        Non-atypical endometrial hyperplasia: risk factors for occult endometrial atypia and malignancy in patients managed with hysterectomy

        ObjectiveTo determine the risk factors for occult endometrial atypia and malignancy in patients diagnosed with non-atypicalendometrial hyperplasia (NEH) on endometrial biopsy. MethodsAll new cases of NEH diagnosed between April 2015 and March 2016 at KK Women’s and Children’s Hospital, whounderwent hysterectomy as first-line treatment, were included in the study. Patients with a history of endometrialhyperplasia or malignancy were excluded from the study. Patient demographics (e.g., age, parity, body mass index[BMI]), medical history, and clinical presentation were obtained for analysis. ResultsIn total, 262 patients were diagnosed with NEH, of which 18.3% (n=48) underwent hysterectomy as first-linemanagement. The average time to surgery was 77.0±35.7 days. All cases were diagnosed by dilation and curettage,and hysteroscopy. The mean age was 51 years, and the mean BMI was 26.9±5.8 kg/m2. Histology from thehysterectomy specimen showed 9 (18.8%) patients with atypical hyperplasia and 2 (4.2%) with grade 1, stage 1Aendometrioid adenocarcinoma. Patients with higher grade final pathology had significantly lower median parity(1 vs. 2, P=0.039), higher mean BMI (30.1±6.5 vs. 25.9±5.3 kg/m2, P=0.033), and BMI ≥30 kg/m2 (54.5% vs. 13.5%,P=0.008, odds ratio 7.68), compared to patients whose final histology showed NEH or no residual hyperplasia. ConclusionOccult endometrial atypia and malignancy were found in 18.8% and 4.2% of patients with an initial diagnosis of NEH,respectively. High BMI and low parity were identified as significant risk factors for high-grade endometrial lesions inpatients with NEH.

        A Study on Residents' Perception towards the Importance of Forest Therapy in the Central Region of China: Based on the Cases in Korea

        Background and objective: Recently, forest therapy has received strong support from the Chinese government and gradually embarked on the road of development. However, compared with South Korea, forest therapy started late in China. And it is still in the exploration stage of theoretical research and industrial practice. To study the development of forest therapy in China, we review the legal system and development of forest therapy in Korea. This study uses online surveys to help us understand the level of awareness of the importance of forest therapy in central China. And it also provides a reference for the further development of forest therapy in China. Methods: The methods used for this analysis were Chi-square test, one-way Analysis of Variance (ANOVA), principal component analysis (PCA), and one-way Multivariate Analysis of Variance (MANOVA) . Results: Our findings showed that the perception about forest therapy was significantly different among people with different educational level (χ² = 25.526, p < .001). The cognition of forest therapy is significantly different from the cognition of the importance of natural healing projects (p < .001) and pro-environmental projects (p < .001). Moreover, there was a significant difference in the perception towards the importance of 18 forest therapy facilities and 9 industrial development influencing factors with or without forest therapy awareness. Conclusion: The conclusion is that our participants have low awareness of forest therapy, but they have great expectations of forest therapy. In addition, the forest environment and emergency medical facilities reflect the recognition of the importance, including treatment programs, treatment facilities and factors affecting industrial development. Therefore, considering the successful experience of forest therapy in Korea, this study proposes practical measures to strengthen publicity, facility construction and treatment program development.

      • <i>Arabidopsis</i> DET1 degrades HFR1 but stabilizes PIF1 to precisely regulate seed germination

        <P><B>Significance</B></P><P>How organisms respond to environment changes is a fundamental and intriguing question in biology. Light is the energy resource and a crucial environmental cue for plant major developmental switches, such as seed germination. Studying the underlying mechanism is important for us to understand the basic principles of plant development and improve crop productions. Here we identify DET1 as a novel central repressor of seed germination. We further reveal that seeds use a multilevel regulatory circuit of triple feed-forward loops to sensitively and precisely mediate light-regulated germination. This study provides a comprehensive framework of how light regulates seed germination.</P><P>Seed is an essential propagation organ and a critical strategy adopted by terrestrial flowering plants to colonize the land. The ability of seeds to accurately respond to light is vital for plant survival. However, the underlying mechanism is largely unknown. In this study, we reveal a circuit of triple feed-forward loops adopted by <I>Arabidopsis</I> seeds to exclusively repress germination in dark conditions and precisely initiate germination under diverse light conditions. We identify that de-etiolated 1 (DET1), an evolutionarily conserved protein, is a central repressor of light-induced seed germination. Genetic analysis demonstrates that DET1 functions upstream of long hypocotyl in far-red 1 (HFR1) and phytochrome interacting factor 1 (PIF1), the key positive and negative transcription regulators in seed germination. We further find that DET1 and constitutive photomorphogenic 10 (COP10) target HFR1 for protein degradation by assembling a COP10–DET1–damaged DNA binding protein 1–cullin4 E3 ligase complex. Moreover, DET1 and COP10 directly interact with and promote the protein stability of PIF1. Computational modeling reveals that phytochrome B (phyB)–DET1–HFR1–PIF1 and phyB–DET1–Protease–PIF1 are new signaling pathways, independent of the previously identified phyB-PIF1 pathway, respectively mediating the rapid and time-lapse responses to light irradiation. The model-simulated results are highly consistent with their experimental validations, suggesting that our mathematical model captures the essence of <I>Arabidopsis</I> seed germination networks. Taken together, this study provides a comprehensive molecular framework for light-regulated seed germination, improving our understanding of how plants respond to changeable environments.</P>

      • Thermo-mechanical properties and shape memory effects of ethylene acrylic acid copolymer/cellulose nanofiber nanocomposite via water-assisted mixing

        Ethylene acrylic acid copolymer(EAA)/cellulose nanofiber(CNF) nanocomposites were successfully prepared by green and simple waterassisted mixing followed by film casting. EAA was dispersed in water neutralized by ammonia, and the stable aqueous dispersion of EAA mixed with CNF water suspension. The morphology, thermal and mechanical properties of the nanocomposites were studied using TEM, DSC, DMA, and tensile testing. The experimental results showed that CNF were homogenously dispersed into the EAA matrix and provided significant reinforcement of the EAA. The presence of a rubbery plateau above the melting temperature of EAA/CNF nanocomposites obtained from DMA indicated that the nanocomposites formed a physically cross-linked structure. The EAA/CNF nanocomposites also exhibited an excellent thermally triggered shape memory effect.

        A Review on Microbial Decaffeination

        Exclusive exposure of caffeine might bring some adverse effects to human health, and cause some risks to environment. Microbial decaffeination might be a new alternative approach because of its high specificity and efficiency. Many studies revealed that several microorganisms can degrade caffeine, including genera Alcaligenes,Rhodococcus, Klebsiella, Acinetobacter, Enterobacter, Stenotrophomonas and Pseudomonas. Biodegradation of caffeine can be significantly influenced by temperature, pH, aeration rate and initial caffeine concentration of the incubation. There are two main pathways of caffeine biodegradation: C-8 oxidation and N-demethylation, while N-demethylation is the general catabolism pathway in bacteria. Application of microbial decaffeination has also been discussed in this review.

        Construction of an Oscillator Gene Circuit by Negative and Positive Feedbacks

        Synthetic oscillators are gene circuits in which the protein expression will change over time. The delay of transcription, translation, and protein folding is used to form this kind of behavior. Here, we tried to design a synthetic oscillator by a negative feedback combined witha positive feedback. With the mutant promoter PLacC repressed by LacIq and PLux activated by AHL-bound LuxR, two gene circuits, Os-LAA and Os-ASV, were constructed and introduced into LacI-deleted E. coli DH5α cells. When glucose was used as the carbon source, a low level of fluorescence was detected in the culture, and the bacteria with Os-ASV showed no oscillation, whereas a small portion of those carrying Os-LAA demonstrated oscillation behavior with a period of about 68.3 ± 20 min. When glycerol was used as the carbon source, bacteria with Os-ASV demonstrated high fluorescence value and oscillation behavior with the period of about 121 ± 21 min.

        Set Stability of Controlled Chua’s Circuit under a Non-smooth Controller with the Absolute Value

        Chua’s circuit is a typical system with the piecewise-smooth vector field via the absolute value. A new piecewise-linear feedback control scheme with the absolute value is introduced to realize stabilization of sets of the controlled Chua’s circuit. The equilibrium sets and the global asymptotical stability with respect to several invariant sets, which may consist of both admissible and boundary equilibrium points or equilibrium manifolds, are investigated for the controlled Chua’s circuit system. The non-smooth controller has a simple structure and available control properties, and it is easy to be applied in engineering practice. It is shown by numerical simulations that this controller can effectively realize chaos control and stabilization of equilibrium states.

        Generalized strictly diagonally dominant matrices have a wide range of applications in matrix theory and practical applications, so it is of great theoretical and practical value to study their numerical determination methods. In this paper, we study the numerical determination of generalized strictly diagonally dominant matrices by using the properties of generalized strictly diagonally dominant matrices. We obtain a new criterion for subdivision iteration determination of the generalized strictly diagonally dominant matrices by subdividing the set of non-prevailing row indices and constructing new iteration factors for the set of predominant row indices, new elements of the positive diagonal factors are derived. Advantages are illustrated by numerical examples.

        <P>This study identifies the LONG HYPOCOTYL IN FAR-RED1 (HFR1)– PHYTOCHROME-INTERACTING FACTOR 1 (PIF1) pair as the central transcriptional regulatory module for rapid light-initiated transcriptome changes in imbibed seeds. HFR1 interacts with PIF1 to prevent PIF1 from binding to DNA, promoting light-initiated germination. The functionally antagonistic HFR1–PIF1 pair constructs a fail-safe mechanism for fine-tuning seed germination under low-level illumination.</P>
