies to reduce damages. Information related to control efficacy of antifungal agents against Fusarium oxysporum and F. solani on inducing wilting symptoms on cucurbits is insufficient. In this study, 108 fungicides registered for controlling fungal disease occurring on watermelon and cucumber were tested to evaluate growth inhibition of Fusarium oxysporum and F. solani on potato dextrose agar medium. Two fungicides, cabendazim + kasukamycin and prochloraz manganese, forming large growth inhibition zone on medium showed against the two species of Fusarium. In the case of seedlings, the same results as mycelium inhibition test was shown. In further study, experimental trials will be carried out to investigate control efficacy of the promising fungicides against Fusarium wilt and crown and foot rot diseases on watermelon and cucumber in field conditions.
