The effect of rituximab dose on infectious complications in ABO-incompatible kidney transplantation

        Lee, Juhan,Lee, Jae Geun,Kim, Sinyoung,Song, Seung Hwan,Kim, Beom Seok,Kim, Hyun Ok,Kim, Myoung Soo,Kim, Soon Il,Kim, Yu Seun,Huh, Kyu Ha Oxford University Press 2016 Nephrology, dialysis, transplantation Vol.31 No.6

<P>Background. Rituximab (RIT) improves the outcomes of ABO-incompatible (ABOi) kidney transplantation (KT), but it has been associated with infectious complications. The aim of this study was to investigate infectious complications according to the dose of RIT in ABOi KT. Methods. We analyzed 213 recipients [118 ABO-compatible (ABOc) KT and 95 ABOi KT] who underwent living donor KT between 2010 and 2014. ABOi KT patients were categorized by RIT dose: standard RIT (375 mg/m(2), n = 76) versus reduced RIT (200 mg, n = 19). All patients received basiliximab and maintained on triple immunosuppression consisting of tacrolimus, prednisone and mycophenolate mofetil. Infectious complications and post-transplant outcomes were analyzed for 1 year following KT. Results. The rates of overall infectious complications among the three groups were comparable (22.9% in ABOc KT, 38.2% in standard RIT and 26.3% in reduced RIT, P = 0.069). In the standard RIT group, hepatitis B virus reactivation occurred in three recipients (3.9%) with hepatitis B surface antigen [-]/anti-hepatitis B core antibody[+]. Three cases (3.9%) of Pneumocystis jirovecii pneumonia occurred in the standard RIT group. Serious infections developed in 13 of the ABOc KT (11.0%), 20 from the standard RIT group (26.3%) and 2 from the reduced RIT group (10.5%, P = 0.015). Standard-dose RIT was found to be an independent risk factor for serious infections [hazard ratio: 2.59 (95% confidence interval: 1.33-5.07), P = 0.005]. There were no significant differences in rejection, renal function, graft survival and patient survival between standard and reduced RIT groups. Conclusions. Standard RIT increased the risk of serious infection when compared with reduced-dose RIT. Reduced-dose RIT might be sufficient for ABOi KT without increasing the risk of serious infection.</P>

        A 50-MHz–1-GHz 2.3-dB NF Noise-Cancelling Balun-LNA Employing a Modified Current-Bleeding Technique and Balanced Loads

Kim, Sinyoung,Kwon, Kuduck IEEE 2019 IEEE Transactions on Circuits and Systems I: Regul Vol.66 No.2

<P>A new noise-cancelling method that employs a modified current-bleeding (CBLD) technique and balanced loads is presented by developing a design for a low-noise and high-linearity balun-low-noise amplifier (LNA) for broadband applications. The basic common-gate (CG)–common-source (CS) balun topology cannot achieve a noise figure (NF) of less than 3 dB. Thus, a practical topology containing a CS transistor of which the transconductance is N times larger than that of the CG transistor and a CS resistor of which the resistance is <TEX>$N$</TEX> times smaller than that of the CG resistor is often used to decrease the NF. However, unsymmetrical load resistors cause a gain and phase imbalance at the differential output. The proposed modified CBLD technique enables the balun-LNA to achieve differential balanced output, low noise, and low-second-order distortion characteristics. The proposed balun-LNA is implemented in 65-nm CMOS technology and covers the frequency range of 50 MHz–1 GHz. It achieves a voltage gain of 30 dB, an S11 of less than −10 dB, an OIP3 of 25.9 dBm, and an OIP2 of 50.6 dBm. The minimum NF is 2.3 dB whereas the average NF is 2.63 dB across the whole band. It operates at a nominal supply voltage of 2.2 V with bias currents of 9 mA. The active die area is 0.0448 mm2.</P>

      • Li and Mg Co-Doped Zinc Oxide Electron Transporting Layer for Highly Efficient Quantum Dot Light-Emitting Diodes

        Kim, Hyo-Min,Cho, Sinyoung,Kim, Jeonggi,Shin, Hyeonjeong,Jang, Jin American Chemical Society 2018 ACS APPLIED MATERIALS & INTERFACES Vol.10 No.28

<P>Zinc-oxide (ZnO) is widely used as an n-type electron transporting layer (ETL) for quantum dot (QD) light-emitting diode (QLED) because various metal doping can be possible and ZnO nanoparticle can be processed at low temperatures. We report here a Li- and Mg-doped ZnO, MLZO, which is used for ETL of highly efficient and long lifetime QLEDs. Co-doping, Mg and Li, in ZnO increases its band gap and electrical resistivity and thus can enhance charge balance in emission layer (EML). It is found also that the O-H concentration at the oxide surface decreases and exciton decay time of QDs on the metal oxide increases by co-doping in ZnO. The inverted green QLEDs with MLZO ETL exhibits the maximum current efficiency (CEmax) of 69.1 cd/A, power efficiency (PEmax) of 73.8 lm/W, and external quantum efficiency (EQEmax) of 18.4%. This is at least two times higher compared with the efficiencies of the QLEDs with Mg-doped ZnO ETL. The optimum Li and Mg concentrations are found to be 10% each. The deep-red, red, light-blue, and deep-blue QLEDs with MLZO ETLs exhibit the CEmax of 6.0, 22.3, 1.9, and 0.5 cd/A, respectively. The MLZO introduced here can be widely used as ETL of highly efficient QLEDs.</P> [FIG OMISSION]</BR>

      • Ligand immobilization on polydiacetylene-coated and surface-enhanced Raman scattering-encoded beads for label-free detection

        Kim, Hyung-Mo,Kang, Yoo-Lee,Chung, Woo-Jae,Kyeong, San,Jeong, Sinyoung,Kang, Homan,Jeong, Cheolhwan,Rho, Won-Yeop,Kim, Dong-Hyuk,Jeong, Dae Hong,Lee, Yoon-Sik,Jun, Bong-Hyun Elsevier 2015 Journal of industrial and engineering chemistry Vol.21 No.-

<P><B>Abstract</B></P> <P>A better understanding of protein–protein interactions can be obtained from multiplex protein detection technologies, and spectrally encoded beads can provide fast and efficient means for this type of detection methods. However, high-throughput detection is challenging due to the requirement of using labeled secondary proteins to detect protein binding events. We have previously reported that polydiacetylene-coated surface-enhanced Raman scattering-encoded beads (PDA–SERS beads) can provide an enhanced encoding capacity owing to their SERS properties as well as their potential for label-free detection from the PDA layer. In this study, we introduced ligands to the PDA–SERS beads by using methods for making free-floating vesicles and planar solid substrates, which enabled the detection of target proteins by PDA fluorescence in a PDA–SERS beads system. By using PDA–SERS beads immobilized with biotin, the fluorescence intensities of biotin-conjugated PDA–SERS beads were increased with an increase in the concentration of streptavidin. And, we could detect 2×10−8 M of streptavidin by measuring the fluorescence intensity without the requirement of an additional labeling step.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

Human Endometrium Derived Induced Pluripotent Stem Cells Are Amenable to Directed Erythroid Differentiation

        Kim Hyun Kyung,Cho SiHyun,Choi Young Sik,Lee Byung Seok,Kim Sinyoung,Kim Hyun Ok,Park Joo Hyun 한국조직공학과 재생의학회 2023 조직공학과 재생의학 Vol.20 No.6

BACKGROUND: A protocol for using human endometrium derived induced pluripotent stem cells (iPSCs) to derive hematopoietic and erythroid lineages will be elaborated, through a two-phase culture system. METHODS: Discarded endometrial tissues were obtained from women receiving hysterectomy in their 4th to 5th decade due to benign uterine conditions. pCE-Sox2, Oct4, Klf4, L-Myc and Lin28 episomal vectors were used to electrotransfect the endometrial stromal cells. The first 8 days involves commitment to hematopoietic stem cells through embryoid body with robust expansion on murine bone marrow stromal cells. The second phase involves feeder free conditions with hydrocortisone, stem cell factor, interleukin-3, and recombinant EPO. After 22 days of feeder free culture, the expression profiles of CD235a?, CD34?, CD43? and CD 71? were analyzed by flow cytometry and Wright-Giemsa staining for differential counting. The oxygen carrying capacity of cultured RBCs was measured using a hemoxanalyser. RESULTS: As a result of inducing these cells via co-culture with murine stromal fibroblasts, all endometrium derived iPSCs were differentiated into erythroblasts with a stable yield of approximately 80% for polychromatic and orthochromatic normoblasts. The protocol for complete induction of erythroid lineage cells starting from human endometrial tissue via iPS cells has been optimized. CONCLUSION: Successful directed erythroid differentiation has occurred from human endometrium-derived iPS cells. A comprehensive process of actually deriving iPS cells using discarded surgical hysterectomy specimens to the erythroid fate has significance in that the scope of using human iPSC cell lines for tissue regeneration could be expanded in the future.
