      • SIRT7 Exhibits Oncogenic Potential in Human Ovarian Cancer Cells

Wang, Hong-Ling,Lu, Ren-Quan,Xie, Su-Hong,Zheng, Hui,Wen, Xue-Mei,Gao, Xiang,Guo, Lin Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.8

Background: Sirtuin7 (SIRT7) is a type of nicotinamide adenine dinucleotide oxidized form (NAD+)-dependent deacetylase and the least understood member of the sirtuins family; it is implicated in various processes, such as aging, DNA damage repair and cell signaling transduction. There is some evidence that SIRT7 may function as a tumor trigger for human malignancy. Here, we aimed to explore the biological function of SIRT7 in ovarian carcinoma cells and its potential mechanism. Materials and Methods: Expression of SIRT7 in ovarian cancer cell lines was detected by western blotting. Transduced cell lines with SIRT7 knockdown or overexpression were constructed. Cell viability, cologenic, apoptosis-associated and motility assays were performed to elucidate the biological function of SIRT7 in ovarian cancer cells. Results: SIRT7 demonstrated a higher level in ovarian cancer cell lines compared with normal cells. On the one hand, down-regulation of SIRT7 significantly reduced ovarian cancer cell growth, repressed colony formation and increased cancer cell apoptosis; on the other hand, up-regulation promoted the migration of cancer cells. Additionally, repression of SIRT7 also induced change in apoptosis-related molecules and subunits of the NF-${\kappa}B$ family. Conclusions: In the present study, our data indicated that SIRT7 might play a role of oncogene in ovarian malignancy and be a potential therapeutic target.

      • Transient Knock Down of Grp78 Reveals Roles in Serum Ferritin Mediated Pro-inflammatory Cytokine Secretion in Rat Primary Activated Hepatic Stellate Cells

        Wang, Chi-Mei,Li, Shan-Jen,Wu, Chi-Hao,Hu, Chien-Ming,Cheng, Hui-Wen,Chang, Jung-Su Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.2

Chronic liver diseases, including cancer, are characterized by inflammation and elevated serum ferritin (SF). However, the causal-relationship remains unclear. This study used primary rat hepatic stellate cells (HSC) as a model to investigate effects of physiological SF concentrations (10, 100 and 1000 pM) because HSCs play a central role in the development and progression of liver fibrosis. Physiological concentrations of SF, either horse SF or human serum, induced pro-inflammatory cytokine $IL1{\beta}$, IL6 and $TNF{\alpha}$ secretion in rat activated HSCs (all p<0.05). By contrast, treatment did not alter activation marker ${\alpha}SMA$ expression. The presence of SF markedly enhanced expression of Grp78 mRNA (p<0.01). Furthermore, transient knock down of Grp78 by endotoxin EGF-SubA abolished SF-induced $IL1{\beta}$ and $TNF{\alpha}$ secretion in activated HSCs (all p<0.05). In conclusion, our results showed that at physiological concentrations SF functions as a pro-inflammatory mediator in primary rat HSCs. We also provide a molecular basis for the action of SF and identified Grp78-associated ER stress pathways as a novel potential therapeutic target for resolution of fibrosis and possible prevention of liver cancer.

Influence of some key factors on material damping of steel beams

        Wang, Yuanfeng,Pan, Yuhua,Wen, Jie,Su, Li,Mei, Shengqi Techno-Press 2014 Structural Engineering and Mechanics, An Int'l Jou Vol.49 No.3

Material damping affects the dynamic behaviors of engineering structures considerably, but up to till now little research is maintained on influence factors of material damping. Based on the damping-stress function of steel, the material damping of steel beams is obtained by calculating the stress distribution of the beams with an analytical method. Some key influence factors of the material damping, such as boundary condition, amplitude and frequency of excitation, load position as well as the cross-sectional dimension of a steel beam are analyzed respectively. The calculated results show that even in elastic scope, material damping does not remain constant but varies with these influence factors. Although boundary condition affects material damping to some extent, such influence can be neglected when the maximum stress amplitude of the beam is less than the fatigue limit of steel. Exciting frequency, load position and cross-section dimension have great effects on the material damping of the beam which maintain the similar changing trend under different boundary conditions respectively.

Seismic analysis of bridges based on stress-dependent damping

        Li Su,Yuanfeng Wang,Pengfei Li,Shengqi Mei,Kun Guo 국제구조공학회 2017 Structural Engineering and Mechanics, An Int'l Jou Vol.62 No.3

Damping value has considerable influence on the dynamic and seismic behaviors of bridges. However, currently the constant damping ratios that are prescribed by most bridge seismic design codes can’t truly represent the complicated damping character of actual structures. In this paper, a cyclic loading experiment was conducted to study the effect of stress amplitude on material damping of concrete to present an analyzing model of the material damping of concrete. Furthermore, based on the fundamental damping of structure measured under ambient vibration, combined with the presented stress-dependent material damping concrete, the seismic response of a bridge pier was calculated. Comparison between the calculated and experiment results verified the validity of the presented damping model. Finally, a modified design and analysis method for bridge was proposed based on stress-dependent damping theory, and a continuous rigid frame bridge was selected as the example to calculate the actual damping values and the dynamic response of the bridge under different earthquake intensities. The calculation results indicated that using the constant damping given by the Chinese seismic design code of bridges would overestimate the energy dissipation capacity of the bridge.

High-efficiency degradation of quinclorac via peroxymonosulfate activated by N-doped CoFe2O4/Fe0@CEDTA hybrid catalyst

        Lezhu Su,Lizhou Ou,Yujiao Wen,Yifan Wang,Weilin Zhao,Zhi Zhou,Mei-e Zhong,YONGFA ZHU,Nan Zhou 한국공업화학회 2021 Journal of Industrial and Engineering Chemistry Vol.102 No.-

Exploring catalyst materials which are advancing, recyclable and with high catalytic performances toremove persistent organic pollutants such as Quinclorac (QNC) is important. In this work, a novel Ndopedcarbon support CoFe2O4/Fe0 hybrid catalyst (CFO/Fe@C) was in situ formed by a simple coprecipitationand calcination process. Fine intergrowth crystal CoFe2O4 and Fe0 were uniformly dispersedon the N-doped porous carbon that derived from the raw material Ethylene Diamine Tetraacetic Acid(EDTA), which also provided chelating effect to prevent the agglomeration of the metals. Interestingly, the Fe0 could only be formed with the presence of cobalt, possibly due to the increased reduction propertyresulted from the particle refinement. The as-formed Fe0 could not only activate the peroxymonosulfate(PMS) but also reduce the Co3+, resulting in a synergistic impact to remarkably enhance thedegradation performance. Besides, the N-doped porous carbon can also benefit the degradation of pollutantby strengthening the electron transfer. A good degradation efficiency of QNC was obtained in CFO/Fe@C-PMS system and most of the QNC had been degraded to carbon dioxide, water and other smallmolecular organisms. The removal rate remained over 70% after four reuses and the material could beeasily recovered from the solution due to the good magnetic properties. Therefore, the as-preparedCFO/Fe@C catalyst should be an ideal catalyst for the removal of organic pollutants.

Degradation of 1,4-dioxane by Newly Isolated Acinetobacter sp. M21 with Molasses as the Auxiliary Substrate

        Peng Wang,Tingchen Cui,Yadong Yang,Jialu Li,Yaoming Su,Na Liu,Hong-Mei Li 한국생물공학회 2022 Biotechnology and Bioprocess Engineering Vol.27 No.3

The elimination of 1,4-dioxane (dioxane), a persistent organic pollutant, is a great challenge owing to its high hydrophilicity and chemical stability. Cometabolic bioremediation technology is an effective approach to remove many organic pollutants. Because of its eco-friendly and inexpensive properties, molasses is widely used as an auxiliary biomaterial to clean up compound-contaminated sites. In this study, a newly isolated bacterium Acinetobacter sp. M21 could effectively remove dioxane using molasses without any apparent lag phase. Under the optimized molasses dosage of 0.3%, M21 could remove 500 mg/L dioxane by 60.0 ± 2.8% within 20 days with a maximum dioxane degradation rate of 1.3 ± 0.2 mg-dioxane/L/h in the first day, and exhibited extraordinary dioxane tolerance up to 1,000 mg/L, while so high dose of dioxane negatively affected the cell growth. The degradation pathway of dioxane was also determined, and was supported by the detection of 2-hydroxyethoxyacetic acid as the key metabolite of dioxane. High level degradation activity of M21 to 20 mg/L dioxane was maintained over a variable of pH (5-11), temperatures (15-45°C), and salinities (up to 8%, as NaCl wt). This is the first report linking the cometabolism of dioxane and molasses by Acinetobacter sp. M21, a bacterium that shows great potential for field dioxane bioremediation.

Synthesis, Crystal Structure and Density Functional Calculationson 1-Phenyl-3-p-fluorophenyl-5-p-chlorophenyl-2-pyrazoline

Pu Su Zhao,Yu Feng Li,Huan Mei Guo,Fang Fang Jian*,Xian Wang 대한화학회 2007 Bulletin of the Korean Chemical Society Vol.28 No.9

1-Phenyl-3-p-fluorophenyl-5-p-chlorophenyl-2-pyrazoline has been synthesized and characterized by elemental analysis, IR, UV-Vis and X-ray single crystal diffraction. Density functional calculations show that B3LYP/6-311G** method can reproduce the structural parameters. The electronic absorption spectra have been predicted based on the optimized structure by using 6-311G** and 6-311++G** basis sets and compared with the experimental values. The results indicate that TD-DFT method can only predict the electronic absorption spectra of the system studied here approximately. On the basis of vibrational analyses, the thermodynamic properties of the title compound at different temperatures have been calculated, revealing the correlations between , , and temperature.
