Single cell gel(SCG) electrophoresis or 'Comet assay' is a rapid and very sensitive fluorescent microscopic method to examine DNA damage and repair at individual cell level. This assay has critically important applications in fields of toxicology ranging from aging and clinical investigations to genetic toxicology and molecular epidemiology. Since the introduction of the alkaline Comet assay in 1988, a number of advancements have greatly increased the flexibility and utility of this technique for detecting various forms of DNA damage (e.g., single- and double-strand breaks, oxidative DNA base damage, and DNA-DNA/DNA-protein/DNA-Drug crosslinking) and DNA repair in virtually any eukaryotic cell.

<P> Nitric oxide (NO) plays an important role in anoxic preconditioning to protect the heart against ischemia-reperfusion injuries. The present work was performed to study better the NO-cGMP-protein kinase G (PKG) signaling pathway in the activation of both sarcolemmal and mitochondrial ATP-sensitive K<SUP>+</SUP> (KATP) channels during anoxic preconditioning (APC) and final influence on reducing anoxia-reperfusion (A/R)-induced cardiac damage in rat hearts. The upstream regulating elements controlling NO-cGMP-PKG signal-induced KATP channel opening that leads to cardioprotection were investigated. The involvement of both inducible and endothelial NO synthases (iNOS and eNOS) in the progression of this signaling pathway was followed. Final cellular outcomes of ischemia-induced injury after different preconditioning in the form of lactate dehydrogenase release, DNA strand breaks, and malondialdehyde formation as indexes of cell injury and lipid peroxidation, respectively, were investigated. The lactate dehydrogenase and malondialdehyde values decreased in the groups that underwent preconditioning periods with specific mitochondrial KATP channels opener diazoxide (100 μM), nonspecific mitochondrial KATP channels opener pinacidil (50 μM), S-nitroso- N-acetylpenicillamine (SNAP, 300 μM), or β-phenyl-1, N<SUP>2</SUP>-etheno-8-bromoguanosine-3′,5′-cyclicmonophosphorothioate, Sp-isomer (10 μM) before the A/R period. Preconditioning with SNAP significantly reduced the DNA damage. The effect was blocked by glibenclamide (50 μM), 5-hydroxydecanoate (100 μM), N<SUP>G</SUP>-nitro-l-arginine methyl ester (200 μM), and β-phenyl-1, N<SUP>2</SUP>-etheno-8-bromoguanosine-3′,5′-cyclic monophosphorothioate, Rp-isomer (1 μM). The results suggest iNOS, rather than eNOS, as the major contributing NO synthase during APC treatment. Moreover, the PKG shows priority over NO as the upstream regulator of NO-cGMP-PKG signal-induced KATP channel opening that leads to cardioprotection during APC treatment. </P>

<P>La<SUB>1-x</SUB>Sr<SUB>x</SUB>MnO<SUB>3</SUB> (x &equal; 0.1, 0.2, and 0.3) nanoparticles (LSMO NPs) were successfully synthesized by a simple sonochemical-assisted method. The prepared samples were investigated by X-ray diffraction (XRD), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). The compositions of samples were determined by energy-dispersive spectroscopy (EDS) analysis. The temperature dependence of the resistivity of samples was measured using four-probe technique, and the magnetic measurement was performed in a Magnetic Property Measurement System (MPMS XL 7.0). SEM, TEM, and XRD show that LSMO NPs are 40∼80 nm in diameter with a very clean surface, and exhibit single phase perovskite structure. The ferromagnetic transition temperature (T<SUB>c</SUB>) and metal-insulator transition temperature (T<SUB>p</SUB>) of the NPs increase with the doping level x. The maximum values of T<SUB>c</SUB> (377 K) and T<SUB>p</SUB> (264 K) occur at x &equal; 0.3. In this work, the effect of strontium doping level on electrical transport and magnetic properties was systematically studied.</P>

This study investigated the effect of the powder produced by ball-milling the outer layer of annatto (Bixa orellana L.) seeds on the physicochemical properties as well as the antioxidant and antimicrobial activities of pork patties over 14 d of refrigerated storage (4±1℃). Five pork patty treatments were produced containing three different concentrations of annatto seeds, 0.1, 0.25 and 0.5% (ANT0.1, ANT0.25, ANT0.5), 0.1% ascorbic acid (AA0.1), and a control (CTL). Based on the results, annatto seed powder appeared to show antioxidant activity. The Hunter color values of pork patties were affected by the addition of annatto seed powder, which increased the redness and yellowness values, but decreased the lightness of the patties (p<0.05). To evaluate the antioxidative effects of annatto on pork patties, thiobarbituric acid reactive substances (TBARS) and peroxide values (POV) were analyzed over 14 d of refrigerated storage. Treatments containing annatto seed showed lower TBARS and POV than control (CTL) samples (p<0.05). The volatile basic nitrogen (VBN) of the pork patties containing annatto seeds were lower than that of CTL at the end of storage (p<0.05). Although no differences in total bacterial counts were observed between control and treated patties, those containing annatto seeds had lower microbial counts for Enterobacteriacease than CTL or AA 0.1%. Therefore, annatto seed powder might be a good source of natural antioxidants for the production of meat products.
Formability and Effect of Hole Bridge in the Single Point Incremental Forming

This paper studies the mechanical properties, formability and effect of hole bridge (hole lancing) in the single point incremental forming for Al5052-O with the thickness of 0.8 mm. The mechanical properties, including anisotropy property and stress-strain relation, were evaluated by uniaxial tensile tests in three directions of 0°, 45°, 90° with the rolling direction. Varying wall angle conical frustum test was used to study the formability. The forming parameters of tool diameter, step depth, spin speed, and feed rate were optimized using multi-objective optimization with grey relational grade. The results show that the tool diameter strongly influences formability, springback and thickness reduction. To verify the effect of hole bridge on the formability, a predesigned hole bridge was machined by a laser on the blank shoulder nearby the forming periphery before forming. The optimized forming parameters found in previous step were applied for this test. As the result, the maximum forming angle was significantly improved in comparison with the non-hole-bridge part. Finite element simulation by ABAQUS explicit software was used to explain the strain evolution in the blank with cut-off hole of the incremental forming process. The simulation results were compared with experiment and discussed.

허혈전처치는 짧은 시간의 허혈과 재관류를 미리 반복해서 실시하면 다음에 오는 장시간의 허혈 및 재관류에 의한 심장기능의 저하, 부정맥의 발생 및 심근세포의 손상이 현저히 완하되는 현상이다. 내인성 심장보호기전으로서의 허혈전처치는 여러 가지의 가능한 작용기전들이 제시되어왔다. 지금까지 알려진 가능한 기전으로Reduced energy demand, mito-chondrial ATPase inhibitor protein, free radicals, stress proteins, prostacyclin, nitricoxide(NO), adenosine, glycolytic flux, ATP-sensitive potassium channel(K_(ATP)-통로)등이 알려져 왔다. 이 중에서 K_(ATP)-통로는 허혈전처치의 end effector로서 중요한 역할을 담당하는 것으로 알려지고 있다. 하지만 허혈전처치의 심장보호효과의 중요 기전으로서 심장 세포막 K_(ATP)-통로의 상대적인 중요성에 대해서는 아직도 논란의 여지가 많은 실정이다. Single-cell gel electrophoresis 방법을 이용하여 허혈전처치의 심장보호효과와 심장 세포막 K_(ATP)-통로 사이의 상호연관성을 규명하고 그 중요성을 밝히는 전략을 살펴보고자 한다. 허혈전처치의 심장보호기전으로서 심장 세포막 K_(ATP)-통로의 역할을 명확히 규명하기 위한 적절한 실험전략의 수립을 통해, 허혈성 심장질환과 관련된 기초의학적 그리고 임상의학적 연구분야에 중요한 실험적 자료가 될 것으로 사료된다.
Chemical constituents and antioxidant activity of Ficus callosa.

<P>One new megastigmane glycoside, ficalloside (1), and eleven known compounds, were isolated from methanol extract of Ficus callosa leaves by repeated column chromatography. Their structures were established on the basis of spectral and chemical evidence. The antioxidant activities of these compounds were measured using the oxygen radical absorbance capacity (ORAC) assay. Compound 8 exhibited potent antioxidant activity of 10.6 microM trolox equivalents at the concentration of 2 microM. At this concentration, compounds 4-7 and 9-12 showed significant antioxidant activity with ranging of 2.1-6.1 microM trolox equivalents.</P>
