The synergistic activation mediator (SAM) system can robustly activate endogenous gene expression by a single-guide RNA. This transcriptional modulation has been shown to enhance gene promoter activity and leads to epigenetic changes. Human interferon-γ is a common natural glycoprotein involved in antiviral effects and inhibition of cancer cell growth. Large quantities of high-purity interferon-γ are important for medical research and clinical therapy. To investigate the possibility of employing the SAM system to enhance endogenous human interferon-γ with normal function in innate immunity, we designed 10 single-guide RNAs that target 200 bp upstream of the transcription start sites of the interferon-γ genome, which could significantly activate the interferon-γ promoter reporter. We confirmed that the system can effectively and highly activate interferon-γ expression in several humanized cell lines. Moreover, we found that the interferon-γ induced by the SAM system could inhibit tumorigenesis. Taken together, our results reveal that the SAM system can modulate epigenetic traits of non-immune cells through activating interferon-γ expression and triggering JAK-STAT signaling pathways. Thus, this strategy could offer a novel approach to inhibit tumorigenesis without using exogenous interferon-γ.
Nrf2 Overexpression Predicts Prognosis and 5-FU Resistance in Gastric Cancer

Objective: NF-E2-related factor 2 (Nrf2) is activated in several human malignancies. However, the role of Nrf2 in gastric cancer (GC) remains incompletely understood. In this study, we therefore analyzed associations of Nrf2 expression status with clinical features and chemotherapeutic resistance in GC. Materials and Methods: A total of 186 samples from GC patients who underwent gastrectomy were used for prognostic assessment. A further 142 samples from GC cases who received first-line combination chemotherapy were applied for investigation of chemoresistance. The Nrf2 expression was evaluated by immunohistochemistry in GC samples, and its relationship with clinicopathological parameters and chemotherapy sensitivity was analyzed. The effect of Nrf2 gene silencing on chemotherapy resistance was also examined by cell viability assay in vivo. Results: Of the 186 patients with GC, 104/186 (55.9%) showed high expression for Nrf2. The overexpression of Nrf2 was an independent predictor of overall survival [OS, hazard ratio (HR) 3.9; P=0.011] and disease-free survival (DFS, HR 4.3; P=0.002). The gene silencing of Nrf2 reduced resistance to cell death induced by 5-FU in GC cell lines. Conclusion: Our data show that Nrf2 is an independent prognostic factor in GC. Furthermore, Nrf2 confers resistance to chemotherapeutic drug 5-FU in GC cells. Taken together, Nrf2 is a potential prognostic marker and predictive for 5-FU resistance in GC.
FABRICATION OPTIMIZATION OF NANOHYDROXYAPATITE ARTIFICIAL BONE SCAFFOLDS

Serious microcracks often occur on the surface of nanohydroxyapatite (n-HAP) artificial bone scaffolds prepared by selectivelaser sintering(SLS) technology. In this study, we found that appropriate preheating before sintering can reduce and attenuate the cracks. The experiments showed that the cracks gradually reduced and then disappeared when the preheating temperature increased from 0 to 600°C while other parameters remain unchanged. The n-HAP particles gradually fused and grew up, while the grain size of sintered n-HAP will be attenuated with the increase of preheating temperature. This provided experimental optimal condition for the preparation of artificial bone scaffolds with nanohydroxyapatite ceramics.

Purpose: To investigate the diet of patients with cervical cancer and precancerosis in the Wufeng area, a high-incidence region in China. Methods: In the case group, 104 patients diagnosed with cervical cancer or cervical intraepithelial neoplasias (CINII/III) were recruited from the Wufeng area. Nine hundred thirty-six healthy women were selected from the same area as the matched controls. A questionnaire, which included questions about general lifestyle conditions, smoking and alcohol status, source of drinking water, green tea intake, and diet in the past year, was presented to all participants. Results: Green tea intake (P=0.022, OR=0.551, 95% CI=0.330-0.919) and vegetable intake (P=0.035, OR=0.896, 95% CI=0.809-0.993) were identified as protective factors against cervical cancer or CINII/III. There was no indication of any associations of other lifestyle factors (smoking status, alcohol status, source of drinking water) or diet (intake of fruit, meat/egg/milk, soybean food, onion/garlic, staple food and pickled food) with cervical cancer. Conclusions: The results suggest that eating more fresh vegetables and drinking more green tea may help to reduce the risk of cervical cancer or CINII/III in people of the Wufeng area.
A Fabrication Process of High Volume Fraction of Jute Fiber/Polylactide Composites for Truck Liner

Aiming at the green manufacturing technology of automotive industry, in the past, some researchers have developed green materials—fully degradable natural fiber reinforced polylactide composites. In the researches, the natural fiber volume fraction (or weight fraction) ranges from 20% to 55%. In order to lower the cost of the composites, an effective way is to increase natural fiber volume fraction and decrease polylactide fraction. In this research, a more uniform fiber blending method is employed, and the natural fiber volume fraction is increased to 70%. The fabrication process includes two steps:felt making and hot pressed molding. Short jute fiber and short PLA fiber are blended on a fiber opening machine in terms of fiber volume fractions, 60% and 70% respectively. The mixtures are carded and needle punched to make felt. Then the felt is hot pressed at 180℃, and the jute/PLA composites are fabricated. Mechanical properties of the composites with different fiber volume fractions are investigated. Real size truck liners are successfully manufactured. The composites have very good formability and processability.
An Effective WSSENet-Based Similarity Retrieval Method of Large Lung CT Image Databases

With the exponential growth of medical image big data represented by high-resolution CT images(CTI), the high-resolution CTI data is of great importance for clinical research and diagnosis. The paper takes lung CTI as an example to study. Retrieving answer CTIs similar to the input one from the large-scale lung CTI database can effectively assist physicians to diagnose. Compared with the conventional content-based image retrieval(CBIR) methods, the CBIR for lung CTIs demands higher retrieval accuracy in both the contour shape and the internal details of the organ. In traditional supervised deep learning networks, the learning of the network relies on the labeling of CTIs which is a very time-consuming task. To address this issue, the paper proposes a Weakly Supervised Similarity Evaluation Network (WSSENet) for efficiently support similarity analysis of lung CTIs. We conducted extensive experiments to verify the effectiveness of the WSSENet based on which the CBIR is performed.
Hot Deformation and Workability of a CrCoNi Medium Entropy Alloy

Hot compression behaviors of CrCoNi medium entropy alloy are were performed on the Gleeble-3800 thermal simulator inthe strain rate range from 0.01 to 10 s−1 and the temperature range from 800 to 1100 °C. According to the true stress-truestrain curves, the true stress decreases with the decreasing of strain rate and the increasing of deformation temperature. Theexperimental parameters were fitted, and a modified model based on Arrhenius hyperbolic sine was established. Amongthem, material constants such as, n, A, and Q are functions of strain. Meanwhile, the processing map of CrCoNi mediumentropy alloy under different strain conditions are established. Importantly, the optimal domain of apparent flow stabilitycan be determined as the deformation temperature from 900 to 1100 °C and the strain rate from 0.01 to 10 s−1. The resultsindicate that the stability domain of the CrCoN medium entropy alloy has a wider range, and the strain has little effect onits processing performance.

Nosema ceranae, a newly emergent parasite invading western honey bees (Apis mellifera L.), is indicated to threaten honey bee health at both individual and colony levels. However, the efficient and environmentallyfriendly treatments are quite limited at present. To find alternative medicine to control Nosema diseases, the effect of 8 types of herbal extracts against N. ceranae infection were screened under laboratory condition. Of which, 1% Andrographis paniculata (A. paniculata) decoction was found to significantly decrease N. ceranae spore numbers on 7 days post infection (dpi) and 13 dpi. Then, our results further revealed that A. paniculata decoction at doses ranging from 1% to 7% displayed significant efficient inhibition of Nosema spore proliferation and improved the infected bees’ survival rates in a dose-dependent manner. A. paniculata decoction was found to protect the gut tissues of infected workers from damage cause by N. ceranae, which might be due to the regu lation of the expression of certain genes in Wnt and JNK pathways, including armadillo, basket, frizzled2 and groucho. Additionally, our study suggested that A. paniculata decoction performed this Nosema spore-reducing potential over its two monomers, andrographolide and dehydrographolide. Taken together, this work enables us to better understand A. paniculata decoction’s potential to inhibit N. ceranae infection, thus providing a new guidance for developing applicable drugs to control Nosema diseases.

We intended to study the mechanism of the inhibitory action of curcumin on human non-small cell lung cancer A549 cell. The cell growth was determined by CCK-8 assay, and the results indicated that curcumin inhibited the cell proliferation in a concentration dependent manner. And to further confirm the relative anti-cancer mechanism of curcumin, RT-PCR was carried out to analysis the expression of relative apoptotic proteins Bax, Bcl-2. We found that curcumin could up-regulate the expression of Bax but down-regulate the expression of Bcl-2 in A549 cells. In addition, curcumin affect the mitochondrial apoptosis pathway. These results suggested that curcumin inhibited cancer cell growth through the regulation of Bcl-2/Bax and affect the mitochondrial apoptosis pathway.

Microcrystalline cellulose was reacted with catalytic amounts of 2, 2, 6, 6-tetramethyl-1-piperidine oxoammonium salt (TEMPO), sodium hypochlorite and sodium bromide in $Na_2CO_3/NaHCO_3$ buffer solution at different temperatures ($30^{\circ}C$, $40^{\circ}C$, $50^{\circ}C$). The oxidation procedures included first oxidation and second oxidation. The yield of cellouronic acid produced in the second oxidation was higher than the yield of cellouronic acid produced in the first oxidation at the same oxidation temperature. Moreover, an interesting "high-hydrophilic" phenomenon appeared at higher temperature during microcrystalline cellulose second oxidation ($30^{\circ}C$, $40^{\circ}C$, $50^{\circ}C$). Properties associated with the chemical characteristics are discussed by XRD, FTIR, $^{13}C$-NMR and Laser Particle Analyzer in view of its interesting high-hydrophilic effects.
