        Ultrasound-Assisted Extraction of Flavonoids and Phenolic Compounds from Ocimum tenuiflorum Leaves

        Ultrasound-assisted extraction (UAE) was optimized for extraction of flavonoids and phenolics from dried leaves of Ocimum tenuiflorum using response surface methodology with a central composite rotatable design and independent variables of ethanol concentration (30-70%), sonication time (5-15 min), and ultrasonic energy density (UED) (0.15-0.35 W/cm3). A significant (p<0.0001) quadratic model described experimental data with a non-significant lack of fit (p>0.05). Linear and quadratic effects of independent variables significantly (p<0.001) influenced flavonoid content (FC) and total phenolic content (TPC) yields. Optimized UAE conditions of ethanol concentration, sonication time, and UED were 55.34%, 11.71 min, and 0.26 W/cm3, respectively, resulting in predicted FC and TPC yields of 6.69 mg of quercetin equivalents QE/g and 9.41 mg of gallic acid equivalents GAE/g, respectively. HPLC analysis revealed different phenolic and flavonoid compounds. UAE extracts exhibited better in vitro antioxidant activities and thermo-oxidative stability values than conventional solvent extracts.

        Energy Requirements and Tillage Performance Under Different Active Tillage Treatments in Sandy Loam Soil

        Purpose To assess the operational energy requirement (OER) and tillage performance index (TPI) of four different active tillage treatments and identification of the best efficient tillage practice among them to achieve an alternative to the conventional practice T1 (1×Cultivator+2×Offset disc harrow), which involves only purely passive type of tillage tools. Methods The active tillage treatments considered were as follows: T2 (1×Combined offset disc harrow), T3 (1×Cultivator+ 1×Single rotor type rotavator), T4 (1×Double rotor type rotavator), and T5 (1×Power harrow). Each implement in the respective treatment was operated at their best suitable forward speed and rotor rpm. Results Compared to the conventional practice T1, the amount of fuel saved per hectare in tillage practices, T2, T4, and T5, was 49.8%, 17.9%, and 20.3%, respectively. However, tillage treatment T3 was found to consume 6.9% more fuel. Compared to T1, the amount of time saved by T2, T3, T4, and T5 tillage practices was found to be 61.1%, 9.54%, 40.6%, and 44.6%, respectively. The maximum OER of 1445.32 MJ ha−1 was observed in T3 followed by 1360.40, 1105.86, 1085.86, and 681.36 MJ ha−1 in T1, T4, T5, and T2, respectively. Further, compared to T1, the TPI for tillage treatments, T2 and T4, was found to be higher by 74.07% and 133.33%, respectively. However, the TPI for tillage treatment T3 was found to be lower by 88.88%. Conclusions Active tillage implements helped in time and fuel-saving in seedbed preparation. The maximum percentage of operational energy was consumed in the form of fuel energy. The front active and rear passive configuration of combined offset disc harrow was found to be the most effective among all the tested active tillage implements in terms of both operational energy and tillage performance criteria.

        Phytochemical composition, in vitro bioactivity evaluation and in silico molecular docking study of fruit essential oils of Zanthoxylum armatum DC collected from Himalayan region of Uttarakhand, India

        The present research aimed to analyze the nematicidal and insecticidal activity of the essential oil obtained from the fruits of Zanthoxylum armatum collected from Gumma village of Dharchula and Chandak village of Pithor agarh. After extraction via hydro distillation, the essential oils were subjected to GC–MS for their compositional analysis. A total of 10 and 13 components were identified for the Dharchula and Pithoragarh collection respectively. Linalool (32.86 %) was the major component in collection from Dharchula whereas it was totally absent in that of collection from Pithoragarh which has trans-sabinene hydrate (11.7 %) as its major component. Essential oils were tested against Meloidogyne incognita for their nematicidal activity and inhibition of egg hatching in laboratory conditions. It was found that the activity of Z. armatum from Dharchula shows high mortality of Meloidogyne incognita and more inhibition to the egg hatching. When tested for insecticidal activity against Lipaphis erysimi, collection from Dharchula presented stronger activity compared to Pithoragarh collec tion as shown by their LC 50 values 0.006 and 0.165 µL/mL respectively after 36 hrs. The in silico PASS study and molecular docking study was conducted to screen the nematicidal and insecticidal potential of the major com ponents on acetylcholinesterase receptors. Results showed good binding affinities and attributed the strongest inhibitory effect to verbenone for the target protein.
