        Punching Behaviour of Reinforced Concrete Footings at Testing and According to Eurocode 2 and fib Model Code 2010

        Punching shear resistance of column footings and foundation slabs varies significantly in different standards. The reason for this is because standards define differently the position of the critical perimeter in which the punching shear resistance should be determined, and quantify the influences of the main parameters like effective depth, shear slenderness, compressive strength of concrete, longitudinal reinforcement ratio and tension yield stress of reinforcement in different ways. In order to quantify the level of safety in Eurocode 2 and in fib MC 2010, their design results are compared with the test results of the series of footings tested in completely realistic boundary conditions in terms of the subgrade soil. Besides the performed tests results, the analysis of the other investigations of the footing punching rested on the real soil is also included. Thus was obtained the answer to the question how individual characteristics of the footings and of the soil affect the punching bearing resistance and how accurately Eurocode 2 and fib MC 2010 predict the bearing capacity of the tested column footings. At the end, based on the test results and on the tests of others, and on and performed numerical analyses, a possible modification of Eurocode 2 in the field of reinforced concrete footing was proposed.

        A comparison of the antinociceptive and temperature responses to morphine and fentanyl derivatives in rats

        In addition to producing antinociception, opioidsexert profound effects on body temperature. This studyaimed at comparing antinociceptive and hyperthermicresponses between two groups of l-opioid receptor agonists:fentanyl (4-anilinopiperidine-type) and morphine(phenanthrene-type) derivatives in rats. Analgesic activitywas assessed by tail immersion test and the body temperatureby insertion of a thermometer probe into the colon. Fentanyl (F), (±)-cis-3-methyl fentanyl (CM), (±)-cis-3-carbomethoxy fentanyl (C), (±)trans-3-carbomethoxyfentanyl (T) and (±)-cis-3 butyl fentanyl (B) produceddose-dependent increase in antinociception and hyperthermia. The relative order of analgesic potency was:CM(11.27)[F(1)[C(0.35)CT(0.11)CB(0.056). Similar tothis, the relative order of hyperthermic potency was:CM(8.43)[F(1)[C(0.46)CT(0.11)CB(0.076). Morphine(M), oxycodone (O), thebacon (T) and 6,14-ethenomorphinan-7-methanol, 4,5-epoxy-6-fluoro-3-hydroxy-a,a,17-trimethyl-, (5a,7a) (E) also produced dose-dependentincrease in antinociception and hyperthermia. Amongmorphine derivatives the relative order of analgesicpotency was: E(56)[O(5)CT(2.6)[M(1), and similar tothis, the relative order of hyperthermic potency was:E(37)[O(3)CT(2.3)[M(1). Morphine (phenanthrene-type)and fentanyl (4-anilinopiperidine-type) derivatives producedhyperthermia in rats at doses about 2 times lower,and 6–11 times higher, than their median antinociceptivedoses, respectively. This study is first to identify differencebetween these two classes of opioid drugs in their potenciesin producing hyperthermia. Further studies are needed toclarify the significance of these findings.

        Parametric Model Variability of the Proximal Femoral Sculptural Shape

        In this paper, a method of parameterisation of a proximal femur model is presented. The created model retains all the anatomical and morphological features of a realistic model. The parametric model of the femur, the so-called master model, is defined by establishing a relation between appropriate femoral regions which extend along the parametrically described axis of the femur. An initial 3D model of the femur is segmented from the CT image and further parameterized to a master model with the potential for customization, i.e. for adaptation to patient-specific values using X-ray images, still maintaining precise anatomical consistency. The first step towards the femur customization is the contour extraction of different types of tissues represented in X-ray images. As medical images can be blurry, image processing was carried out and a Canny edge detector operator was applied. The numerical values of the parameters were determined by manual measuring of defined regions on an X-ray image. The variability testing was performed on 12 femurs. The proposed model can greatly contribute to preoperative planning, implant selection, as well as to the overall shortening of intervention time.

        Permanent disposal of Cs ions in the form of dense pollucite ceramics having low thermal expansion coefficient

        A promising method for removal of Cs ions from water and their incorporation into stable crystal structure ready for safe and permanent disposal was described. Cs-exchanged X zeolite was hot-pressed at temperature ranging from 800 to 950 ℃ to fabricate dense pollucite ceramics. It was found that the application of external pressure reduced the pollucite formation temperature. The effect of sintering temperature on density, phase composition and mechanical properties was investigated. The highest density of 92.5 %TD and the highest compressive strength of 79 MPa were measured in pollucite hot-pressed at 950 ℃ for 3 h. Heterogeneity of samples obtained at 950 ℃ was determined using scanning electron microscopy. The pollucite hot-pressed at 950 ℃ had low linear thermal expansion coefficient of ~4.67 × 10<sup>-6</sup> K<sup>-1</sup> in the temperature range from 100 to 1000 ℃.

        TNF, IL12B, and IFNG Gene Polymorphisms in Serbian Patients with Psoriasis

        Background: Psoriasis is a common chronic inflammatory skin disease with a strong genetic basis. Cytokines such as tumor necrosis factor alpha (TNF-α), interleukins (ILs) such are IL-12 and IL-23, and interferon gamma (IFN-γ) are released from various inflammatory and resident cells, and have been implicated in the initiation/maintenance of inflammation. Certain alleles of the aforementioned cytokines may be associated with disease susceptibility/severity. Objective: To investigate the association of three common functional gene polymorphisms, namely TNF -308 G/A (rs1800629), IL12B (encoding the p40 subunit of IL-12/23) +1188 A/C (rs3212227), and IFNG +874 T/A (rs2430561) with psoriasis development and severity in Serbian patients. Methods: We genotyped 130 patients with psoriasis (26 of whom also had psoriatic arthritis) and 259 controls; rs1800629 and rs3212227, and rs2430561, by real-time PCR assay. Results: The TNF GG genotype was detected at a higher frequency in patients with psoriasis compared to control subjects (OR, 1.420; 95% CI, 0.870∼2.403) without statistical significance (p= 0.191). Lack of the TNF G allele was associated with lower psoriasis severity (p=0.007). The IL12B AC genotype was underrepresented in the patients with psoriatic arthritis compared to healthy subjects (OR, 0.308; 95% CI, 0.090∼ 1.057; p=0.049). The distribution of the rs2430561 allele and genotype frequencies was similar between patients with psoriasis and controls. Conclusion: Our study demonstrates an effect of the rs1800629 on psoriasis severity, and a marginal impact of the rs3212227 on susceptibility to psoriatic arthritis. Collectively, our results obtained in a Serbian cohort expand current knowledge regarding individual predisposition to psoriatic disease.(Ann Dermatol 27(2) 128∼132, 2015)

        Population Pharmacokinetic Analysis of the Oral Absorption Process of Tacrolimus in Healthy Volunteers

        연구배경: 타크로리무스는 이식환자들의 동종이식 거부반응을 예방하기 위해 사용된다. 타크로리무스는 치료 허용 범위가 좁고 개인 간, 개인 내 약동학적 차이가 높으므로 혈중 농도의 모니터링을 이용한 약물 투여 용량 조절이 필수적이다. 현재 타크로리무스의 흡수 기전에 관한 연구가 미흡하여 이번 연구를 통해 건강 피험자 중 타크로리무스의 최적의 흡수 모델을 정의하는 것을 목표로 하였다. 방 법: 31명의 건강한 성인들에게 각각 4 mg의 타크로리무스를 경구투여하였다. 채혈시점은 약물투여 전및 투여 후 0.5, 1, 1.5, 2, 3, 6, 8, 10, 12, 24, 48, 60 ,72 시간이었으며 혈장 내 타크로리무스의 농도 분석은HPLC-MS-MS를 이용하였다. 약동학적 분석을 위해 NONMEM을 사용하였고, 영차 흡수, 일차 흡수, 트랜짓 분획 흡수 모델에 2분획적 모델을 적용하여 비교하였다. 결 과: 트랜짓 분획 흡수 모델이 타크로리무스의 약동학적 흡수 과정을 가장 잘 설명하는 것으로 나타났다. 여기서 적용된 3 가지의 흡수 모델 중 트랜짓 분획 흡수 모델에서 OFV와 RMSE가 가장 감소하였고, 각 개인 피험자들의 흡수기를 나타낸 그래프와 산점도 그림에서도 트랜짓 분획 흡수 모델이 일차 흡수 모델보다 더 우수했다. 결 론: 우리는 최근에 고안되어진 트랜짓 분획 모델이 타크로리무스의 불규칙한 약동학적 흡수 과정을 나타내는 것에 유용한 모델임을 알 수 있었다. 타크로리무스의 흡수 과정이 제형에 관계 없이 불규칙적인 것으로 알려져 있으므로, 트랜짓 분획 모델이 흡수 지연 시간을 적용한 약동학적 흡수 과정의 특징을 생리학적으로 보다 잘 나타낼 것으로 기대한다.

        Elucidation of the profound antagonism of contractile action of phenylephrine in rat aorta effected by an atypical sympathomimetic decongestant

        Vasoconstrictive properties of sympathomimetic drugs are the basis of their widespread use as decongestants and possible source of adverse responses. Insufficiently substantiated practice of combining decongestants in some marketed preparations, such are those containing phenylephrine and lerimazoline, may affect the overall contractile activity, and thus their therapeutic utility. This study aimed to examine the interaction between lerimazoline and phenylephrine in isolated rat aortic rings, and also to assess the substrate of the obtained lerimazoline-induced attenuation of phenylephrine contraction. Namely, while lower concentrations of lerimazoline ($10^{-6}M$ and especially $10^{-7}M$) expectedly tended to potentiate the phenylephrine-induced contractions, lerimazoline in higher concentrations ($10^{-4}M$ and above) unexpectedly and profoundly depleted the phenylephrine concentration-response curve. Suppression of NO with NO synthase (NOS) inhibitor $N^w$-nitro-L-arginine methyl ester (L-NAME; $10^{-4}M$) or NO scavanger $OHB_{12}$ ($10^{-3}M$), as well as non-specific inhibition of $K^+$-channels with tetraethylammonium (TEA; $10^{-3}M$), have reversed lerimazoline-induced relaxation of phenylephrine contractions, while cyclooxygenase inhibitor indomethacin ($10^{-5}M$) did not affect the interaction between two vasoconstrictors. At the receptor level, non-selective 5-HT receptor antagonist methiothepin reversed the attenuating effect of lerimazoline on phenylephrine contraction when applied at $3{\times}10^{-7}$ and $10^{-6}M$, but not at the highest concentration ($10^{-4}M$). Neither the 5-$HT_{1D}$-receptor selective antagonist BRL 15572 ($10^{-6}M$) nor 5-$HT_7$ receptor selective antagonist SB 269970 ($10^{-6}M$) affected the lerimazoline-induced attenuation of phenylephrine activity. The mechanism of lerimazoline-induced suppression of phenylephrine contractions may involve potentiation of activity of NO and $K^+$-channels and activation of some methiothepin-sensitive receptors, possibly of the 5-$HT_{2B}$ subtype.

        An Investigation of Formulation Factors Affecting Feasibility of Alginate-Chitosan Microparticles for Oral Delivery of Naproxen

        In the present work we investigated the feasibility of chitosan treated Ca-alginate microparticles for delivery of naproxen in lower parts of GIT and evaluated influence of formulation factors on their physicochemical characteristics and drug release profiles. Investigated factors were drug/polymer ratio, chitosan molecular weight, chitosan concentration in hardening medium, and hardening time. Sixteen microparticle formulations were prepared utilizing 24 full factorial design (each factor was varied at two levels). Microparticles size varied between 262.3 ± 14.9 and 358.4 ± 21.7 μm with slightly deformed spherical shape. Low naproxen solubility and rapid reaction of ionotropic gelation resulted in high encapsulation efficiency (> 75.19%). Under conditions mimicking those in the stomach, after two hours, less than 6.18% of naproxen was released. Significant influence of all investigated factors on drug release rate was observed in simulated small intestinal fluid. Furthermore, experimental design analysis revealed that chitosan molecular weight and its concentration had the most pronounced effect on naproxen release. Release data kinetics indicated predominant influence of a pH-dependent relaxation mechanism on drug release from microparticles.
