      • Trichostatin A enhances proliferation and migration of vascular smooth muscle cells by downregulating thioredoxin 1

        <P><B>Aims</B></P><P>A reduction in the level of thioredoxin 1 (Trx1) has been proposed as a possible mechanism for the tumor-specific growth arrest caused by inhibition of histone deacetylases (HDACs). In this study, we investigated the effect of trichostatin A (TSA), a potent HDAC inhibitor, on the proliferation and migration of vascular smooth muscle cells (VSMCs), and we examined the role of reduced Trx1 levels in this effect.</P><P><B>Methods and results</B></P><P>TSA treatment time-dependently decreased Trx1 expression in rat VSMCs at both the mRNA and protein levels. It also enhanced platelet-derived growth factor (PDGF)-induced proliferation and migration of the VSMCs. By potentiating Akt phosphorylation, the siRNA-induced downregulation of Trx1 also enhanced VSMC proliferation and migration in response to PDGF or serum treatment. Consistent with these results, TSA administration increased neointimal thickening in a murine model of post-angioplastic restenosis.</P><P><B>Conclusion</B></P><P>These data demonstrate that TSA enhances vascular proliferative activity by downregulating Trx1, thus activating an Akt-dependent pathway. Our results indicate that, in addition to its apoptotic effects in tumour cells, the downregulation of Trx1 has a proliferative role in primary VSMCs.</P>

        경혈 교육을 위한 3D 및 증강현실 기술을 활용한 한의학 통합교육 테이블 개발

        Objectives : Acupoints education is important in that it can determine the clinical competency of Korean Medicine Doctors (KMDs). Accordingly, we aimed to develop a practical simulator for acupoints education, acupoints training, acupoints practice, and acupoints evaluation. Methods : Korean Medicine (KM) SMART Table can be divided into hardware, server and components, and is organically linked. We develop KM SMART Table that combines the hardware of a human-sized table with a UHD display capable of multi-touch in two cases and software that can teach acupoints. We make Augmented Reality (AR) contents linked with KM SMART Table contents and develop applications that can use contents using mobile devices. By developing an AR image tracking module to react with KM SMART Table, it enables acupoint learning according to the mobile device platform and human anatomy. Results : The current system is a prototype where some 3D technology has been implemented, but the AR function will be produced later. New learning using 3D and AR will be required during acupoints education and acupoints practice. It will be used a lot in OSCE (Objective Structured Clinical Examination) practices for strengthening the competency of KMDs, and it will be of great help not only in KM education as a unique simulator of KM, but also in the practice of acupuncture and chuna for musculoskeletal diseases. Conclusions : The KM SMART Table is a technology that combines 3D and AR to learn acupoints, and to conduct acupoints OSCE practice, and we suggest that it can be usefully used for educational evaluation.

        The RFID the technique is non-contact radio recognition technique and overcomes the bar codes of the limit of slow recognition speed, recognition ratio and storage capability. It will be like that and it is paid attention with the fact that it will bring a renovation change in enterprise goods activity or information transmission system. It is a focus technique that simultaneously takes charge of a ubiquitous network sensor function. The u-CRM uses a RFID technique. In ubiquitous environment, the u-CRM is to collect, to store a customer data and to manage the customer. And at real-time it will collect and will be able to analyze information of the customer. Therefore it will be able to provide an immediate service. This study, it compares the u-CRM that uses the RFID and existing CRM and it proposes the efficient u-CRM model.

      • Continuous blood cell separation by hydrophoretic filtration

        <P>We propose a new hydrophoretic method for continuous blood cell separation using a microfluidic device composed of slanted obstacles and filtration obstacles. The slanted obstacles have a larger height and gap than the particles in order to focus them to a sidewall by hydrophoresis. In the successive structure, the height and gap of the filtration obstacles with a filtration pore are set between the diameters of small and large particles, which defines the critical separation diameter. Accordingly, the particles smaller than the criterion freely pass through the gap and keep their focused position. In contrast, the particles larger than the criterion collide against the filtration obstacle and move into the filtration pore. The microfluidic device was characterized with polystyrene beads with a minimum diameter difference of 7.3%. We completely separated polystyrene microbeads of 9 and 12 µm diameter with a separation resolution of ∼6.2. This resolution is increased by 6.4-fold compared with our previous separation method based on hydrophoresis (S. Choi and J.-K. Park, <I>Lab Chip</I>, 2007, 7, 890, ref. 1). In the isolation of white blood cells (WBCs) from red blood cells (RBCs), the microfluidic device isolated WBCs with 210-fold enrichment within a short filtration time of ∼0.3 s. These results show that the device can be useful for the binary separation of a wide range of biological particles by size. The hydrophoretic filtration as a sample preparation unit offers potential for a power-free cell sorter to be integrated into disposable lab-on-a-chip devices.</P> <P>Graphic Abstract</P><P>This paper describes a novel hydrophoretic method for continuous blood cell separation using filtration obstacles, which play not only the role of filtering large particles blocked by the obstacles but also the role of focusing small particles able to pass through the gap between the obstacle and the top or bottom of the channel. <IMG SRC='http://pubs.rsc.org/services/images/RSCpubs.ePlatform.Service.FreeContent.ImageService.svc/ImageService/image/GA?id=b705203k'> </P>

      • Involvement of JNK-Dependent Mitochondrial Respiratory Burst in Oxidative Stress induced Neurinal Cell Death

        Reactive oxygen species(ROS), especially hydrogen peroxide(H₂O₂) and superoxide anion, exert detrimental effects on cell survival and are known to be involved in a variety of pathophysiologic conditions. In this study, we demonstrate that exogenous H₂O₂ exerts cytotoxic effects via a late-phase surge of endogenous ROS(mainly superoxide anion) generated by activated mitochondria. Such delayed generation of ROS was suppressed by diphenylene iodonium or rotenone, potent inhibitors of mitochondrial respiratory chain complex Ⅰ. Secondary production of ROS was preceded by a transient hyperpolarization of the mitochondrial transmembrane potential(ΔΨm), which was accompanied by increased production of ATP. Delayed production of superoxide anion was also observed in the reperfused ischemic brains, and blockage of mitochondrial hyperactivation by rotenone reduced ROS production and the infarction volumes. Sustained activation of c-Jun N-terminal kinase(JNK) has been known to mediate oxidative stress-induced neuronal cell death in ischemic-reperfusion injury. We further demonstrate here that the JNK pathway is responsible for the mitochondrial hyperactivation, which results in delayed ROS generation and subsequent neuronal cell death in vitro and in vivo.

      • Sheathless Focusing of Microbeads and Blood Cells Based on Hydrophoresis

        <P>This paper presents a microfluidic device for sheathless focusing of microbeads and blood cells based on a hydrophoretic platform comprising a V-shaped obstacle array (VOA). The VOA generates lateral pressure gradients that induce helical recirculations. Following the focusing flow particles passing through the VOA are focused in the center of the channel. In the device, the focusing pattern can be modulated by varying the gap height of the VOA. To achieve complete focusing within 4.4% coefficient of variation, the relative size differences between the gap and the particle were 3 and 4 µm for 10 and 15 µm beads, respectively. Red blood cells were used to study the hydrophoretic focusing pattern of biconcave, disk-shaped particles.</P> <B>Graphic Abstract</B> <P>A sheathless focusing method is developed based on a hydrophoretic platform comprising a V-shaped obstacle array (VOA). Hydrophoretic focusing of microparticles is performed along transverse pressure gradients and recirculation flows generated by the VOA.         교과서에 실린 예시 단어의 사용 빈도와 전형성 분석

        이 연구에서는 2011 교육과정에 따른 중학교 교과서 14종을 대상으로 하여서 단어 형성법 단원 파생어 내용을 설명하면서 실린 예시 단어의 빈도와 전형성을 분석하였다. 분석의 범위를 교과서 개념 이해 영역에서 다루어진 예시 단어로 국한하였다. 파생어 사용 빈도 분석 결과, 접두 파생어 중 사용 빈도가 높은 5개는 차례로 ‘치솟다(323), 맨발(221), 짓밟다(201), 한여름(195), 참기름(170)’이었고 사용 빈도가 0-2인 파생형이 ‘덧버선, 풋김치, 개철쭉’ 등 24개가 있었다.접미 파생어 중 사용 빈도가 높은 5개는 차례로 ‘손님(2500), 놀이(1613), 길이(1156), 높이(920), 날개(876)’이었고 사용 빈도가 0-2인 파생형이 ‘구두장이(2), 먹보(1), 잠보(1), 침질(1), 꾀보(0)’의 5개가 있었다. 전형성 분석 결과, 접사에서는 다른 범주와의 경계가 모호한 접두사로 ‘늦-’, ‘참-’이 있었고 다른 범주와의 경계가 모호한 접미사로 ‘-님’, ‘-기’가 있었다. 파생어에서는 공시적으로 ‘어근+접사’ 구성이 아닌 파생어 ‘풋내기’가 교과서에서 사용되었음이 밝혀졌다. 빈도와 전형성 분석 결과 빈도에 따라 예시 단어가 재검토되어야 하고 이때 접사 빈도와 파생어 빈도뿐만 아니라 어근 빈도 또한 복합적으로 고려되어야 함이 논의되었고 빈도와 전형성에 있어 그 정도가 불일치하는 단어의 경우 개념 이해 단계가 아닌 탐구 단계에서 다루어져야 한다고 하였다. The purpose of this study is to analysis frequency and typicality of example words used in textbooks. We use 14 textbooks corresponding 2011 educational curriculum for analysis and the unit we examined was about word formations, especially derived words. the scope of this study was limited to areas of understanding of the concepts. The result of analysis of derived words is summarized as follows. Top5 derived words by prefix on frequency were ‘치솟다(323), 맨발(221), 짓 밟다(201), 한여름(195), 참기름(170)’, and ‘0-2’ frequency were 24 words included ‘덧버선, 풋김치, 개철쭉’ etc. Top 5 derived words by suffix on frequency ‘손님(2500), 놀이(1613), 길이(1156), 높이(920), 날개(876)’, and ‘0-2’ frequency were ‘구두장이(2), 먹보(1), 잠보(1), 침질(1), 꾀보(0)’. The resultof analysis of typicality is summarized as follows. The prefixes which blur the boundaries were ‘늦-’, ‘참-’, and the suffixes were ‘-님’, ‘-기’. Also, There is a ‘풋내기’ which was not composed of ‘root+affix’ synchronically. We discussed about result of analysis of frequency and typicality. 1) Example words used textbook must be reexamined on results of analysis of frequency. 2) The root frequency is also concerned. 3) Words which have the discrepancy between frequency and typicality are included in areas of inquiry learning not understanding of concepts.
