      • Alcohol Dehydrogenase의 활성에 미치는 인삼 Saponin의 영향

        인삼 total saponin류를 protopanaxadiol과 protopanaxatriol을 aglycon으로 하는 diol계 saponin과 triol계 saponin으로 분획하여 각 분획물과 total saponin이 간장 alcohol dehydrogenase(ADH, EC 1.1.1.1)활성에 미치는 영향을 in vitro에서 관찰하고 다음의 결과를 얼었다. (1) 인삼 total saponin이나 diol계 saponin이 적당량(??) 존재할 때 ADH는 활성화되어 ethanol의 산화 속도가 control에 비하여 약 11% 촉진되었으나, triol계 saponin은 ADH의 활성에 별다른 영향을 미치지 않는 것으로 관찰되었다. (2) 인삼 total saponin과 diol계 saponin, triol계 saponin 존재하에서 ADH의 ethanol에 대한 km 값을 측정한 결과 ??%때 가장 낮은 km값을 나타내었으나, diol계 saponin존재할 때의 km값은 triol계 saponin이 존재할 때보다 낮은 값을 나타내었다. 위와 같은 실험 결과로 미루어 보아 인삼 saponin은 중 diol계 saponin이 주로 ADH 활성에 영향을 미치는 것으로 생각되며, 인삼 saponin은 ADH의 ethanol에 대한 친화도를 증가하는 것으로 예측된다. The effects of total ginseng saponins, protopanaxadiol and protopanaxatriol containing saponin fractions of korean ginseng root (Panax ginseng C.A. Meyer) on the oxidation of ethanol by liver alcohol dehydrogenase (ADH,EC l.l.l.l)in vitro has observed and the following results were obtained. (1) In the presence of moderate amounts (??) of total ginseng saponins or protopanaxadiol containing saponins, the oxidation of ethanol by the activation of ADH was found stimulated about 11% compared with control. But no noticeable effect of the protopanaxatriol containing saponins on ADH activity was observed. (2) The km values of ADH to ethanol in the presence of the total saponins and protopanaxadiol containing saponins and protopanaxatriol containing saponins were found the lowest km in ??% and km value of ADH to ethanol in the presence of diol saponins was lower than that of triol saponins. From the above results, it was realized that the protopanaxadiol containing saponins of the total saponins might effect on the ADH activity by lowering km of ADH ethanol.

      • Methyl Iinoleate가 mouse 혈청 및 간의 단백질 효소 활성에 미치는 영향

        순수한 methyl linoleate를 mouse에 장기간 경구투여 하였을 때의 생체에 미치는 영향을 구명하기 위하여 methyl linoleate의 일정량을 1일 1회 20일 동안 경구투여하여 혈청 및 간장에서의 단백질 및 효소활성에 미치는 영향을 검토한 결과는 다음과 같다. 실험군의 혈청에서는 정상군에 비하여 TBA값이 증가하였으며, TBA값의 증가에 따라 실험군의 혈청내 acid phosphatase, alkaline phosphatase, glutamic-oxaloacetic transaminase(GOT) 및 glutamic-pyruvic transaminase(GPT)의 활성은 모두 증가하고 있었다. 실험군의 간에서의 acid phosphatase와 alkaline phosphatase의 활성은 증가하였으나, GOT와 GPT의 활성은 감소하는 경향을 나타내었다. 한편, 혈청단백질의 전기영동상은 실험군에서는 정상군에 비하여 albumin과 α₂-globulin은 증가하였고, β-globulin과 r-globulin은 감소하였다. 전기영동 결과 lactic dehydrogenase(LDH) isozyme들의 백분율은 LDH₁은 증가한 반면, LDH₄, ??의 활성은 감소하였다. 이상의 결과로부터 methyl linoleate를 mouse에 장기간 투여하였을 때 혈청 및 간의 단백질 및 효소활성에 크게 영향을 미치고 있음을 알 수 있었다. In order to investigate the effect of pure methyl linoleate fed to the mouse on the protein and enzyme activity in the mouse serum and liver, we adminstered the fixed dosage of methyl linoleate to mice once per day for 20 days by using stomach tube. And the following results were obstaind. TBA value in the serum of test group was increased than those of normal group. The activities of acid phosphatase, alkaline phosphatase, glutamic oxaloacetic transaminase(GOT), and glutamic pyruvic transaminase(GPT) in the serum of test group were increased than those of normal group. The activities of acid phosphatase and alkaline phosphatase in the liver of test group were increased than those of normal group but the activities of GOT and GPT were slightly decreased in the test group. In the serum protein of test group, the amounts of albumin and α₂-globulin were increased, on the other hand the amounts of β-globulin and r-globulin were decreased. According to the electrophoresis, the activity of LDH₁were increased while these of LDH₄and ?? were decreased. From these results we conclude that the pure methyl linoleate fed to mouse influences on the protein and enzyme activity in the mouse serum and liver.

        자동산화 Methyl Linoleate가 Mouse혈청의 효소활성에 미치는 영향 (급성 독성)

        In order to investigate the acute toxicity of autoxidized methyl linoleate(AOML) on the activity of serum enzymes in the mouse, we administered once 0.45ml of AOML to ICR strain mouse by using stomach tube. The following results were obtained: The total lactic dehydrogenase(LDH) activities in the serum of AOML group were generally increased than those of normal group. According to electrophoresis, the activities of LDH, were increased while those of LDH, were decreased. The activities of glutamic oxaloacetic transaminase(GOT), glutamic-pyruvic transaminase(GPT) and α-amylase in the serum of AOML group were increased more than those of normal group. The activities of alkaline phosphatase in the serum of AOML group were increased but those of isozyme were not confirmed in the normal and AOML group. In the serum protein of AOML group, albumin was increased, on the other hand γ-globulin was decreased. At the peripheral blood slide smear, lymphocytes were significantly decreased but neutrophils were increased and the morphological change of erythrocytes was observed. From these results we conclude that the AOML fed to mouse influences on the activity of various serum enzymes and blood cells in the mouse.

        자동산화 Methyl Linoleate가 Mouse간장의 효소활성에 미치는 영향

        자동 산화유가 mouse의 간장 효소에 미치는 영향을 구명하기 위하여 autoxidized methyl linoleate의 일정량을 mouse에게 1일 1회 20일 동안 경구 투여하여 간에서의 효소 활성을 조직화학적 및 생화학적 방법으로 검토한 결과는 다음과 같다.<br/> 실험군의 간에서의 POV, COV 및 TBA값은 정상군에 비해 크게 증가하여 AOML이 많이 흡수되어 있음을 알 수 있었다.<br/> 실험군의 간에서의 phospholipid, triglyceride 및 total cholesterol의 양은 정상군에 비해 증가하였다.<br/> 실험군의 간에서의 acid phosphatase, alkaline phosphatase의 활성은 정상군에 비해 증가하였으며, ATPase의 활성은 감소하였다.<br/> 실험군의 간에서의 형태학적 변화는 정상군에 비해 핵산의 감소, 지질의 축적 및 간 세포의 손상이 있었다.<br/> 이상의 결과로부터 mouse에 경구 투여한 자동 산화 methyl linoleate는 간에서의 효소 활성과 간의 형태학적 변화에 크게 영향을 주고 있음을 알 수 있었다. In order to investigate the effect of autoxidized oil on the enzyme activity in the mouse liver, we administered the fixed dosage of autoxidized methyl linoleate (AOML) to mice once per day for 20 days by using stomach tube and investigated the enzyme activity with the histochemical staining method and biochemical analysis.<br/> The following results were obtained:<br/> The POV, COV and TBA value in the liver of AOML group were significantly increased than those of normal group. The phospholipid, triglyceride and total cholesterol in the liver of AOML group were slightly increased than those of normal group. The activities of acid phosphatase and alkaline phosphatase in the liver of AOML group were increased than those of normal group but ATPase activity was decreased in the AOML group. The decrease of RNA, accumulation of fat and damage of liver cells were observed as the morphological changes in the liver of AOML group.<br/> From the results obtained we conclude that the autoxidized methyl linoleate influenced upon the various enzyme activity and the morphological changes in the mouse liver.

      • 송옆중의 항산화성 물질에 관한 연구 : 제1보 : 송옆의 각종 용매 추출물의 항산화작용 The Antioxidative Action of Various Solvent Extracts of Pine Needles

        송옆중의 항산화성 물질의 존재와 항산화성의 정도를 검토하기 위하여 각종 용매 추출물이 linoleic acid의 자동산화에 미치는 영향을 in vitro에서 oven test법 및 peroxide value, carbonyl value 측정등으로 조사한 결과는 다음과 같다. 1. 송옆의 각종 용매 추출물들은 모두 항산화작용을 나타내어 linoleic acid의 자동산화를 억제하여 자동산화의 유도기간을 연장시켰으며, 그중 methanol 추출물은 가장 강한 항산화효과를 나타내었다. 2. Linoleic acid의 자동산화 유도기간은 methanol 추출물의 첨가 농도에 비례하여 예장되었다. 3.Methanol 추출물의 항산화성 물질은 methanol 추출물의 acetone 분획물에 존재하고 있었으며, acetone 분획물은 methanol 추출물과 같은 정도의 항산화성을 나타내었으나, methanol 추출물의 잔류물은 항산화성을 거의 나타내지 않았다. The antioxygenic effect of chloroform-methanol, methanol and water extracts of pine needles on the autooxidation of linoleic acid has been investigated in vitro. The following results were obtained: 1. All of the extracts of pine needles showed the antioxygenic activity and inhibited the autooxidation of linoleic acid with extending the induction period. And the methanol extract showed the most prominent antioxygenic activity. 2. The induction period of the formation of hydroperoxides was extended with concentration of methanol extract. 3. Antioxygenic substances in methanol extract were found to remove into acetone fraction. The acetone fraction showed the same degree of antioxygenic activity as methanol extract. But residue of methanol extract didn't show the antioxygenic activity.

      • Methyl Linoleate 가 Mouse 혈청 및 간의 지질대사에 미치는 영향

        순수한 methyl linoleate를 mouse에 장기간 경구투여 하였을 때의 생체에 미치는 영향을 구명하기 위하여 methyl linoleate의 일정량을 1일 1회 20일 동안 투여하여 혈청 및 간장에서의 지질대사를 검토한 결과는 다음과 같다. 실험군의 혈청 triglyceride. phospholipid, total cholesterol 양 및 혈청 TBA 값은 정상군에 비하여 모두 증가되었다. 간에서의 TOV, COV및 TBA 값은 정상군에 비하여 실험군에서 모두 크게 증가하고 있으 며, 또 한편 형청 lipoprotein의 전기영동 결과 형청 lipoprotein의 조성 백분율은 albumin과 α-lipoprotein 은 증가하였으며, pre-β-lipoprotein과 β-lipoprotein은 감소되었다. 이상의 결과로부터 methyl linoleate를 mouse에 장기간 투여하였을 때 혈청 및 간의 지질대사에 크게 영향을 미치고 있음을 알 수 있다. In order to investigate the effect of pure methyl linoleate on the lipid metabolism in the mouse serum and liver, we administered the fixed dosage of methyl linoleate to mice once day for 20 days by stomach tube. The results as follows; The amounts of triglyceride, phospholipid, total cholesterol and TBA value in the blood serum to test grovp were all increased when they compared with of control group. The POV,COV and TBA values increased significantly, and also the amounts of pyospholipid. triglyceride and total cholesterol were slightly increased in the test group liver, respectively. Accroding to the electrophoresis of serum lipoprotein, the amounts of albumin and α-lipoprotein were increased in the test group. On the other hand, the amounts of pre-β-lipoprotein were decreased. From these results we concluded that the pure methyl linoleate fed to mouse affected significantly on the lipid metabolism.

      • ECOSYSTEM ON THE GULF OF YEONGIL IN THE EAST SEA OF KOREA 4. Horizontal and vertical distribution of salinity and density

        The oceanographic characteristics of the Gulf of Yeongil were studied by using salinity and density readings obtained "in situ" from September 1985 to August 1987. The characteristics of salinity and density are directly affected by fresh waters induced from precipitation in summer and evaporation induced from latent heat loss in winter. Seasonal and annual variations of all horizontal stations are very similar to one another. Vertical variations of stations located in order to observe the vertical characteristics appear sharply to have an effect on the stratification patterns, such as halocline and pycnocline in summer and the homogeneous patterns in winter. Interesting variations are found out the stratification patterns at the surface layer in July and the upwelling phenomena at the bottom layer in August. It indicates that the former is the result of an inflow of fresh water from the Hyungsan River and the latter is the result of an intrusion of cold water from the East Sea.
