        Two Unrecorded Species of the Snapper (Perciformes: Lutjanidae) Collected from Jeju Island, Korea

        Two unrecorded species of the snapper, Lutjanus malabaricus (296.0 mm standard length [SL]) and L. stellatus (350.0 mm SL) belonging to the family Lutjanidae, were first collected from the western coastal waters of Jeju Island, Korea. L. malabaricus is characterized by having a dark marking on the upper half of the caudal peduncle, a band of vomerine teeth that does not protrude posteriorly at the middle, and nine anal soft rays. Compared to its Korean relative, L. malabaricus is distinguishable by having rows of scales that run obliquely in the dorsal-posterior direction above the lateral line (vs. parallel to the lateral line in L. argentimaculatus). L. stellatus can be identified by the absence of wavy blue lines on the head (vs. many blue lines in L. rivulatus) and presence of a white spot above the lateral line (vs. a black blotch on the lateral line in L. rivulatus). We propose new Korean names, Jin-hong-tung-dom” and “Huin-jeom-tung-dom,” for L. malabaricus and L. stellatus, respectively.

      • Mutational analysis of mononucleotide repeats in dual specificity tyrosine phosphatase genes in gastric and colon carcinomas with microsatellite instability

APMIS 2010; 118: 389–93.</P><P>Coordinated protein phosphorylation and dephosphorylation are crucial in the regulation of cell signaling, and disruption of the coordination is known to play important roles in cancer development. Recent reports revealed that classical protein tyrosine phosphatase (PTP)-encoded genes are somatically mutated in human colorectal cancer. However, data on dual specificity phosphatase (DPTP) gene mutations in human cancers are lacking. By analyzing a public genomic database, we found that five DPTP genes, <I>CDC14A</I>, <I>MTM1</I>, <I>MTMR3</I>, <I>SSH1</I>, and <I>SSH2</I>, have mononucleotide repeats in their coding DNA sequences. To see whether these genes are mutated in cancers with microsatellite instability (MSI), we analyzed the mononucleotide repeats in 26 gastric cancers (GC) with MSI (MSI-H), 12 GC with low MSI (MSI-L), 45 GC with stable MSI (MSS), 33 colorectal cancers (CRC) with MSI-H, 14 CRC with MSI-L, and 45 CRC with MSS by single-strand conformation polymorphism (SSCP). We found <I>CDC14A</I> and <I>MTMR3</I> mutations in five and one cancer (s), respectively. These mutations were detected in MSI-H cancers, but not in MSI-L or MSS cancers. The GC and CRC with MSI-H harbored the mutations in 15% and 6%, respectively. The <I>CDC14A</I> and <I>MTMR3</I> mutations detected in the GC and CRC were deletion or duplication mutations of one base in the nucleotide repeats that would result in premature stops of the amino acid syntheses. Our data show that frameshift mutations of DPTP genes in MSI-H cancers occur at moderate frequencies. The data suggested that alterations in the <I>CDC14A</I> and <I>MTMR3</I> genes may play a role in the development of GC and CRC with MSI-H by deregulating phosphatase functions possibly together with mutations of classical PTP genes.</P>

        v-Crk Induces Rac-dependent Membrane Ruffling and Cell Migration in CAS-deficient Embryonic Fibroblasts

        Crk-associated substrate (CAS) is a focal adhesion protein that is involved in integrin signaling and cell migration. CAS deficiency reduces the migration and spreading of cells, both of which are processes mediated by Rac activation. We examined the functions of v-Crk, the oncogene product of the CT10 virus p47gag-crk, which affects cell migration and spreading, membrane ruffling, and Rac activation in CAS-deficient mouse embryonic fibroblasts (CAS-/- MEFs). CAS-/- MEFs showed less spreading than did CAS+/+ MEFs, but spreading was recovered in mutant cells that expressed v-Crk (CAS-/-v-Crk MEF). We observed that the reduction in spreading was linked to the formation of membrane ruffles, which were accompanied by Rac activation. In CAS-/- MEFs, Rac activity was significantly reduced, and Rac was not localized to the membrane. In contrast, Rac was active and localized to the membrane in CAS-/-v-Crk MEFs. Lamellipodia protrusion and ruffle retraction velocities were both reduced in CAS-/- MEFs, but not in CAS-/-v-Crk MEFs. We also found that microinjection of anti-gag antibodies inhibited the migration of CAS-/-v-Crk MEFs. These findings indicate that v-Crk controls cell migration and membrane dynamics by activating Rac in CAS-deficient MEFs.

        ppGpp-Mediated Stationary Phase Induction of the Genes Encoded by Horizontally Acquired Pathogenicity Islands and cob/pdu Locus in Salmonella enterica serovar Typhimurium

        Salmonella enterica is highly diverse in terms of genome structure, which is at least partly due to the horizontal transfer of genetic elements from various sources. In this study, we examined the expression profiles of such genes in $\underline{S}$almonella $\underline{P}$athogenicity $\underline{I}$slands (SPIs) and the cob/pdu locus, horizontally acquired large DNA segments, during growth under standard growth conditions. Transcripts from exponentially growing and early stationary phase Salmonellae were compared using various methods including cDNA microarray analysis. Nearly all genes encoded by SPIs and the cob/pdu locus were induced at the onset of the stationary phase in a stringent molecule ppGpp-dependent but stationary phase ${\sigma}$, ${\sigma}$38-independent manner. Although, it has been suggested that ppGpp acts in concert with DksA, we found the stationary phase induction of those SPI genes was not DksA dependent. It is suggested that ppGpp stimulates the expression of these stress-inducible genes encoded by horizontally acquired DNA, by itself or in concert with DksA.

        A low-bandgap alternating copolymer containing the dimethylbenzimidazole moiety

        <P>A new acceptor unit containing dimethyl-2<I>H</I>-benzimidazole was synthesized and utilized for the synthesis of a conjugated polymer containing electron donor–acceptor pairs for OPV devices. The dimethyl-2<I>H</I>-benzimidazole unit was designed to act as a substitute for the BT unit of <B>PCDTBT</B>. This novel dimethyl-2<I>H</I>-benzimidazole unit has two methyl groups which can supply higher solubility than those of the BT series. A thin film of <B>PCDTMBI</B>, containing the dimethyl-2<I>H</I>-benzimidazole unit, shows two broad absorption bands with maxima at 400 and 636 nm and an absorption onset of 756 nm, corresponding to a band gap of 1.64 eV. The oxidation onset of <B>PCDTMBI</B> was estimated to be 0.67 V, which corresponds to a HOMO energy level of −5.47 eV. The LUMO energy level of the polymer was thus determined to be −3.82 eV. A device with a <B>PCDTMBI</B>:PC<SUB>71</SUB>BM blend had a <I>V</I><SUB>OC</SUB> value of 0.65 V, a <I>J</I><SUB>SC</SUB> value of 10.0 mA cm<SUP>−2</SUP>, and a <I>FF</I> of 0.48, leading to an efficiency of 3.12%.         Background: We investigated how many patients with Raynaud`s phenomenon (RP), but not systemic sclerosis (SSc) would be reclassifi ed as SSc, and we analyzed the predictive values for the reclassifi cation as SSc using the 2013 ACR/EULAR classifi cation criteria Methods: We consecutively enrolled 64 patients with RP and 60 patients with SSc. We applied the new classifi cation criteria to patients with RP, reclassifi ed them, and compared variables between those who were newly reclassifi ed as SSc and those who were not or previously diagnosed as SSc. The predictive values for the reclassifi cation as SSc were also assessed. Results: Seventeen of 64 patients (26. 5%) with RP were newly classifi ed as SSc according to the 2013 ACR/EULAR classifi cation criteria. Patients newly classifi ed as SSc had sclerodactyly, digital tip ulcer, telangiectasia, abnormal nailfold capillaries and the presence of anti-centromere antibody more frequently than those not. The independent predictive values for the reclassifi cation as SSc were sclerodactyly (OR 60. 025),telangiectasia (OR 13. 353) and the presence of anti-centromere antibody (OR 11. 168). Telangiectasia and anti-centromere antibody in newly classifi ed patients were more frequently than those in previously diagnosed SSc patients. Conclusions: 26. 5% of patients with RP were newly classifi ed as SSc by the 2013 ACR/EULAR classifi cation criteria, and sclerodactyly, telangiectasia, and the presence of anti-centromere antibody were the independent predictive values for the reclassifi - cation as SSc.

        Effect of Inclusion of Lacquer (Rhus verniciflua Stokes) Meal on Carcass Traits and Meat Quality in Growing-finishing Pigs

        In this study, pigs [n=117; (Landrace ${\times}$ Yorkshire) ${\times}$ Duroc; $64{\pm}0.5$ Kg initial body weight] were used to investigate the effect of feeding different levels of lacquer (Rhus verniciflua Stokes) meal on performance, carcass traits and quality of meat kept under refrigeration at $3{\pm}1^{\circ}C$. The pigs were randomly allotted to 3 treatments on the basis of body weight and sex and each treatment was replicated 3 times (13 pigs in each replicate). Lacquer meal in sawdust form obtained from the stem bark and heartwood of sun-dried lacquer trees was added to the grower and finisher diets at 0, 20 and 40 g/kg diet. The experimental diets were fed for 8 wk. Inclusion of lacquer meal had no influence (p>0.05) on growth performance of pigs. Improvement in carcass traits and decreased back fat thickness were noticed in pigs fed diets added with 20 and 40 g/kg lacquer meal. Longissimus muscle obtained from pigs fed lacquer meal had higher moisture and lower fat content, thiobarbituric acid reactive substances and water holding capacity. Meat from lacquer fed pigs was also darker and redder. The data indicates that lacquer meal can be incorporated up to 40 g/kg in the diet of fattening pigs without affecting growth performance. Also, lacquer meal increases carcass lean content and improves the oxidative stability of the meat.

Since different structures provide different coordination geometry of each metal ion, it is clear that selection of appropriate metal ions can control the coordination geometry of each metal ion to form different crystal structures. Reactivity study of seven compounds for the transesterification of a variety of esters has shown that <B>4</B>-Zn and <B>5</B>-Zn are very efficient and the best among them. Reactivity study of the compounds <B>1</B>–<B>7</B> for the transesterification of a variety of esters has shown that <B>4</B>-Zn and <B>5</B>-Zn are very efficient and the best among them. The catalyst <B>6</B>-Cd containing Cd ion, well known as an inert metal ion for the ligand substitution, also catalyzed efficiently the transesterification of a variety of esters, and its reactivity is comparable to <B>4</B>-Zn and <B>5</B>-Zn. Moreover, the redox-active metal-containing polymers, <B>1</B>-Co, <B>3</B>-Cu, and <B>7</B>-Mn, have shown efficient catalytic reactivities for the transesterification reactions, while <B>2</B>-Ni has displayed a very slow conversion. The reactivities of the compounds used in this study are in the order of <B>5</B>-Zn><B>4</B>-Zn><B>6</B>-Cd><B>7</B>-Mn∼<B>3</B>-Cu><B>1</B>-Co><B>2</B>-Ni, indicating that the non-redox metal-containing compounds (<B>5</B>-Zn, <B>4</B>-Zn, and <B>6</B>-Cd) show better activity than the redox-active metal-containing compounds (<B>7</B>-Mn, <B>3</B>-Cu, <B>1</B>-Co, and <B>2</B>-Ni). These results suggest that it is possible to tune the catalytic activities by changing from Zn to those metals such as Cd, a kinetically inert metal, or Cu, Mn, and Co, the redox-active metals.</P>

      • Robot-Assisted Gastrectomy With Lymph Node Dissection for Gastric Cancer: Lessons Learned From an Initial 100 Consecutive Procedures

        OBJECTIVE:: To evaluate the technical feasibility, effectiveness, and safety of robot-assisted gastrectomy (RAG) with lymphadenectomy, using the da Vinci system through analyses of our initial series of 100 consecutive patients. SUMMARY BACKGROUND DATA:: The application of robotic surgery was proven to be one of the best cutting-edge technologies for successful minimally invasive surgery by providing solutions to the many drawbacks of laparoscopic surgery, yet few reports have studied robotic surgery in gastric cancer. METHODS:: A review of a prospectively designed database at our institute from July 2005 to October 2007 revealed a series of 100 consecutive RAG patients with a preoperative diagnosis of early gastric cancer. Clinicopathologic characteristics and surgical outcomes were analyzed. RESULTS:: All operations were performed successfully without open or laparoscopic conversion. There were 33 total gastrectomies and 67 subtotal gastrectomies with D1+&bgr; or extended lymphadenectomy (D2). The mean total operation time and console time were 231 and 150 minutes, respectively. There were 13 postoperative morbidities and 1 postoperative mortality. The first flatus was noted on postoperative day 2.9, soft diet was started on postoperative day 4.2, and the mean postoperative hospital stay was 7.8 days. Although all patients were diagnosed as early gastric cancer preoperatively, the final pathology report revealed that 19 patients exhibited a depth deeper than T2. The mean number of retrieved lymph nodes was 36.7 (range, 11–83). None of the specimens showed microscopic tumor involvement in the resection line. CONCLUSIONS:: This study demonstrated that RAG with lymphadenectomy can be applied safely and effectively for patients with gastric cancer.
