        탄소강 배관 재료의 DSA 거동에 미치는 노치 영향 평가

        본 연구에서는 탄소강 배관 재료의 동적변형시효 거동에 미치는 노치에 의한 응력과 변형 집중효과를 파악하기 위해서, 표준 시편과 노치 시편을 이용하여 다양한 온도와 시험 속도에서 인장시험을 수행하였다. 또한, 시편의 형상에 따른 응력과 변형률 상태의 차이를 파악하기 위해서, 각 시편에 대한 유한요소 시뮬레이션을 수행하였다. 노치 시편을 이용한 인장시험 결과에서도 표준 시편에서 관찰된 것과 같은 동적변형시효의 증거인 serration과 인장강도의 증가 현상이 관찰되었다. 동일한 시험 속도에서 인장시험이 수행된 경우에 표준 시편에 비해 노치 시편에서 동적변형시효 현상이 고온에서 나타났다. 유한요소 해석 결과에 의하면 표준 시편에 비해 노치 시편에서 동적변형시효가 고온에서 나타나는 것은 노치부에서 응력과 변형의 집중으로 인한 높은 변형률 속도에 기인하는 것으로 확인되었다. In this study, tensile tests were performed using standard and notched-bar specimens under two different displacement rates and various temperatures, in order to investigate the effects of the stress and strain concentration at the notched section on the dynamic strain aging (DSA) behavior of carbon steel piping material. In addition, finite element simulations were conducted to evaluate quantitatively the stress and strain states for both types of specimen under uniaxial tensile loading. The results showed that serration and an increase in tensile strength, which are considered to be evidence of DSA in carbon steels, can be observed from tensile tests for notched-bar specimens. It was also found that the temperature region of DSA observed in the notched-bar specimens was higher than the DSA region observed in the standard tensile specimens tested under the same displacement rate. The results of finite element analysis showed that this behavior is associated with the high strain rate at the notched section, which is caused by the stress and strain concentration.

        The 8-hydroxyisocoumarin, named Centipedin, which has a significant antibiotic activity, was separated and solubilized with organic solvents, such as diethyl ether from centipede Scolopendra subspinipes multilans L. Koch. The Centipedin was purified by silicic acid column and high S cation exchange chromatography followed by reverse-phase HPLC. It was confirmed that Centipedin has a potent antibiotic effectiveness against Gram-negative Klebsiella pneumoniae ATCC 8308. The results showed that Centipedin blocks both DNA replication and RNA transcription during the growth of this pathogen in vivo. The biosynthesis of antibiotic 8-hydroxyisocoumarin was studied in vivo by feeding $[^{14}C]-labelled$ compound as a precursor to live centipede, in which $[^{14}C]acetate$ was the most efficiently incorporated into the Centipedin within 30 h after injection. Also, in vitro study on the biosynthesis of Centipedin showed that efficient incorporation of $[^{14}C]acetate$ occurred at pH range 5.0-7.0 for 10 h incubation and decreased significantly after then. It is suggested that 8-hydroxyisocoumarin is one of the defense compounds acting on bacterial infection in Scolopendra subspinipes.

        Centipede Scolopendra subspinipes multilans L. KOCH로부터 정제된 항균 물질 Centipedin의 항균 Spectrum 및 작용 Mechanism 연구

        지네 Scolopendra subspinipes multilans L. Koch로부터 정제한 Centipedin의 gram-negative, gram-positive bacteria 및 fungi에 대한 광범위 항균 spectrum을 조사하였으며 액체배지를 사용하여 각 균주에 대한 minimal inhibitory concentrations(MICs)값을 측정하였다. 인체의 기관중 주로 폐에 감염되는 gram-negative bacteria인 Klebsiella pneumoniae ATCC 8308 균주에 대한 MIC값이 $2{\mu}g/ml$로서 가장 강한 항균력을 나타내었고 화농균인 Proteus vulgaris NRRL B-123 균주에 대해서도 MIC값이 $16{\mu}g/ml$로 비교적 높은 항균력을 나타내었으며 항균제로서의 작용 mechanism을 시험한 결과 prokaryotic RNA transcription을 억제함을 알 수 있었다. Human blood leukemia(HL-60)와 mouse B lymphocyte myeloma cell 등의 동물세포에 대한 Centipedin의 cytotoxicity는 없는 것으로 확인되었다. A broad spectrum of antibiotic action was studied with Centipedin purified from centipede Scolopendra subspinipes multilans L. Koch aganist gram-positive, gram-negative bacteria and fungi. The minimal inhibitory concentrations(MICs) were determined in liquid medium. The significant antibiotic activity was obtained aganist gram-negative Klebsiella pneumoniae ATCC 8308 responsible for causing infection at lung and intestine. The MIC value against Klebsiella pneumoniae ATCC 8308 was $2{\mu}g/ml$, and this Centipedin was active against Proteus vulgaris NRRL B-123. In addition, it has been shown that Centipedin blocks procaryotic RNA transcription and a little of DNA replication system in vitro. Centipedin did not exhibit any significant cytotoxicity against animal cells such as human blood leukemia (HL-60) and mouse B lymphocyte myeloma cell.

        단위가축영양 : 수퇘지의 정액 성상과 혈중 항산화물 농도에 대한 통마늘분말 급여의 영향

        본 연구는 통마늘분말의 급여가 수퇘지의 정액 성상과 혈중 항산 화물 농도에 미치는 영향을 구명하기 위해 12개월령의 듀록종 수퇘지 9두를 공시하여 시험사료만을 급여한 대조구(4두)와 시험사료에 0.3% 통마늘분말을 첨가하여 급여한 실험구(5두)를 이용해 13주간 수행되었다. 통마늘분말의 급여가 수퇘지의 정액생산량과 정자농도에 미치는 영향을 8주간에 걸쳐 평가한 결과, 정액생산량에서는 통마늘분말 급여 여부에 따른 차이를 보이지 않았으나, 수퇘지 정액의 평균 총정자수는 0~4주의 평균 총정자수는 통마늘분말 급여여부에 상관없이 차이가 없었다. 그러나, 5~8주에 채취된 정액의 평균 총정자수는 통마늘분말을 급여한 실험구가 대조구에 비해 유의적으로 높게 나타났다(P<0.05). 통마늘분말 급여가 수퇘지의 성욕에 미치는 영향을 알아보기 연속 사정횟수와 전체 사출정액에 대한 총정자수를 조사한 결과, 통마늘분말을 급여한 실험구가 대조구에 비해 사정횟수와 총정자수가 유의적으로 높게 나타났다(P<0.05). 수퇘지에 대한 통마늘분말의 급여가 정장 및 혈장 내 polyphenol 농도에 미치는 영향에서는 정장 내에서의 polyphenol 농도는 통마늘분말 급여여부에 상관없이 차이를 보이지 않았다. 그러나, 혈장 내에서의 polyphenol 농도는 9주, 12주 및 13주차에 통마늘분말을 급여한 실험구가 대조구에 비해 유의적으로 높은 수치를 나타내었다(P<0.05). 본 실험의 결과를 종합하면, 정액채취용 수퇘지에 대한 통마늘분말의 급여는 성욕의 향상에 영향을 미쳐 정액채취빈도와 정액내 총 정자수를 증가시키고, 혈액 내 항산화물 농도를 높이는 것으로 나타나, 통마늘분말이 정액채취용 수퇘지의 성적을 향상시키는데 사료적 가치가 있는 것으로 사료된다. The objective of this study was to determine the effects of dietary supplementation of whole garlic powder (WGP) on semen characteristics and blood antioxidant level in boars. For this study, nine Duroc boars of 12 months age were used. Semen and blood samples were collected for 13 weeks, once in each week. The boars were fed the basal diet (BD; control) or BD supplemented with 3% WGP. There were no significant differences in the semen volume and sperm concentration between control and WGP group on all collection weeks. However, total sperm number per ejaculate was higher in the WGP group than that in the control group on collection weeks 6, 7 and 8 (P<0.05). Also, on collection weeks 5, 6, 7 and 8, mean of total ejaculated sperm numbers per boar were significantly higher in the WGP group compared to control group (P<0.05). On the other hand, ejaculation frequency per boar (boar`s libido) and total ejaculated sperm number per boar were significantly increased in the WGP group compared to the control group, respectively (P<O.05). Although there was no difference in polyphenol level in seminal plasma between two treatment groups, polyphenol level in blood serum was significantly higher in the WGP group on collection weeks 9, 12 and 13 (P<0.05). These results indicate that dietary supplementation of 3% WGP improves boar libido and semen productivity such as ejaculation frequency per boar, total sperm number per ejaculate, mean of total ejaculated sperm number per head. and elevate the blood level of antioxidant (polvphenon in boar serum.
