Persimmon Oligomeric Proanthocyanidins Extend Life Span of Senescence-Accelerated Mice

The anti-aging activities of persimmon oligomeric proanthocyanidins (POPs), reported to improve life span and behavioral characteristics associated with the aging process, were investigated using the senescence-accelerated mouse (SAM) P8, which is a good model for studies on aging-related behavioral changes as well as life span. We demonstrated that the administration of POPs extended the life span of SAMP8. In addition, POPs elevated Sirt1 expression, which is recognized as an essential factor for life span extension in the brain. On the other hand, the administration of POPs did not induce stereotypical behaviors such as rearing, jumping, and hanging from the lid of a cage, whereas food restriction increased these frequencies without a significant change in motor function. The present study suggests a promising role of POPs as anti-aging agents to extend life span, although further studies elucidating their anti-aging mechanisms acting are needed.
A novel therapeutic approach of Hachimi-jio-gan to diabetes and its complications

Great efforts have been made to improve both the quality of life and life expectancy of diabetes by treating problems associated with chronic complications such as neuropathy, retinopathy and nephropathy. In particular, diabetes is an increased risk of developing several types of kidney disease, and the predominant cause of end-stage renal disease in patients with this disorder is diabetic nephropathy. Therefore, prevention of the occurrence and progression of diabetes and its complications has become a very important issue. The scientific observations of an animal model of streptozotocin-induced diabetes, spontaneously occurring diabetes and diabetic nephropathy in this study suggest that one of the Kampo prescriptions, Hachimi-jio-gan comprising eight constituents, is a novel therapeutic agent.
Effectiveness of Green Tea Tannin on Rats with Chronic Renal Failure

The effects of green tea tannin on nephrectomized rats were examined. There were increases in blood urea nitrogen, serum creatinine, and urinary protein, and a decrease in creatinine clearance in the nephrectomized control rats, whereas better results for these parameters were obtained in rats given green tea tannin after nephrectomy, demonstrating a suppressed progression of the renal failure. When the renal parenchyma was partially resected, the remnant kidney showed a decrease in the activity of radical scavenger enzymes. Green tea tannin, however, was found to lighten the kidney under such oxidative stress. Mesangial proliferation and glomerular sclerotic lesions, which were conspicuous in the rats that were not given green tea tannin after nephrectomy, were also relieved.

Background : Excess alcohol consumption can lead to alcoholic liver damage and gastritis. Korean thistle (Cirsium japonicum var. maackii) has been reported that it’s administration protective from liver from lipid accumulation and toxicity. Also, Korean dandelion (Taraxacum coreanum Nakai) has been traditionally used for treating various inflammation-related diseases including ethanol-induced gastritis. In this study, we aimed to determine whether the combination treatment of Korean thistle and Korean dandelion could regulate alcoholic liver damage and gastritis in mice. Methods and Results : Seven-week-old mice were treated with ethanol (5 g/㎏ of body weight) by gavage every 12 h for a total of three doses to induce acute liver damage and gastritis. Ethanol extract of Korean thistle, Korean dandelion or combination was gavaged simultaneously with ethanol for three doses. The results showed that the combination treatment of Korean thistle and Korean dandelion significantly inhibited alcohol-induced increase of serum alanine transaminase and aspartate transaminase levels, liver weight, and hepatic caspase-3 expressions. Hepatic histopathological changes induced by alcohol were also remarkably improved by the combination treatment. The combination treatment inhibited alcohol-induced Morphological changes of gastritis and attenuated the expressions of pro-inflammatory protein, such as inducible nitric oxide synthase. Conclusion : The results of our study suggest that the combination treatment of Korean thistle and dandelion may simultaneously alleviate acute alcohol-induced gastritis and liver damage.

oxygen species(ROS) or fre radicals are produced in the pathogenesis of human diseases including ath-erosclerosis, diabetes, cancer, and aging. Antioxidants are associated with the prevention of ROS-induced tissue and cellulardamage in the various diseases. This study investigated the antioxidative activities of the methanol extract of Alisma plantago-aquatica var. orientale and its main component under conditions of radical generation using allophycocyanin and ferric-thio-cyanate assay. Alisol B 23-acetate as a main component was isolated from the methanol extract of Alisma plantago-aquaticavar. orientale. In results, the extract of Alisma plantago-aquatica var. orientale showed inhibitory activity on AAPH [2,2’-azo-bis(2-amidinopropane)dihydrochloride]-induced protein oxidation. Also, the extract of Alisma plantago-aquatica var. orientaleand alisol B 23-acetate inhibited lipid peroxidation. To reduce cardiovascular disease (CVD) by reducing circulating cholesterol concentrations, much attention has been focused on the search for dietary interventions for hypercholesterolemia. Corni Fructus is known to exhibit several biological activities. Therefore, in the present study, its protective effect on diet-induced hypercholesterolemia was studied using a rat model fed 1% cholesterol and 0.5% cholic acid. Corni Fructus extract was administered at an oral dose of 50, 100, or 200mg/kg of body weight/day for 10 days. The administration inhibited the elevation of both systolic and diastolic blood pressure. In addition, it lowered serum total cholesterol levels with a decrease in esterified cholesterol. Moreover, the atherogenic index was decreased in a dose-dependent manner, suggesting its protective role against CVD through regulating cholesterol and lipoprotein levels. The hepatic levels of total and free cholesterol were also reduced by Corni Fructus. This implies that free cholesterol was used for catabolism or efflux. Consequently, the levels of total cholesterol and bile acid in feces were significantly increased. Our present results also showed that the administration of Corni Fructus decreased lipid peroxidation, suggesting a protective effect against oxidative stress induced by hypercholesterolemia. Furthermore, hypercholesterolemic control rats had significantly lower expression of sterol regulatory element-binding protein (SREBP)-2 protein without any difference in SREBP-1 protein expression. On the other hand, the protein expression of SREBP-2 was significantly up-regulated by Corni Fructus. Furthermore, the protein expression of peroxisome proliferator-activated receptor α was elevated, indicating that Corni Fructus would activate fatty acid oxidation. In conclusion, Corni Fructus may protect against CVD by regulating blood pressure, cholesterol levels, and expression of proteins related to lipid metabolism.
