        Three-Dimensional Skin Tissue Printing with Human Skin Cell Lines and Mouse Skin-Derived Epidermal and Dermal Cells

        Since the skin covers most surfaces of the body, it is susceptible to damage, which can be fatal depending on the degree of injury to the skin because it defends against external attack and protects internal structures. Various types of artificial skin are being studied for transplantation to repair damaged skin, and recently, the production of replaceable skin using three-dimensional (3D) bioprinting technology has also been investigated. In this study, skin tissue was produced using a 3D bioprinter with human skin cell lines and cells extracted from mouse skin, and the printing conditions were optimized. Gelatin was used as a bioink, and fibrinogen and alginate were used for tissue hardening after printing. Printed skin tissue maintained a survival rate of 90% or more when cultured for 14 days. Culture conditions were established using 8 mM calcium chloride treatment and the skin tissue was exposed to air to optimize epidermal cell differentiation. The skin tissue was cultured for 14 days after differentiation induction by this optimized culture method, and immunofluorescent staining was performed using epidermal cell differentiation markers to investigate whether the epidermal cells had differentiated. After differentiation, loricrin, which is normally found in terminally differentiated epidermal cells, was observed in the cells at the tip of the epidermal layer, and cytokeratin 14 was expressed in the lower cells of the epidermis layer. Collectively, this study may provide optimized conditions for bioprinting and keratinization for three-dimensional skin production.

      • A case of multiple, severe adverse effects of immunotherapy after complete remission of lung cancer

        Introduction: Although immunotherapy is promising modality of cancer management, adverse effects are often unpredictable and rapidly change. We experienced a case of lung cancer with multiple, severe adverse effects. Especially in this case, it was difficult to decide whether high dose steroids were infused immediately after recovery from diabetic ketoacidosis. Case Presentation; A 65-year-old male patient was diagnosed as squamous cell carcinoma of lung, and recurred after surgical resection. PET-CT(Fig. A) scan showed bony matastasis. On immunohistochemical staining, PD-L1 was 100% positive. The lesion regressed after pembrolizumab. On 7th cycle, dyspnea, wheeze, urticaria occurred and relieved by peniramine, hydrocortisone which features were consistent with angioedema. Follow-up PET-CT scan(Fig. B) showed complete remission. At that time he complained of dry mouth and FBS was 178 mg/dL. Two days later, he visited emergency room for weakness. Blood test was consistent with diabetic ketoacidosis: glucose 711 mg/dL, C-peptide 0.74 ng/mL, pH 7.185, ketone 8.49 mMol/L. As there is no history of diabetes, it was thought as a side effect of pembrolizumab. Seven days later, blood glucose level returned to 182 mg/dL with insulin. The day before his discharge, he showed abnormal behavior. Twelve hours later, generalized seizure was occurred. Brain MRI finding was nonspecific, and EEG finding was consistent with diffuse encephalopathy. Consciousness level aggravated to semicoma. High dose steroids(methyl prednisolone 1 g/day for 3 days) was started. Two days later, mental status improved to stupor. However, after 3 months of fluctuating mental status, he died of pneumonia. Discussion: Encephalitis is rare, but potentially fatal adverse events of immune checkpoint inhibitors. Prompt use of high dose steroids is essential. In this case, it was very difficult decision to prescribe that. Although it is not always possible, we should be alert to all the possible, rare side effects of therapy.

        Millettia erythrocalyx 에탄올 추출물의 항산화 활성 및 항암 활성에 관한 연구

        Millettia erythrocalyx는 콩과(Fabaceae)에 속하는 식물로 중국, 태국, 인도 등 열대·아열대 지역에 분포하며, 항바이러스 활성을 보유하고 있다는 보고가 있으나 항산화능과 항암활성 등에 관한 연구는 보고된 바가 없다. 따라서 본 연구에서는 M. erythrocalyx의 에탄올 추출물(EEME)을 사용하여 항산화능을 측정하고, 인체간암세포주인 HepG2에 대한 항암활성과 그 분자적 기전에 관하여 분석하였다. 먼저 DPPH radical scavenging activity를 통해 분석한 결과, EEME의 IC50는 2.74 μg/ml로 뛰어난 항산화능을 보유하였음을 확인하였다. 또한 EEME 농도 의존적으로 HepG2 세포의 성장을 억제하였다. EEME의 HepG2 세포 사멸 효과의 기전을 분석하기 위하여 세포주기를 분석한 결과, EEME 농도의존적으로 SubG1 세포가 증가하였으며, Annexin V 염색과 DAPI 염색을 통해 apoptotic 세포 및 apoptotic body가 증가됨을 확인하였다. 또한 apoptosis 관련 단백질들의 발현변화를 분석한 결과, EEME 처리에 의해 사멸수용체인 Fas와 pro-apoptotic 단백질인 Bax의 발현이 증가되었으며, caspase-3, -8, -9가 활성화되고 최종적으로 PARP가 분해되어 apoptosis가 유도되었음을 확인하였다. 이러한 결과들로부터 EEME는 내인성 및 외인성 경로를 통한 apoptosis 유도에 의하여 HepG2 세포의 증식을 억제하는 항암활성을 보유하였음을 확인하였다. Millettia erythrocalyx, a species of plant in the Fabaceae family, is widely distributed in the tropical and subtropical regions of the world, such as the Indies, China, and Thailand. The antiviral activity of flavonoids from M. erythrocalyx has been reported; however, the antioxidative and anticancer activities of M. erythrocalyx remain unclear. In this study, we evaluated the antioxidative and anticancer effects of ethanol extract of M. erythrocalyx (EEME) and the molecular mechanism of its anticancer activity in human hepatocellular carcinoma HepG2 cells. EEME exhibited significant antioxidative effects, with a concentration at 50% inhibition (IC50) value of 2.74 μg/ml, as measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay; moreover, it inhibited cell proliferation in a dosedependent manner in HepG2 cells. Cell cycle analyses showed that EEME induced HepG2 cell accumulation in the subG1 phase in a dose-dependent manner. EEME also induced apoptosis of HepG2 cells, with increases in apoptotic cells and apoptotic bodies, as detected by Annexin V and 4,6-diamidino-2-phenylindole (DAPI) staining, respectively. Treatment with EEME resulted in increased expression of First apoptosis signal (Fas), a death receptor, and Bcl-2-associated X protein (Bax), a proapoptotic protein, and the activation of caspase-3, 8, and 9, resulting in the cleavage of poly (Adenosine diphosphate-ribose) polymerase (PARP). Collectively, these results suggest that EEME may exert an anticancer effect in HepG2 cells by inducing apoptosis via both the intrinsic and extrinsic pathways.

        인체 폐암 세포주 A549에서 Euonymus porphyreus 추출물의 항산화 및 항암활성 분석

        Euonymus porphyreus는 노박덩굴과에 속하는 식물로 동아시아 지역에 널리 분포하며, 식물학상의 특징에 대한 보고는 있으나 항산화능과 항암활성 등에 관한 연구는 아직까지 밝혀진 바가 없다. 이에 본 연구에서는 인체 폐암세포인 A549를 사용하여 E. porphyreus 에탄올 추출물(EEEP)의 항산화 및 항암활성과 그 분자적 기전에 관하여 연구하였다. 먼저 EEEP의 총 폴리페놀 화합물과 플라보노이드 함량을 측정한 결과, 각각 115.42 ㎎/g, 23.07 ㎎/g이었다. EEEP의 DPPH radical 소거활성을 측정한 결과, IC50가 11.09 ㎍/ml로 뛰어난 항산화능을 보유한 것을 확인하였다. 또한 EEEP는 농도의존적으로 인체폐암세포주인 A549의 세포 성장을 저해하였으며, 세포 주기변화를 분석한 결과 A549 세포의 SubG1기 세포비율이 증가하는 것을 확인하였다. Annexin V 염색과 DAPI 염색으로 EEEP 처리에 의해 apoptotic 세포와 apoptotic body가 증가하는 것을 확인하였으며, 이러한 결과는 EEEP에 의해 A549 세포의 apoptosis가 유도되는 것을 시사한다. 또한 관련 단백질들의 발현변화를 분석한 결과, EEEP에 의해 Fas, p53, Bax의 발현이 증가하고 Bcl-2의 발현은 감소하였으며, caspase-8, -9와 caspase-3의 활성화를 통해 PARP가 분해되어 apoptosis가 유도되었음을 확인하였다. 이러한 결과들로부터 EEEP는 내인성 및 외인성 경로를 통한 apoptosis 유도에 의하여 A549 세포의 증식을 억제하는 항암활성을 보유하였음을 확인하였다. Euonymus porphyreus, a species of plant in the Celastraceae family, is widely distributed in East Asia, especially in Southern China. The botanical characteristics of E. porphyreus have been reported, but its antioxidative and anticancer activities remain unclear. In this study, we evaluated the antioxidative and anticancer effects of ethanol extracts of E. porphyreus (EEEP) and the molecular mechanism of its anticancer activity in human lung adenocarcinoma A549 cells. The total polyphenol and flavonoid compound contents from EEEP were 115.42 ㎎/g and 23.07 ㎎/g, respectively.         사범대학의 교육실습체제 개선 사례 및 성과 분석

        이 연구는 ``교원양성교육 선도 사범대학 지원사업``에 선정되어 적극적으로 교육실습체제 개선을 추진한 A 사범대학의 사례와 성과를 분석하고 시사점을 확인함으로써, 그 성과를 공유하고 효과적인 결과들을 확산하려는데 목적이 있다. 2011년부터 2014년까지 4년간 사업비를 지원받은 A 사범대학은 사업 첫해부터 교육실습 개선을 위한 연구와 실행을 본격적으로 진행하였고, 참관실습-교육봉사-현장실습의 연계체제를 강화하고자 하였다. 현장교육실습 사전사후교육 강의와 현장교육실습 포트폴리오를 개발하는 등 교육실습 지원체제를 구축하였고, ``글로컬 교육실습``을 지향점으로 교육소외집단에 대한 실습과 봉사, 그리고 연변 조선족자치주에 대한 국제교육실습을 운영하였다. 현장교육실습 사전사후교육 및 성찰활동이 강화됨으로써 현장교육실습생의 현장이해 및 교사 역량이 연차별로 향상되었으며, 현장교육실습 활동에 대한 실습생과 실습학교의 만족도도 증가하였다. 그리고 국제교육실습생 수와 만족도가 상승하였으며, 실습생의 상호교류도 나타나게 되었다. 마지막으로 연구결과를 토대로 시사점 및 의의, 그리고 연구의 제한점을 고려한 제언을 하였다. This study aimed to analyze the case of student-teacher training practicum systemsmodification process a "A" College of Education, and identify the outcomes and implications. "A" college was selected and funded by a government project. The project was titled ``Leading College of Education Support``. From 2011 to 2014, "A" College of Education tried to improve student-teacher practicum systems, and changed the curriculum for student-teacher education including pre-post preparation education. For example, "A" college tried to link observation-educational volunteering-field practicum and constructed practicum support systems including the development of portfolios and the reinforcement of pre-post lecturing for practicums. In addition, "A" college sent its student-teachers to a college in YeonBeon in China, with the perspective of ``Global Practicum``, and the colleges exchanged student-teachers with each other. The number was increased and student teachers` and college`s interests and supports were enhanced. To improve the systems, "A" college connected observation-educational volunteering - field practicum relation, and evaluated degrees of satisfaction and the effectiveness of student-teacher training from the view of practicum schools and participants undertaking practicums. The results revealed a gradual improvement in satisfaction and effectiveness. Finally, the implications of this research were proposed.
