      • Histone Deacetylase 3 Induces miR-224 Expression to Drive EMT and Fibroblast Migration Through FOXA1 Downregulation

        Epithelial-mesenchymal transition (EMT), a process by which epithelial cells undergo a phenotypic conversion that gives rise to myofibroblasts, plays a pivotal role in the development of lung diseases such as pulmonary fibrosis. Recent studies revealed that hypoxia induces EMT of alveolar epithelial cells and that histone deacetylases (HDACs) are aberrantly overexpressed in basal cells of idiopathic pulmonary fibrosis (IPF) lungs. Previously, we found that HDAC3 promotes transition of alveolar epithelial cells to mesenchymal cells and fibroblast migration under hypoxic conditions, thereby accelerating the pulmonary fibrosis. In this study, we aimed to discover the microRNAs (miRNAs) that are controlled by HDAC3 in hypoxic conditions and also identify their target genes. In doing so, we planned to elucidate the mechanism by which HDAC3 accelerates the pulmonary fibrosis process. To identify the miRNAs that are regulated by the HDAC3, we performed a new-generation sequencing analysis using HDAC3-overexpressing A549 cells and found that miR-224 is upregulated by HDAC3 overexpression. MiR-224 expression was found to be increased in lungs from bleomycin-induced mice; it also induced the migration and invasion of hypoxia-exposed fibroblast cells. In addition, we analyzed the target genes of miR-224 using the TargetScan database and selected from these the genes that are associated with EMT for further study. Of these genes, Forkhead Box A1 (FOXA1) was found to be increased by HDAC3 siRNA and miR-224 inhibitor in hypoxia-exposed alveolar cells. Additionally, FOXA1 regulated the expression of mesenchymal markers in HDAC3 siRNA-transfected alveolar cells. FOXA1 also controlled the migration and invasion of HDAC3 siRNA-transfected fibroblast cells under hypoxia. Furthermore, the expression of FOXA1 was downregulated in lungs of bleomycin-induced mice and IPF patients. Therefore, we suggest that HDAC3 induces miR-224 expression to drive alveolar EMT through FOXA1 downregulation in hypoxic conditions, which may accelerate the pulmonary fibrosis.

      • Association of miRNA-Mrna in PM-induced Inflammation in Human Bronchial Epithelial Cells

        Background Particulate matters (PM) are known to be associated with respiratory diseases such as pneumonia and chronic obstructive pulmonary disease (COPD). We hypothesized PM exacerbates inflammatory response in pulmonary diseases. However, the molecular mechanisms of PM-mediated inflammation are still unclear. Therefore, this study aimed to identify the dysregulated genes that related to inflammatory response in PM-exposed bronchial epithelial cells, and to explore potential microRNA (miRNA) mRNA correlations to inhibit PM-induced inflammation. Methods We performed a next-generation sequencing (NGS) and bioinformatics study to evaluate the changes of mRNA and miRNA profiles in PM-exposed BEAS 2B cells. Also, we analyzed the miRNA-mRNA correlations by ingenuity pathway analysis (IPA). To validate the Results from NGS analysis, the expression levels the miRNAs and mRNAs involved in PM-induced inflammation were assessed by qRT-PCR and western blot analysis. Results We identified 1,088 dysregulated genes, mostly involved in the inflammation and airway chronic inflammatory disorder. Using the miRmap database, 12 dysregulated miRNAs were identified, and 166 target mRNAs were predicted. After validation using TargetScan and qRT-PCR, we identified three downregulated miRNAs (hsa-miR-155-5p, hsa-miR-210-5p, hsa-miR-6515-5p) and their target genes (CXCL1, CXCL3, CSF2, CSF3, ICAM1, CYP1A1 and HMOX1). Overexpression of their miRNAs suppressed the production of pro-inflammatory cytokines and expression of inflammatory genes induced by PM exposure. Furthermore, overexpression of their miRNAs decreased the expression of predicted target genes in PM-exposed epithelial. Conclusion We discovered hsa-miR-155-5p, hsa-miR-210-5p, hsa-miR-6515-5p miRNAs, and their target mRNAs, CXCL1, CXCL3, CSF2, CSF3, ICAM1, CYP1A1 and HMOX1in PM-induced Beas 2B cells, suggesting that association of these miRNA-mRNA might contribute to increased inflammation of PM-exposed bronchial epithelial cells.

        한국판 긍정성 공명 척도의 타당화 연구

        본 연구의 주 목적은 Fredrickson(2016)의 긍정성 공명 이론을 바탕으로 Major, Le Nguyen, Lundberg와 Fredrickson(2018)이 제작한 긍정성 공명 척도(Positivity Resonance Scale)를 한국판으로 번안 및 타당화하는 데 있다. 대학생 335명을 대상으로 탐색적 요인분석과 확인적 요인분석을 실시하였다. 탐색적 요인분석 결과 1요인 모형 구조를 확인하였으며, 확인적 요인분석 결과 1요인 모형의 모형 적합도가 양호하게 나타나 한국판 긍정성 공명 척도의 1요인 구조를 확정하였다. 공존 타당도를 검증한 결과, 긍정성 공명은 긍정 정서, 정신적 안녕, 사회적 유대감, 관계 유능성과는 정적 상관을, 부정 정서, 우울, 외로움과는 부적 상관을 나타냈다. 위계적 회귀분석을 실시한 결과 긍정성 공명은 긍정 정서를 통제하고도 유의한 증분 설명량을 나타냈다. 이러한 결과를 통해 한국판 긍정성 공명 척도가 신뢰롭고 타당한 도구임을 밝혔다. 마지막으로 본 연구 결과의 의의와 한계점 및 추후 연구에 대해 제언하였다. This study aimed to validate the Korean version of the Positivity Resonance Scale developed by Major, Le Nguyen, Lundberg, & Fredrickson(2018) based on Fredrickson’s(2016) positivity resonance theory. We conducted an exploratory factor analysis and confirmatory factor analysis using the data set of 335 undergraduate students. Results provided support for the one-factor structure of the Korean version of the Positivity Resonance Scale. The Korean version of the Positivity Resonance Scale was positively correlated with positive affect, mental health, social connectedness, interpersonal competence, and was negatively correlated with negative affect, depression, and loneliness, supporting good concurrent validity. Also, a hierarchical multiple regression analysis resulted in the incremental validity of the Korean version of the Positivity Resonance Scale, which suggests that the construct of positivity resonance is well discriminated from positive affect measures. Overall, these findings indicated that the Korean version of the Positivity Resonance Scale is a reliable and valid tool for measuring the concept of positivity resonance. This study ends with a discussion of the implications, limitations, and directions for future research.

        First Report on Plum Pocket Caused by Taphrina deformans in South Korea

        Plum pocket caused by the dimorphic ascomycetous fungi, Taphrina spp., results in unsightly malformations and crop loss. In 2016, Japanese plums (Prunus salicina Lindl.) with plum pocket symptoms were found in Gimcheon. Three isolates were collected from symptomatic P. salicina fruits and identified as Taphrina deformans based on morphological characteristics and molecular sequence analysis of including internal transcribed space (ITS) and the mitochondrial small ribosomal subunit (SSU) regions of the three isolates. Pathogenicity test on plum fruits confirmed that, the present T. deformans isolates are causal agent of plum pocket. To the best of our knowledge, this is the first report of plum pocket caused by T. deformans in South Korea.

        The interior design was emerged to determine the merchantability of Automobile due to the quality enhancement and diversification of Automobile consumer Needs. Especially, the interior lighting was one of the main factors being obtained the various emotional effects for the image of Automobile. This study thus focused to evaluate the various emotional images that the users felt by the lighting color of Automobile Interior. As this results, the warm color series like as Orange and Red was highly evaluated in the image of 「Luxury·profound」, and the white blue series was highly evaluated in the image of 「Leading-edge」 and 「Gorgeous」. Type B, D and E not integrated with Cluster and Button types highly evaluated in the image of 「Changeable」 In addition, it seemed to be considered Design types followed each material and Button types of Interior skin and factors with the zoning in the further study. Therefore, this study will be presented the basic materials for the study conducted in the future.

        Prevalence and Risk Factors of Abusive Behaviors in the Caregivers of People with Dementia in Korea

        Objective We investigated the prevalence and risk factors of physically abusive behaviors (PhAB) and psychologically abusive behaviors (PsAB) towards people with dementia (PWD) in family caregivers, and compared their prevalences between East Asian and Western countries. Methods We estimated the prevalence and risk factors of PhAB and PsAB in 467 Korean pairs of community-dwelling PWD and their primary family caregivers. We evaluated abusive behaviors using the Modified Conflict Tactics Scale. In addition, we compared the prevalence of abusive behaviors between Asian and Western countries through a meta-analysis on 12 studies including the current one. Results More than a half of the caregivers reported PsAB and about one out of seven caregivers admitted PhAB within past three months. PsAB and PhAB were slightly more prevalent in East Asian countries than in Western countries. Non-Alzheimer type and moderate to severe behavioral and psychological symptoms of dementia were associated with the risk of PhAB but not with the risk of PsAB. Severe care burden and low income were associated with the risk of PhAB and PsAB. Conclusion PhAB and PsAB were as prevalent in the family caregivers of PWD in Asian countries including Korea as in Western countries. Prevention strategies should be implemented according to the type of abusive behaviors.

      • Fluorapatite diagenetic differences between Cretaceous skeletal fossils of Mongolia and Korea

        <P><B>Abstract</B></P> <P>The skeletal tissue of modern bones is mostly composed of hydroxyapatite, which contains calcium and phosphate. During diagenesis, calcium, phosphorus and minerals of the hydroxyl group can be exchanged with other elements to form carbonate-fluorapatite. It is well-established that paleoenvironmental evolution and geological events have played a significant role in the compositional changes of fossilized bones. For example, skeletal fossils from the Hasandong Formation (Aptian- early Albian) of the Gyeongsang Basin in Korea are characterized by black and dark colors, whereas dinosaur bones from the Nemegt Formation (early Maastrichtian) of the Nemegt and Altan Uul ranges in Mongolia are light brown and white. This study investigated the mineralogical and geochemical causes for the differences in coloration between these two groups of fossilized bones. Multiple synchrotron-based techniques were utilized, including synchrotron–XRF, –XRD, –X-ray micro-computed tomography (μ-CT), micro-XRF and SEM-EDX data to analyze the elemental composition and mineral phases in dark Korean fossilized bones, which are characterized by the presence of iron, aluminum, magnesium and other trace elements. Chamosite was identified as a secondary mineral at 5% by weight of the total fossil mass, which is primarily composed of carbonate-fluorapatite. However, skeletal fossils from Mongolia are characterized by secondary minerals, such as barite, goethite and calcite, which accumulate in pore spaces. Since different secondary minerals result from different alteration procedures, the presence of chamosite in the Korean fossils suggests alteration by spatial replacement, and the presence of barite in the Mongolian fossils suggests alteration by accumulation. The investigation of these two skeletal groups, analyzed using a suite of synchrotron-based multidisciplinary techniques, revealed contrasting mineralogical and geochemical details and helps to determine the origin of fossil colorization.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Two distinct colorizations in fossilized bones show different degrees of alteration in the original bone material. </LI> <LI> Secondary mineral phases have indicated spatial replacement and precipitation processes during respective alteration. </LI> <LI> Synchrotron-based X-ray techniques are shown to be useful in revealing mineralogical and chemical information of fossils. </LI> </UL> </P>

        반려견 보유세 도입에 대한 탐색적 연구

        사회·문화·경제적 변화에 따라 반려동물의 사육 수가 증가되고, 반려동물의 적절한 관리를 위해 필요한 재원부담도 늘고 있다. 농림축산식품부는 “동물복지 5개년 계획”을 통해 장기적으로 반려동물 보유세를 검토하겠다고 발표하였다. 이 연구는 반려동물 보유세에 대한 학술적 검토와 논의가 활성화되지 않은 상태에서 동 조세 도입의 사회적 합의가 있다는 가정하에 전략적 도입방안을 검토하는데 목적을 두었다. 조세의 신설은 부담자와 편익자 등 다수의 저항을 불러올 수 있으며 찬・반 양론이 대립 중임을 감안해 반려동물 보유세 도입 여부에 대한 판단은 연구에서 제외하였다. 이 연구에서는 반려동물 중 가장 큰 비중을 차지하는 반려견만을 대상으로 분석하였고, 이를 기반으로 반려견 보유세 도입의 전제와 방식을 제안하였다. 반려견 보유세를 도입한다고 가정할 경우 단기 대안과 장기 대안을 구분하여 제시하였다. 단기적으로는 “면허세” 방식으로 도입하되 장기적으로는 “책임・규제세” 형식으로 전환함이 타당함을 제시하였다. 구체적으로 원인자 부담과 수익자 부담 원칙을 따르되 목적세 형식으로 도입하고, 종량세 형식을 취하되 견종과 개체 수에 따라 차등 세율을 적용하며, 외부경제와 불경제가 발생되는 기초자치단체에 과세권을 부여하는 방식을 제시하였다. 이 연구는 아직 지방재정 분야에서 논의가 확산되지 않은 반려견 보유세 도입에 대한 탐색적 연구로서 향후의 논의 기초를 제공한다는 함의를 갖는다. In line with the increase in companion animals, the Ministry of Agriculture, Food and Rural Affairs reviewing the tax on companion animals. But, There has been no social consensus on companion dog ownership tax. Also, academic debate on tax introduction is not spreading. And the establishment of a tax can lead to resistance from the bearer. Since the pros and cons are still in conflict, the decision on whether to introduce tax was excluded from the discussion. In this context, this study aims to exploratory review the introduction of the companion dog ownership tax. In the short term, it is reasonable to introduce the license tax. In the long run, it makes sense to introduce it in the form of a responsible regulatory tax. Specifically, the Companion dog ownership tax was introduced in the form of a purpose tax, and a method of applying a differential tax rate while taking the form of a specific tax, and giving the tax right to basic municipalities was suggested. This has implications as basic data in the field of local finance research, which has not yet spread.
