        Evaluation of Ascorbic Acid Treatment in Clinical and Subclinical Mastitis of Indian Dairy Cows

        A study was carried out to assess the therapeutic effect of ascorbic acid in mastitis of dairy cows. The herd with a population of 250-275 lactating cows was screened for clinical and subclinical mastitis for a period of 5 months. Based on inclusion and exclusion criteria, eighteen animals each with clinical and subclinical mastitis in one quarter only were selected as study population. Twelve cows (group A) with normal udder and health were also selected as a healthy control. Clinical mastitis cows were grouped as B (n=12) and C (n=6). Cows of group B were treated with ascorbic acid at 25 mg/kg, subcutaneously for 5 consecutive days and intramammary infusion (Ampicillin sodium 75 mg and Cloxacillin sodium 200 mg/infusion) based on antibiotic sensitivity test, till complete recovery. Group C cows received only intramammary infusion till the complete recovery. Eighteen subclinical mastitis cows were divided in group D (n=12) and E (n=6). Cows of group D were treated with ascorbic acid at 25 mg/kg subcutaneously for 5 consecutive days while group E did not receive any treatment. California mastitis test (CMT), somatic cell count (SCC), physical changes of udder and milk were used to diagnose and classify the mastitis. Evaluation of the therapy was based on CMT score and physical changes of udder and milk. Sample size calculation was also performed but was not followed for control groups due to scarcity of cases. Adequate blinding was done when and where required to avoid the biases. Confounding variables like herd, age of the cow, stage of the lactation, season and geographical region were duly considered and adequate blocking was followed. Ascorbic acid was administered in clinical and subclinical cases even after cure considering its immunostimulatory and healing inducing effects. The recovery rate was faster in cases of clinical mastitis treated with ascorbic acid along with an intramammary infusion (group B) than the quarters of group C cows. Quarter wise the average duration/number (3.16${\pm}$0.11 days) of antimicrobial intramammary infusion was significantly (p<0.01) less in group B than that of average duration/number (5.33${\pm}$0.20 days) of group C. Subclinical mastitis cows treated with ascorbic acid showed 83.33% recovery while 16.77% did not respond to treatment till last day of study. Cows of group E (untreated) did not recovered from the mastitis. Subjective parameters viz. swelling, pain reflex of udder and physical changes in milk from quarter of ascorbic acid treated cows (group B) disappeared earlier than that of group C cows. It is concluded from this study that the ascorbic acid might be useful as an adjunct in case of clinical mastitis to get quick recovery with less number of intramammary infusions. High recovery rate in subclinical mastitis quarters of group D cows is appreciable and opens a new avenue to conduct further trials in a larger population in various field conditions. However, the pharmacology of ascorbic acid with particular reference to health of mammary gland needs to be investigated.

      • Effect of azadirachtin on parasitism rate of Gryon japonicum (Ashmead) and Ooencyrtus nezarae Ishii on pre-treated host eggs

        As a broad-spectrum insecticide azadirachtin may cause acute toxicity and alter behavior of natural enemies. We evaluatedthe toxicity of azadirachtin on two egg parasitoids of Riptortus pedestris (Fabricius) (Hemiptera: Alydidae), Ooencyrtusnezarae Ishii (Hymenoptera: Encyrtidae) and Gryon japonicum (Ashmead) (Hymenoptera: Platygastridae), by pre-treatinghost eggs with five concentrations, i.e., 15, 10, 7.5, 5, and 2.5 mL/L. Azadirachtin significantly reduced parasitism byG. japonicum in 15 and 10 mL/L concentrations than other lower concentrations. Similar effect was also observed inO. nezarae. The parasitism reduction capacity (R) for G. japonicum was 40.8 and 33.1% in 15 and 10 mL/L concentrations,but it was 81.6 and 71.4% in O. nezarae, respectively. Based on the IOBC guideline, azadirachtin can be slightly ormoderately harmful to O. nezarae and slightly harmful or harmless to G. japonicum.

        Response of Upper Ocean and Impact of Barrier Layer on Sidr Cyclone Induced Sea Surface Cooling

        In the present study an attempt has been made to investigate the impact of salinity stratification on the SST during the tropical cyclone (TC) passage. In this context, a severe post monsoon cyclone, Sidr, (Category 4) that developed over the south-eastern Bay of Bengal (BoB) during 11-16 November, 2007 was chosen as a case study. Pre-existence of a thick barrier layer (BL), temperature inversions and a higher effective oceanic layer for cyclogenesis (EOLC) were noticed along the path of the Sidr cyclone. The analysis of available Argo floats along the Sidr cyclone track also revealed less cooling during as well as after its passage as was reported from satellite derived SST. The role of BL on Sidr induced sea surface cooling was investigated using a diagnostic mixed layer model. Model results also depict the reduced sea surface cooling during the passage of Sidr. This is attributed to the presence of BL which results in the inhibition of the entrainment of cool thermocline water into the shallow mixed layer. Climatological as well as in situ observations of tropical cyclone heat potential (TCHP) and EOLC shows that the Sidr cyclone propagated towards the regions of higher EOLC.

      • Identification and evaluation of Beauveria bassiana ARP14 against Riptortus pedestris (Hemiptera: Alydidae) and its parasitoids

        An entomopathogenic fungus, Beauveria bassiana ARP14, was isolated from field collected Riptortus pedestris (F.). Lethal concentration of ARP14 was compared with commercialized GHA strain against R. pedestris and its egg parasitoids Ooencyrtus nezarae Ishii and Gryon japonicum (Ashmead). Mortality and infection rate were evaluated in five concentrations, i.e., 1×10 9 , 1×10 8 , 1×10 7 , 1×10 6 , and 1×10 5 conidia/mL using glass scintillation vial. The LC50 of 2nd, 4th instar, and adult stage of R. pedestris was not different between the two strains. The infection rate of ARP14 was higher than that of GHA in 4th instar and adult stage in 1×10 8 conidia/mL. The infection rate was low in ARP14 than GHA in both egg parasitoids. Therefore, our results suggest that the ARP14 strain can be used against R. pedestris with less negative effect on egg parasitoids than GHA strain.

        Role of Coflex as an Adjunct to Decompression for Symptomatic Lumbar Spinal Stenosis

        Study design: Prospective cohort study. Purpose: To assess whether additional implantation of Coflex following spinal decompression provided better clinical outcomes compared to decompression alone for symptomatic lumbar spinal stenosis (LSS) and to determine whether improvement in clinical outcomes correlated with changes in the radiological indices studied. Overview of Literature: Literature on benefits of additional Coflex implantation compared to decompression alone for symptomatic LSS is limited. Methods: Patients with symptomatic LSS who met the study criteria were offered spinal decompression with Coflex implantation. Those patients who accepted Coflex implantation were placed in the Coflex group (n=22); while those opting for decompression alone, were placed in the comparison group (n=24). Clinical outcomes were assessed preoperatively, six-months, one-year and two-years postoperatively, using the Oswestry disability index, 100 mm visual analogue scale (VAS)-back pain and VAS-leg pain, and short form- 36 (SF-36). Radiological indices (disc height, foraminal height and sagittal angle) were assessed preoperatively, six months, one year, and two years postoperatively. Results: Both groups showed statistically significant (p <0.001) improvement in all the clinical outcome indicators at all points in time as compared to the preoperative status. However, improvement in the Coflex group was significantly greater (p <0.001) than the comparison group. Changes in the radiological indices did not correlate significantly with the improvement in clinical outcome indicators. Conclusions: Additional Coflex implantation after spinal decompression in symptomatic LSS offers better clinical outcomes than decompression alone in the short-term. Changes in radiological indices do not correlate with the improvements in clinical outcomes after surgery for symptomatic LSS.

        Evidences from the past century have recorded that global warming has been increasing constantly and will continue to rise at the rates that are estimated to be between 0.1 to 0.2℃ per decade in the next century. We cannot negate the fact that rising temperatures would also effect harshly to animal sector particularly dairy sector. So far, it has been noticed that rising temperatures causing heat stress among dairying. However, at cellular level molecular chaperones (genes/heat shock proteins) gets activated and released. Genes that get up/down regulated following heat stress have been shown to be involved in protein folding, protein degradation, protein synthesis and detoxification, oxygen transport, phosphorylation etc. in different animals. In this study we discuss the impact of heat stress on cow’s dairy capacity, health and alongside the responses of different molecular chaperones in protecting the existing proteins for routine health and production. Additionally, the different mechanism that underlie activation of the heat shock proteins response and the role of heat shock proteins as cyto-protective molecules to develop them into active molecules as regulators of heat stress in animals.

      • Application of flonicamid and Beauveria bassiana GHA to manage soybean insect pests without disturbing natural enemies

        Negative effects of insecticides on non-target organism can be reduced by using selective and less toxic insecticides. We assessed the effect of subsequent application of flonicamid and Beauveria bassiana GHA on pests and natural enemies in soybean fields by recording their density in 6-8 d interval. Flonicamid controlled Aphis glycines Matsumura in 6 day after spray (DAS). Nymphs of Riptortus pedestris (F.) were also reduced by 46 and 48% in 12 and 19 DAS, respectively. Application of B. bassiana reduced R. pedestris nymph 50% in 16 DAS whereas adult R. pedestris, Medythia nigrobilineata Motschulsky and A. glycines were not significantly reduced in treated fields. Ladybird beetles, syrphid flies, hymenopteran wasps, and parasitism rate of R. pedestris eggs were not different between the treatment and control. These results suggest that flonicamid and B. bassiana showed less negative effect on the natural enemies and effective against aphids and nymphs stage of R. pedestris.

        Deubiquitinating enzymes as cancer biomarkers: new therapeutic opportunities?

        Cancer remains a life-threatening disease and accounts for the major mortality rates worldwide. The practice of using biomarkers for early detection, staging, and customized therapy may increase cancer patients’ survival. Deubiquitinating enzymes (DUBs) are a family of proteases that remove ubiquitin tags from proteins of interest undergoing proteasomal degradation. DUBs play several functional roles other than deubiquitination. One of the important roles of DUBs is regulation of tumor progression. Several reports have suggested that the DUB family members were highly-elevated in various cancer cells and tissues in different stages of cancer. These findings suggest that the DUBs could be used as drug targets in cancer therapeutics. In this review, we recapitulate the role of the DUB family members, including ubiquitin-specific protease, otubain protease, and important candidates from other family members. Our aim was to better understand the connection between DUB expression profiles and cancers to allow researchers to design inhibitors or gene therapies to improve diagnosis and prognosis of cancers.
