        Post-cancer Treatment with Condurango 30C Shows Amelioration of Benzo[a]pyrene-induced Lung Cancer in Rats Through the Molecular Pathway of Caspa- se-3-mediated Apoptosis Induction -Anti-lung cancer potential of Condurango 30C in rats-

        Objectives: The present investigation aimed at examining if post-cancer treatment with a potentized homeopathic drug, Condurango 30C, which is generally used to treat oesophageal cancer, could also show an ameliorating effect through apoptosis induction on lung cancer induced by benzo[a]pyrene (BaP) in white rats (Rattus norvegicus). Methods: Lung cancer was induced after four months by chronic feeding of BaP to rats through gavage at a dose of 50 mg/kg body weight for one month. After four months, the lung-cancer-bearing rats were treated with Condurango 30C for the next one (5th), two (5th -6th) and three (5th-7th) months, respectively, and were sacrificed at the corresponding time- points. The ameliorating effect, if any, after Condurango 30C treatment for the various periods was evaluated by using protocols such as histology, scanning electron microscopy (SEM), annexinV-FITC/PI assay, flow cytometry of the apoptosis marker, DNA fragmentation, reverse transcriptase-polymerase chain reaction (RT-PCR), immunohistochemistry, and western blot analyses of lung tissue samples. Results: Striking recovery of lung tissue to a near normal status was noticed after post-cancerous drug treatment, as evidenced by SEM and histology, especially after one and two months of drug treatment. Data from the annexinV-FITC/PI and DNA fragmentation assays revealed that Condurango 30C could induce apoptosis in cancer cells after post-cancer treatment. A critical analysis of signalling cascade, evidenced through a RT-PCR study, demonstrated up-regulation and down-regulation of different pro- and anti-apoptotic genes, respectively, related to a caspase-3-mediated apoptotic pathway, which was especially discernible after one-month and two- month drug treatments. Correspondingly, Western blot and immunohistochemistry studies confirmed the ameliorative potential of Condurango 30C by its ability to down-regulate the elevated epidermal growth factor receptor (EGFR) expression,a hallmark of lung cancer. Conclusion: The overall result validated a positive effect of Condurango 30C in ameliorating lung cancer through caspase-3-mediated apoptosis induction and EGFR down-regulation.

      • Fluorometric detection of nitroaromatics by fluorescent lead complexes: A spectroscopic assessment of detection mechanism

        <P><B>Abstract</B></P> <P>Three Schiff base ligands such as 2‑[(2‑Hydroxy‑3‑methoxy‑benzylidene)‑amino]‑2‑hydroxymethyl‑propane‑1,3‑diol (<B>HL1</B>), 2‑[(2‑Hydroxy‑benzylidene)‑amino]‑2‑hydroxymethyl‑propane‑1,3‑diol (<B>HL2</B>), 2‑[(3,5‑Dichloro‑2‑hydroxy‑benzylidene)‑amino]‑2‑hydroxymethyl‑propane‑1,3‑diol (<B>HL3</B>) have been synthesized by condensation of aldehydes (such as 3,5‑Dichloro‑2‑hydroxy benzaldehyde, 2‑Hydroxy‑benzaldehyde, and 2‑Hydroxy‑3‑methoxy‑benzaldehyde) with <I>Tris</I>‑(hydroxymethyl)amino methane and characterized by IR, UV–vis and <SUP>1</SUP>H NMR spectroscopy. Then all these three ligands have been used to prepare Pb(II) complexes by reaction with lead(II) acetate tri-hydrate in methanol. In view of analytical and spectral (IR, UV–vis and Mass) studies, it has been concluded that, except <B>HL2</B>, other two ligands form 1:1 metal complexes (<B>1</B> and <B>3</B>) with lead. Between two complexes, complex <B>3</B> is highly fluorescent and this property has been used to identify the pollutant nitroaromatics.       • Metabolomics affirms traditional alcoholic beverage raksi as a remedy for high-altitude sickness

        Raksi, a fermented distilled alcoholic beverage, is an ethnic drink consumed in high altitude regions of Singalila Ridge of the Himalayas and in adjoining high altitude places in Nepal, northern and north eastern part of India and Tibetan plateau. Like jaanr, tongba, nigar, chhyang, and other fermented ethnic beverages, raksi is considered as an element of ethnopharmacology of high altitude with claims of medicinal properties. An ethnobiological survey was done in the study area prior to collection and identification of raksi samples. In this research, two raksi samples (khokim raksi and chimphing raksi) of Singalila Ridge of the Himalayas were investigated by metabolite profiling using gas chromatography-mass spectrometry analysis. Results of the experiment showed presence of several respiratory protective, cardioprotective, neuroprotective, anti-inflammatory, and antioxidant components which have properties to prevent various high altitude illnesses. Moreover, large quantities of bioactive terpenoids, fatty acid derivatives, coumarins, and peptides were detected whose chemotaxonomy and biosynthesis pathways were further studied. This metabolomics investigation not only affirmed Raksi as a remedy for high-altitude sickness but also helped in understanding the importance of this type of ethnic foods in high altitude ethnoecology. This research on raksi in the fields of ethnobiology and metabolomics is a cumulative approach which has opened the door for in-depth investigations on similar ethnic foods. However, further experiments on raksi are surely needed in ethnobiology, microbiology, biochemistry, and food technology.

        Understanding Bacterial Biofilm Stimulation Using Different Methods - a Criterion for Selecting Epiphytes by Plants

        Earlier studies by our group revealed that gallic acid in phytochemicals stimulated biofilm production in epiphytes, while caffeic acid in phytochemicals inhibited biofilm production in non-epiphytes. It is well documented that antimicrobial secretion by some epiphytic bacteria inhibits non-epiphytic bacterial growth on leaf surfaces. These selection criteria help plants choose their microbial inhabitants. Calcium and iron in phytochemicals also stimulate biofilm formation and thus, may be selection criteria adopted by plants with respect to their native epiphytic population. Furthermore, the processing of leaves during phytochemical extraction impacts the composition of the extract, and therefore its ability to affect bacterial biofilm formation. Computation of the Hurst exponent using biofilm thickness data obtained from the Ellipsometry of Brewster Angle Microscopic (BAM) images is an efficient tool for understanding the impact of phytochemicals on epiphytic and non-epiphytic populations when compared to fluorescent microscopy, scanning electron microscopy, and staining techniques. To the best of our knowledge, this is the first report that uses the Hurst exponent to elucidate the mechanism involved in plant microbe interaction.

        Seasonal Variability of Marine Fish Diversity in Relation to Water Quality of East Midnapore Coast of West Bengal, India

        Marine fishes are one of the important factors in stabilizing the local aquatic ecosystem and regulating the nutritional socioeconomy of local fisher folks. The recent increases in anthropogenic activity, pollution and overfishing have led to the decline of marine fish species richness and their local aquatic habitats. In this study we sought to determine the inter-relationship between water quality, anthropogenic activity, and fish landing stations through a 31 km stretch of the East Midnapore coast in West Bengal, India which is known for its tourist destinations. The study was conducted monthly on different trawler fish landing sites from Dec 2018 to Dec 2021. During this period, we took fish samples and identified them. We obtained water quality data regarding Sea Surface Temperature (SST), Concentration of Chlorophyll-a (Chl-a), Turbidity, and Dissolved Oxygen (DO) in order to identify further correlation between the water quality analysis and species diversity. 154 numbers of commercially important marine fish species were documented. As per the IUCN database, 13% of the total fish species fall under the red list category and 16% of the species reveal a decreasing population trend. The availability of those red-listed fish throughout the season has been shown in the matrix plot to detect their gradual decrease in sighting. After analyzing the water quality data, we found out that DO, SST, Turbidity, and Chl-a correlate with the species richness on some sites and the water parameters are also differs during the seasons. Both fish species richness and water quality have been affected on those fish landing sites which have been subject to heavy anthropogenic loads.
