Hypoxia-inducible factor 1α inhibitor induces cell death via suppression of BCR-ABL1 and Met expression in BCR-ABL1 tyrosine kinase inhibitor sensitive and resistant chronic myeloid leukemia cells

        Masanobu Tsubaki,Tomoya Takeda,Takuya Matsuda,Akihiro Kimura,Remi Tanaka,Sakiko Nagayoshi,Tadafumi Hoshida,Kazufumi Tanabe,Shozo Nishida 생화학분자생물학회 2023 BMB Reports Vol.56 No.2

Chronic myeloid leukemia (CML) has a markedly improvedprognosis with the use of breakpoint cluster region-abelson 1(BCR-ABL1) tyrosine kinase inhibitors (BCR-ABL1 TKIs). However, approximately 40% of patients are resistant or intolerantto BCR-ABL1 TKIs. Hypoxia-inducible factor 1α (HIF-1α) is ahypoxia response factor that has been reported to be highlyexpressed in CML patients, making it a therapeutic target forBCR-ABL1 TKI-sensitive CML and BCR-ABL1 TKI-resistant CML. In this study, we examined whether HIF-1α inhibitors inducecell death in CML cells and BCR-ABL1 TKI-resistant CML cells. We found that echinomycin and PX-478 induced cell death inBCR-ABL1 TKIs sensitive and resistant CML cells at similar concentrationswhile the cell sensitivity was not affected with imatinibor dasatinib in BCR-ABL1 TKIs resistant CML cells. In addition, echinomycin and PX-478 inhibited the c-Jun N-terminalkinase (JNK), Akt, and extracellular-regulated protein kinase 1/2(ERK1/2) activation via suppression of BCR-ABL1 and Met expressionin BCR-ABL1 sensitive and resistant CML cells. Moreover, treatment with HIF-1α siRNA induced cell death by inhibitingBCR-ABL1 and Met expression and activation of JNK,Akt, and ERK1/2 in BCR-ABL1 TKIs sensitive and resistant CMLcells. These results indicated that HIF-1α regulates BCR-ABL andMet expression and is involved in cell survival in CML cells, suggesting that HIF-1α inhibitors induce cell death in BCR-ABL1TKIs sensitive and resistant CML cells and therefore HIF-1α inhibitorsare potential candidates for CML treatment.

E-Commerce in the Historical Approach to Usage and Practice of International Trade

춘홍차,Tsubaki, Koji The Korean Research Institute of International Com 2003 貿易商務硏究 Vol.19 No.-

The author believes that the main task of study in international trade usage and practice is the management of transactional risks involved in international sale of goods. They are foreign exchange risks, transportation risks, credit risk, risk of miscommunication, etc. In most cases, these risks are more serious and enormous than those involved in domestic sales. Historically, the merchant adventurers organized the voyage abroad, secured trade finance, and went around the ocean with their own or consigned cargo until around the $mid-19^{th}$ century. They did business faceto-face at the trade fair or the open port where they maintained the local offices, so-called "Trading House"(商館). Thererfore, the transactional risks might have been one-sided either with the seller or the buyer. The bottomry seemed a typical arrangement for risk sharing among the interested parties to the adventure. In this way, such organizational arrangements coped with or bore the transactional risks. With the advent of ocean liner services and wireless communication across the national border in the $19^{th}$ century, the business of merchant adventurers developed toward the clear division of labor; sales by mercantile agents, and ocean transportation by the steam ship companies. The international banking helped the process to be accelerated. Then, bills of lading backed up by the statute made it possible to conduct documentary sales with a foreign partner in different country. Thus, FOB terms including ocean freight and CIF terms emerged gradually as standard trade terms in which transactional risks were allocated through negotiation between the seller and the buyer located in different countries. Both of them did not have to go abroad with their cargo. Instead, documentation in compliance with the terms of the contract(plus an L/C in some cases) must by 'strictly' fulfilled. In other words, the set of contractual documents must be tendered in advance of the arrival of the goods at port of discharge. Trust or reliance is placed on such contractual paper documents. However, the container transport services introduced as international intermodal transport since the late 1960s frequently caused the earlier arrival of the goods at the destination before the presentation of the set of paper documents, which may take 5 to 10% of the amount of transaction. In addition, the size of the container vessel required the speedy transport documentation before sailing from the port of loading. In these circumstances, computerized processing of transport related documents became essential for inexpensive transaction cost and uninterrupted distribution of the goods. Such computerization does not stop at the phase of transportation but extends to cover the whole process of international trade, transforming the documentary sales into less-paper trade and further into paperless trade, i.e., EDI or E-Commerce. Now we face the other side of the coin, which is data security and paperless transfer of legal rights and obligations. Unfortunately, these issues are not effectively covered by a set of contracts only. Obviously, EDI or E-Commerce is based on the common business process and harmonized system of various data codes as well as the standard message formats. This essential feature of E-Commerce needs effective coordination of different divisions of business and tight control over credit arrangements in addition to the standard contract of sales. In a few word, information does not alway invite "trust". Credit flows from people, or close organizational tie-ups. It is our common understanding that, without well-orchestrated organizational arrangements made by leading companies, E-Commerce does not work well for paperless trade. With such arrangements well in place, participating E-business members do not need to seriously care for credit risk. Finally, it is also clear that E-International Commerce must be linked up with a set of government EDIs s

A Study of the PDCA and CAPD Economic Designs of the $\bar{x}$ Control Chart

Sun, Jing,Tsubaki, Michiko,Matsui, Masayuki Korean Institute of Industrial Engineers 2007 Industrial Engineeering & Management Systems Vol.6 No.1

The PDCA (Plan, Do, Check and Act) cycle is often used in the field of quality management. Recently, business environments have become more competitive, and the due time of products has shortened. In a short production run process, to increase efficiency of management, the necessity for distinguishing the PDCA design that starts with PLAN and the CAPD design that starts with CHECK has been clarified. Starting from Duncan (1956), there have been a number of papers dealing with the economic design of control charts from the viewpoint of production run. Some authors (Gibra, 1971; Ladany and Bedi, 1976; etc.) have studied the economic design for finite-length runs; other authors (Crowder, 1992; Del Castillo and Montgomery, 1996; etc.) have studied the economic design for short runs. However, neither the PDCA nor the CAPD design of control charts has been considered. In this paper, both the PDCA and CAPD designs of the $\bar{\x}$ chart are defined based on Del Castillo and Montgomery's design (1996), and their mathematical formulations are shown. Then from an economic viewpoint, the optimal values of the sample size per each sampling, control limits width, and the sampling interval of the two designs are studied. Finally, by numerically analyzing the relations between the key parameters and the total expected cost per unit time, the comparisons between the two designs are considered in detail.

The development of a high functional continuous filtration system for sericite powders

        최희규,Ochirhuyag Bayanjargal,JunIchiro Tsubaki,Takamasa Mori,Tadamitsu Sugimoto,JungEun Lee 한국화학공학회 2008 Korean Journal of Chemical Engineering Vol.25 No.5

A cake-less continuous filtration equipment has been developed based on an evaluation of inorganic powder slurry characteristics by many kinds of measurement methods, such as a sedimentation test under gravity and a hydrostatic pressure test. We have been developing a high-performance cake-less filtration system in which a condensed inorganic powder slurry layer maintains its fluidity. The development of this novel filtration system has allowed us to cease the scraping operation of the cake as well as to collect the highly condensed slurry more easily. These results clearly show that a new device may be realized for the deposition of highly condensed, fluid slurries. It is confirmed that the concentration of the condensed slurry amounted to 35 vol% while still retaining fluidity. The specifications of our new filtration system and its operation conditions for scale-up can be determined by theoretical methods. There is good potential for successfully collecting high condensation slurry more easily in a multiple filter system

Isolation and characterization of acid-soluble bluefin tuna (Thunnus orientalis) skin collagen

        Tanaka, Teruyoshi,Takahashi, Kenji,Tsubaki, Kazufumi,Hirata, Maika,Yamamoto, Keiko,Biswas, Amal,Moriyama, Tatsuya,Kawamura, Yukio The Korean Society of Fisheries and Aquatic Scienc 2018 Fisheries and Aquatic Sciences Vol.21 No.4

In this study, we isolated and characterized the acid-soluble skin collagen of Pacific bluefin tuna (PBT, Thunnus orientalis). The PBT skin collagen was composed of two ${\alpha}$ chains (${\alpha}1$ and ${\alpha}2$) and one ${\beta}$ chain. The denaturation temperature of PBT collagen was low although it was rich in proline and hydroxyproline. The primary structure of PBT skin collagen was almost identical to that of calf and salmon skin collagen; however, it differed with respect to the epitope recognition of the antibody against salmon type I collagen. These results suggest that the primary structure of skin collagen was highly conserved among animal species, although partial sequences that included the epitope structure differed among collagens.
