        Tannic acid-induced apoptosis in FaDu hypopharyngeal squamous cell carcinoma

        Tannic acid (TA) is a water-soluble polyphenol compound found in various herbal plants. We investigated the chemopreventive effects of TA on FaDu hypopharyngeal squamous carcinoma cells. In an 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, TA showed dose-dependent cytotoxicity with a half maximal inhibitory concentration (IC50) of 50 µM. Cell cycle analysis and immunofluorescence imaging demonstrated that under low-dose (25 µM) treatment, FaDu cells were arrested in G2/M phase, and as the dose of TA was increased, apoptosis was induced with the increase of cell population at sub-G1 phase. The expressions of various cyclins, including cyclin D1 and cyclin-dependent kinases (CDK-1 and CDK-2), were down-regulated at low doses of TA, whereas apoptotic effectors such as cleaved caspase 3, cleaved caspase 7, and poly (ADP-ribose) polymerase (PARP) were expressed in a dose-dependent manner in Western blotting. In addition, TA-induced apoptosis of FaDu cells might be mediated by the extracellular signal-regulated kinase (ERK)/mitogen-activated protein kinase pathway, with the upregulation of p-AKT/p-PKB (phosphorylated protein kinase B) and p-ERK. Overall, our data support the hypothesis that TA is a potential candidate agent for the treatment of hypopharyngeal cancer.

        Investigation of the Relationship Between Cognitive Functions and Retinal Findings From Spectral Optical Coherence Tomography in Patients With Schizophrenia and Their Healthy Siblings

        Objective Retinal structural changes which were investigated by optical coherence tomography (OCT) have been reported in schizophrenia. Since cognitive dysfunction is a core feature of schizophrenia, the correlations between retinal findings and cognitive functions of patients and their healthy siblings may provide insight into the pathophysiological processes of the disorder. We aimed to investigate the relationship between neuropsychiatric tests and retinal changes in schizophrenia patients and their healthy siblings.Methods We measured OCT parameters and cognitive performance (via Trail Making Tests, verbal fluency tests, and The Digit Span Tests) of 72 participants (36 patients with schizophrenia and 36 healthy siblings) and disease severity (with Positive and Negative Syndrome Scale, Global Assessment of Functioning, and Clinical Global Impression scales) in patients with schizophrenia and evaluated the relationship between retinal findings and clinical parameters, especially neurocognitive tests.Results We found decreased ganglion cell layer-inner plexiform layer thickness and macular volume in the patient group. There were strong correlations between neurocognitive tests and OCT findings in both groups. On the other hand, there was not any correlation between retinal findings and disease parameters.Conclusion The cognitive symptoms of schizophrenia may be more closely related to structural changes in the retina.

        Physciosporin suppresses the proliferation, motility and tumourigenesis of colorectal cancer cells

        <P><B>Abstract</B></P> <P><B>Background</B></P> <P>Lichens, which represent symbiotic associations of fungi and algae, are potential sources of numerous natural products. Physciosporin (PHY) is a potent secondary metabolite found in lichens and was recently reported to inhibit the motility of lung cancer cells via novel mechanisms.</P> <P><B>Purpose</B></P> <P>The present study investigated the anticancer potential of PHY on colorectal cancer (CRC) cells.</P> <P><B>Methods</B></P> <P>PHY was isolated from lichen extract by preparative TLC. The effect of PHY on cell viability, motility and tumourigenicity was elucidated by MTT assay, hoechst staining, flow cytometric analysis, transwell invasion and migration assay, soft agar colony formation assay, Western blotting, qRT-PCR and PCR array <I>in vitro</I> as well as tumorigenicity study <I>in vivo</I>.</P> <P><B>Results</B></P> <P>PHY decreased the viability of various CRC cell lines (Caco2, CT26, DLD1, HCT116 and SW620). Moreover, PHY elicited cytotoxic effects by inducing apoptosis at toxic concentrations. At non-toxic concentrations, PHY dose-dependently suppressed the invasion, migration and colony formation of CRC cells. PHY inhibited the motility of CRC cells by suppressing epithelial-mesenchymal transition and downregulating actin-based motility markers. In addition, PHY downregulated β-catenin and its downstream target genes cyclin-D1 and c-Myc. Moreover, PHY modulated KAI1 C-terminal-interacting tetraspanin and KAI1 expression, and downregulated the downstream transcription factors c-jun and c-fos. Finally, PHY administration showed considerable bioavailability and effectively decreased the growth of CRC xenografts in mice without causing toxicity.</P> <P><B>Conclusion</B></P> <P>PHY suppresses the growth and motility of CRC cells via novel mechanisms.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Treatment of hydrocyanic galvanic effluent by electrocoagulation: Optimization of operating parameters using statistical techniques and a coupled polarity inverter

        The treatment of hydrocyanic galvanic effluent with Zn, Ni and Cu ions by electrocoagulation (EC) using aluminum electrodes and a polarity inverter was evaluated. 32 experiments were performed and the optimal operating conditions were obtained using statistical analysis: 30minutes, 5.00 g/L NaCl, 1 cm between electrodes, electrode surface area of 104cm2/L and current density of 8mA/cm2. The results for removal with the polarity inverter were: 99.55% total cyanide, 22.49% total nickel, 52.66% total copper and 100% total zinc. After the EC experiments, localized and general corrosion of the aluminum electrodes was evaluated both internally and externally by SEM. Contaminant removal was confirmed by X-Ray powder diffraction (XRD) and Fourier transform infrared (FT-IR) which demonstrated the presence of contaminants in the galvanic sludge generated in the process.

        Learner-friendly textbooks: Chemistry texts based on a constructivist view of learning

        In this study, the use of inquiry methods, learning cycles, a conceptual change model and analogy in creating alternative science texts was discussed. An alternative text on the topic of acids and bases was created by integrating the methods and models discussed in this paper. The alternative text and a sample of a traditional text taken from a textbook, which is still used in Turkish high schools, were given to two groups, totaling 42 pre-service teachers-the alternative text was given to an experimental group and the traditional text to a control group-in an experimental setting and their understandings of acids and bases were compared. In addition, in the second step of the study, the pre-service teachers read both texts and indicated their preferences in terms of interest, understandability and helpfullness.

        Microdissected Prefabricated Flap: An Evolution in Flap Prefabrication

        When traditional flap techniques are not feasible, we apply flap prefabrication, which is more complicated and sophisticated but supplies large and thin flaps. There are some disadvantages to the technique that require improvement, such as venous congestion after flap transfer, which requires months for neoangiogenesis and necessitates a vascular carrier. Here, the author presents a new technique, called as ‘microdissected prefabricated flap,’ to successfully produce a safe, large, and thin flap. This technique is based on the microdissection of the perforators to the greatest extent possible, spreading them out into the subdermal level and using them as a carrier. The details and the application of this technique are presented and reported.

        Photodynamic antimicrobial activity of new porphyrin derivatives against methicillin resistant Staphylococcus aureus

        Methicillin resistant Staphylococcus aureus (MRSA) with multiple drug resistance patterns is frequently isolated from skin and soft tissue infections that are involved in chronic wounds. Today, difficulties in the treatment of MRSA associated infections have led to the development of alternative approaches such as antimicrobial photodynamic therapy. This study aimed to investigate photoinactivation with cationic porphyrin derivative compounds against MRSA in in-vitro conditions. In the study, MRSA clinical isolates with different antibiotic resistance profiles were used. The newly synthesized cationic porphyrin derivatives (PM, PE, PPN, and PPL) were used as photosensitizer, and 655 nm diode laser was used as light source. Photoinactivation experiments were performed by optimizing energy doses and photosensitizer concentrations. In photoinactivation experiments with different energy densities and photosensitizer concentrations, more than 99% reduction was achieved in bacterial cell viability. No decrease in bacterial survival was observed in control groups. It was determined that there was an increase in photoinactivation efficiency by increasing the energy dose. At the energy dose of 150 J/cm2 a survival reduction of over 6.33 log10 was observed in each photosensitizer type. While 200 μM PM concentration was required for this photoinactivation, 12.50 μM was sufficient for PE, PPN, and PPL. In our study, antimicrobial photodynamic therapy performed with cationic porphyrin derivatives was found to have potent antimicrobial efficacy against multidrug resistant S. aureus which is frequently isolated from wound infections.

        Improved Equations for the Profile of a Vertical Air-Core Vortex

        The available formulas for the profile of a vertical non-air-entraining vortex are not practical because they involve unknown parameters and need measurements of the local viscous-core radii across the height of the air-core vortex. Also, these formulas can not be used for the air-entraining vortices. In the present study, the available formulas involving unknown parameters were further improved. Findings are as follows. 1) the magnitudes of the unknown parameters vary across the height of the profile of the air-core vortex and they are the function of the ratio of the height of the air-core vortex to the submergence of the intake; 2) simple charts and formulas were obtained for the variations of the unknown parameters for the air-core vortices with and without an intake; 3) in lieu of the local viscous-core radius, the radius at the half-depth of the profile of the air-core vortex was used; 4) no laborious work of measurements are needed to determine the local viscous-core radii along the profile of the vortex; and 5) the improved formulas are in good agreement with available test data for the profiles of both the non-air-entraining and the air-entraining vortices with or without a vertically flowing downward intake.
