        The basal concentrations of three major prostaglandins (PG) were determined by the very sensitive radioimmunoassay (RIA). When rat was sacrificd by microwave irradiation, the values of PGD_2, PGE_2 and PGF_2α were 1.35±0.22, 0.74±0.17 and 0.27±0.13 ng/ whole brain respectively, whereas other methods like simple decapitation, perfusion or perfusion with indomethacine showed far higher levels of the corresponding prostaglandins. In order to investigate the basal concentrations of the prostaglandins in any specific part of the brain, rat brain was dissected into 13 parts. PGD_2 was the most significant in pineal gland (38.4±9.6 ng/g wet weight) and posterior pituitary (17.8±4.1 ng/g wet weight). On the other hand, PGE_2 was found to be high in hypothalamus(14.8±0.9 ng/g wet weight), and anterior and posterior pituitary (18.2±0.4 and 17.3±2.7 ng/g wet weight). The concentration of PGF_2α was generally insignificant in contrast to PGD_2 orPGE_2 but still apparent in thalamus and hypothalamus with 4.2±0.8 and2.7±0.6 ng/g wet weight respectively.

      • HTB 176 임파종 세포에 대한 상백피(Mori Cortex)의 세포증식 억제효과

        Lymphoblastoma ATOC HTB 176 암종 세포를 인공 배양하면서 강화도 산 약쑥의 물과 메탄올 추출액을 첨가한 결과 특히 물추출액을 가한 경우에 세포의 증식이 약 50% 정도로 크게 억제되었다. 이 쑥의 세포독성 효과는 항암과 발암의 주요 원인 물질로 알려진 superoxide dismutase(SOD) 활성의 변화와 비례하여 나타났는데, 추출액의 농도 및 배양기간에 비례하여 SOD의 활성이 2배이상 증가하였으며 세포내 소기관중에서는 mitochondria 분획의 SOD활성이 특히 크게 유발되었다. 이와같은 SOD 효소 활성의 증가는 이효소의 전구체인 유독한 활성산소인 O2-의 발생이 특히 mitochondria에서 증가함을 시사한다고 볼 수 있다. 한편 쑥의 첨가에 따른 세포의 생존율이 급격히 감소한 것을 발견하였다. 이러한 결과를 종합하면 TIB 67 암세포에 쑥의 물추출액을 가한경우 세포내 mitochondria에서 유독한 O2-가 발생되며 이 O2-에 대해 자신을 보호하기 위해 SOD활성이 유발되지만 완전하게 보호를 하지 못하고 결국은 생존율이 떨어지고 암세포 자신의 사멸로 이어지는 것을 알 수 있었다. The cell growth inhibition effect of Mori Cortex was elucidated using HTB 176 lymphoblastoma cells. Mon' Cortex extract, specially H₂O extract, displayed a significant effect of cell growth inhibition in relation with extract concentrations and culture days. Such cytotoXic effect was also found to be correlated with the increment of SOD activities in mitochondria fraction. This result suggests that Mori Cortex may stimulate the production of O₂- intmcdulary, which may attack and kill himself, HTB 176 cell. Viability test also showed a steady decrease according to the Mod Cortex concentrations and culture days.

      • TIB 67 Macrophageblastoma 세포에 대한 시호(Bupeurum falcatum) 의 세포독성 효과와 Superoxide dismutase (SOD) 활성과의 관계

        중요한 전통 생약제의 한가지로서 소염, 진통, 진정, 면역 증강, 해열, 이뇨 등의 효과를 나타내는 시호(Bupleurum falcatum)로부터 항암제를 개발하기 위하여 열수 및 메탄을 추출액을 조제하여 각각의 TIB 67 세포에 대한 세포독성과 항산화 효소활성과의 관계를 검색하였다. 시호의 열수추출액과 메탄올추출액 모두 0.1ml를 가하여 4일간 배양을 시켰을 때 50% 이상의 세포독성 효과를 나타내었으며 이 세포 독성효과와 비례하여 세포질 분획이나 미토콘드리아 분획 모두 약 두배정도의 total SOD활성증가가 이루어졌는데 이 활성의 대부분은 Mn-SOD활성으로 밝혀졌다. 이결과들로부터 아마도 시호내의 상당히 극성을 띠는 유기물질이 TIB 67 세포에 대하여 항암효과를 나타내며 이 작용은 초과산화산소 소거제인 superoxide dismutase (SOD), 특히 Mn-SOD의 활성증가와 비례한 것으로 보아 항암현상이 활성산소와 밀접한 관계가 있는 것을 알 수 있었다. 아울러서 원래 기원이 미토콘드리아인 Mn-SOD가 세포질내에서도 거의 동일한 활성치를 보인 것은 항암현상의 발현에는 Mn-SOD의 장소이동도 한가지 중요한 요인이 된다는 것을 시사하고 있다. The present study was taken in an attempt to develop the nontoxic anticancer agent from Bupleurum falcatum which has been known widely for displaying a variety of phamacological effects such as antiinflammation, sedation, analgesic, immunopotentiation, diuretic, antipyretic. Water and methanol extracts prepared from Bupleurum falcatum showed good cytotoxicities with significant value of 52% when 0.2ml extract was added with culture period of 4 days, and over 58% with 0.2ml extract addition and 4 culture days. These cytotoxic activities were accompanied by coincident twofold increments of superoxide dismutase (SOD) activities, specially Mn-SOD of both cytoplasmic and mitochondria fractions. These results suggest that somewhat hydrophilic organic components of Bupleurum falcatum show explicit anticancer activity against TIB 67 cell, one of the leukemic tumors and their reaction mechnism may be performed probably via augmented activity of Mn-superoxide dismutase, which has origin in mitochondria. Hence it can be surmised that the tranlocation of Mn-SOD from mitochondria to cytoplam fraction also would be one of the most important factors involved in the course of anticancer action of natural products.

      • 밤나무 배배양에 미치는 배지조성과 식물호르몬의 영향

        본 연구는 한국종 밤나무(Castanea crenata Sieb and Zucc)의 배배양을 위한 최적조건을 규명코져 시행되었다. 배지의 성분과 식물성장호르몬이 배의 발아 및 잎과 뿌리의 성장에 영향을 미쳤다. 배지중에서 KM과 WPM의 효과가 가장 높았으며 식물성장호르몬으로는 BAP(6-benzyladenine)가 0.2~0.5ppm 농도에서 가장 높은 촉진효과를 나타냈다. In the current study, the optimum condition for the embryo culture of chestnut(Castanea crenata Sieb and Zucc) was investigated. Medimum composition and plant hormone were found to affect the development of embryo shoot and root. KM and WPM were the most effective media. Plant hormone BAP(6-benzyladenine) showed the most stimulating effect in the range of 0.2 to 0.5 ppm.

        Very large PPC effect(σ_ppc/σ_A=10^5) in doping-modulated npnp a-Si: H superlattices could be obtained. B and P atoms, respectively, diffuse and so some parts compensated. Therefore, annealing activation energy of E_a=0.67eV is larger than one of reference n-type sample of E_a=0.26eV and annealing dark conductivity of σ_A=4×10^-[ohm cm]^-1 is lower than that of n-type reference sample of σ_A=2×10 exp (4)[ohm cm]^-1. From this results, very large PPC relatively have obtained. In this paper, we have observed that PPC effect depends on temperature and exposure time. On the basis of the experimental results, we discuss the existing models.

        Antiproliferative Effect of Artemisia argyi Extract against J774A.1 Cells and Subcellular Superoxide Dismutase (SOD) Activity Changes

        The water and methanol extracts of Artemisia argyi showed significant cytotoxicities against J774A.1 cells but not so much against normal leukocytes. The cytotoxicities were found to be dependent on the extract concentration and the incubation time. The concentration of water and methanol extracts inhibiting 50% of cell proliferation (IC?) were estimated to be 44.2 mg/ml and 71.6 mg/ml, respectively. In the presence of Artemisia argyi water extract, total superoxide dismutase (CuZnSOD and MnSOD) activities of media, cytoplasmic and mitochondrial fractions of J774A.1 cells increased in accordance with cytotoxicity. MnSOD was found to be the main component of enhanced total SOD activities, particulary in the mitochondrial fraction. In contrast to SOD, catalase and glutathione peroxidase (GPx) were not found in any instance of the current investigation. In addition, substantial amount of O?? appeared to be generated in the mitochondrial fraction under the influence of Artemisia argyi. All data put together, it is postulated that Artemisia argyi extracts seem to stimulate O? generation in mitochondria of J774A.1 cells with concomitant increases of SODs. Since H?O?, the reaction product of SOD on O?, is known to be readily converted to very toxic OH·in the absence of catalase and/or GPx cooperation, toxicity derived from ROS such as O?, H?O? and OH·may be the main cause of necrosis and/or apoptosis of J774A.1 cells.

      • Farnesol이 ICR 마우스의 대식세포의 증식, O2-, ROS 효소 (SOD, GPx), RNS (iNOS) 효소활성에 미치는 영향

        The current study was performed to find out the immunestimuating anticancer activity of farnesol, one of the monoterpenes. Farnesol showed generally the proliferating effect on the cell number of macrophages from ICR mouse. The most prominent proliferating effect was found to be 135.7% of control in 1.0 μg/ml for 48 hr. Such effect was accompanied by the coincident alterations of RNS (reactive nitrogen species) related enzymes activities such as inducible nitric oxide synthase (iNOS) and O2- ion together with and ROS (reactive oxygen species) related enzymes activities such as superoxide dismutase(SOD) and glutathione peroxidase (GPx). When L1210 cells was cultured in the presence of farnesol (1.0 μg/ml for 48 hr), O2- ion, SOD and GPx activities were increased considerably with values of 178.4 %, 275,4 %, 175.3% respectively. In addition, farnesol exhibited cytotoxic effect (76.4%) on L1210 cell in 1.0 μg/ml for 48 hr.

        기아를 일으킨 흰쥐의 생명연장에 미치는 천연약물의 효과

        The current study was undertaken to select the crude drug showing life prolonging effect on the starved rats(Sprague-Dawley) and elucidate the reaction mechanism there of. As distilled water extracts prepared from twenty kinds of natural drugs were administered orally to female starved rats, Bupleuri Radix demonstrated the most prominent effect with 34.6% increment in the survival time. To investigate underlying mechanism of life prolonging effect of Bupleuri Radix, the concentrations of adrenocorticosteroids(corticosterone, testosterone and aldosterone) and enzyme activities of superoxide dismutase(SOD), glutamate oxaloacetate transaminase(GOT), glutamate pyrurate transaminase(GPT) were assayed in the serums of rats starved for 2, 4 and 6 days respectively. The results manifested much elevated values of corticosterone, aldosterone(2 days) and rather decreasing tendency afterwards, specially in the late periods of starvation(6 days). With respect to such a considerable changes according to starvation periods, Bupleur Radix restored these values much near to normal suggesting that Buplerum Radix may play a life prolonging action during starvation probably through reinforcing the homeostatic properties of corticosteroids and some enzyme activities.

        노화과정에 따른 뇌신경의 핵단백질의 변화

        Rat cerebellum nuclei showed some significant changes of non-histone chromosomal protein (NHCP), a kind of chromosomal protein, as a function of age by electrophoresis profile analysis. Non-histone chromosomal protein was found to be prominent in peak numbers 23, 24 and 37 during developmental stages generally. In adult cerebellum, peak numbers 1, 4, 22 and 26 were distinct as compared with those of neonatal or fetal rats. Further, electrophoresis profile of high mobility group (HMG) protein included in the nonhistone chromosomal protein category revealed the presence of HMG bands 1, 2 and 17 in both neonatal and adult stages but HMG 14 band was distinct in adult cerebellum exceptionally. The HMG protein pattern obtained from nerve cell types demonstrated no much difference between the neuron and glia cell rich found fractions of adult cerebellum.
