        Laccase- and Peroxidase-Free Tyrosinase Production by Isolated Microbial Strain

        Laccase- and peroxidase-free tyrosinase has commercial importance in the production of L-3, 4-dihydroxyphenylalanine (L-DOPA), which is mainly used in the treatment of Parkinson`s disease. In the present study, isolation of an actinomycetes microbial strain capable of producing only tyrosinase is reported. Among all soil isolates, three individual colonies revealed black color around the colony in the presence of tyrosine. Further screening for laccase and peroxidase activities using syringaldazine denoted that one of the isolates, designated as RSP-T1, is laccase and peroxidase negative and produces only tyrosinase. The microbe was authenticated as Streptomyces antibioticus based on 16S ribotyping. Effective growth of this isolate was noticed with the use of medium (pH 5.5) containing casein acid hydrolysate (10.0 g/l), K2HPO4 (5.0 g/l), MgSO4 (0.25 g/l), L-tyrosine (1.0 g/l), and agar (15 g/l). The scanning electron micrograph depicted that the microbe is highly branched and filamentous in nature. The enzyme production was positively regulated in the presence of copper sulfate. The impact of different fermentation parameters on tyrosinase production depicted that the maximized enzyme titer values were observed when this isolate was grown at 6.5 pH and at 30oC temperature under agitated conditions (220 rpm). Among all the studied physiological parameters, agitation played a significant role on tyrosinase production. Upon optimization of the parameters, the yield of tyrosinase was improved more than 100% compared with the initial yield.

        Pharmacopuncture of Bauhinia variegata Nanoemulsion Formulation against Diabetic Peripheral Neuropathic Pain

        Objectives: The objective of the study was to prepare Bauhinia variegata loaded nanoemulsion(formulation and determine the efficacy of herbal drug formulation against diabetic peripheral neuropathic pain through acupuncture technique. Methods: Nine different ba tches of nanoemulsion (NE1 NE9) of BVN was prepared by varying the S<sub>mix</sub> ratio and the concentration of oil. BVN was characterized to determine particle size, shape, zeta potential, polydispersity index, optical transmittance, drug release profile and stora ge stability. The optimized formulation was subjected to plantar test, behavioral tests of neuropathic pain and Von Frey filament stimulation test. Diabetes was induced by intraperitoneal injection of freshly prepared solution of Streptozotocin (60 mg/kg) to the experimental rats. Animals were made diabetic divided into four groups, Group I was untreated normal control group, Group II was diabetic control group, Group III was Bauhinia variegata extract ( treated group (100 mg/kg/day, p.o) and Group IV was BVN treated groups (100 mg/kg/day, p.o) acute and chronically. Results: The prepared B. variegata loaded nanoemulsion was nanosized (124 nm), spherical, uniform and stable over the period of 180 days with no change in physiochemical properties. The bl ood glucose and body weight of animals was normalizing after four weeks of treatment that was significant with BVN in comparison to diabetic control group. The chronic administration of BVN significantly (P<0.001) decreased hind paw withdrawal latency an d attenuated mechanical allodynia as compared with diabetic rats. Conclusion: Thus, BVN may be an effective drug formulation against diabetic peripheral neuropathic pain.

        Three dimensional hollow sulphide nanocomposites for supercapacitor electrodes

        The transition metal sulphides have gained sufficient attraction due to their high theoretical capacitance, low electronegativity, good thermal and electrical conductivity and good redox behaviour. In the present work, nanocomposites of nickel cobalt sulphide (NiCoS), zinc cobalt sulphide (ZnCoS), copper cobalt sulphide (CuCoS) and nickel-zinc-copper cobalt sulphide (NiZnCuCoS) have been prepared, by hydrothermal synthesis method, for their use as the supercapacitor electrode material. The X-ray diffraction analysis, Field Emission Scanning Electron Microscopy images and UV–Visible studies suggest the formation of quantum dots (as the crystallite size is in the range of 3.5 nm–6.0 nm) in all the nanocomposites. The aim of this study is to observe the change in the electrochemical properties of the transition metal sulphides by multi-metal doping. The electrochemical measurements reveal that among all the prepared nanocomposites, highest specific capacitance (150 F g-1 at 5 mV s-1) has been exhibited by CuCoS nanocomposites (nanowires like structure) whereas highest potential window (1.7 V) and hence highest energy density (16.5 W h kg-1 at 0.1 A g-1) has been exhibited by NiZnCuCoS nanocomposites. Presently, the main goal of supercapacitors is to provide high energy density. It is observed that multi-metal sulphides (NiZnCuCoS), when used as the supercapacitor electrode, provide better electrochemical performance which may be owing to the specific nanosheets like structures as well as good synergism between Ni, Zn, Cu and Co transition metals. Hence it is observed that by multi-metal doping, the supercapacitive behaviour of transition metal sulphides has improved.

        Testicular Cell Indices and Peripheral Blood Testosterone Concentrations in Relation to Age and Semen Quality in Crossbred (Holstein Friesian×Tharparkar) Bulls

        Present study analyzed the changes in peripheral blood testosterone concentrations and testicular cytogram in relation to age and semen quality in crossbred males. Three different age groups of crossbred males viz. bull calves (6 months, n = 5), young bulls (15 months, n = 5) and adult bulls (4 to 6 years, n = 8) were utilized for the study. Testicular fine needle aspiration cytology technique was used to quantify testicular cytology and their indices. Peripheral blood testosterone concentrations were measured using enzyme-linked immunosorbent assay method. Semen samples collected from adult bulls were microscopically evaluated for quality parameters. Mean peripheral blood testosterone concentrations in bull calves, young bulls and adult bulls were $2.28{\pm}0.09ng/mL$, $1.42{\pm}0.22ng/mL$ and $5.66{\pm}1.08ng/mL$ respectively, and that in adult bulls were significantly different (p<0.01) from young bulls and bull calves. There was no significant difference between the proportion of different testicular cells in bull calves and young bulls. Between young and adult bulls, significant differences (p<0.01) were observed in the proportion of spermatocytes, spermatozoa, and sperm: Sertoli cell ratio. The proportions of Sertoli cells showed a significant difference (p<0.01) between the three age groups. The number of primary spermatocytes had a positive correlation with peripheral blood testosterone concentrations in bull calves (r = 0.719, p<0.01). Number of Sertoli cells per 100 germ cells was negatively correlated with blood testosterone concentration in young bulls (r = -0.713, p<0.01). Among different semen parameters in adult bulls, ejaculate volume (r = 0.790, p<0.05) had positive relationship, and sperm motility had significant negative correlation (r = -0.711, p<0.05) with testosterone concentrations. The number of Sertoli cells and Sertoli cell index had a positive correlation with various semen quality parameters (p<0.001). Results of the present study conclude that number of Sertoli cells and Sertoli cell index are good indicators of semen quality, but peripheral blood testosterone concentrations may not have a direct relationship with various seminal attributes in crossbred bulls.

        Effect of Feeding Chemically Treated Mustard (Brassica campestris) Straw on Rumen Fibre Degrading Enzymes in Sheep

        The aim of the experiment was to study the changes in the activities of various rumen fibre degrading enzymes due to the feeding of chemically treated mustard (Brassica campestris) straw in sheep. Mustard straw (MS) (<5 cm particle size) was treated either with urea (4% (w/w), or with 2% sodium hydroxide (NaOH), or with alkaline hydrogen peroxide (2% NaOH and 1.5% hydrogen peroxide ($H_2O_2$)) and/or supplemented with 2% (w/w) urea. Seven maintenance type rations were prepared using MS (70 parts) with molasses (5 parts) and concentrate (25 parts). They were untreated MS (CMS), urea treated MS (UMS), urea supplemented MS (MSUS), alkali treated MS (AMS), alkali treated and urea supplemented MS (AMS-US), alkali $H_2O_2$ treated MS (AHMS) and alkali $H_2O_2$ treated and urea supplemented MS (AHMS-US). They were then compressed into a complete feed block with the help of block making machine. Forty two male hoggets of Malpura breed sheep were equally distributed into each treatment group and (were) offered feed and water ad libitum. At the end of 21 days of feeding trial, rumen liquor was collected through stomach tube from three animals in each group at 0 h, 4 h, 8 h, 12 h of post feeding. Results showed that the level of enzyme varied from 8.52 to 11.12, 40.85 to 50.37, 3.22 to 3.78, 2.09 to 2.77 and 31.44 to 44.24 units/100 ml SRL respectively for carboxymethyl cellulase (CMCase), $\alpha$-amylase, microcrystalline cellulase (MCCase), filter paper (FP) degrading enzyme and $\alpha$-glucosidase. Processing of MS affected the enzyme activities, in a way, that NaOH and AHP treatment significantly reduced CMCase and FP degrading enzyme. The effect of urea treatment showed an increase in the activity of MCCase and $\alpha$-glucosidase. But the supplementation of urea increased the activity of CMCase, FP degrading enzyme and $\alpha$-glucosidase. The CMCase, $\alpha$-amylase, $\alpha$-glucosidase activities were highest at 4hr whereas MCCase and FP degrading enzyme had maximum activities at 12 h post feeding Results suggested that MS might need longer time in the rumen for its effective degradation.

      • Comparative studies of gasification potential of agro-waste with wood and their characterization

        In this work, an experimental study of the gasification on wood was carried out in downdraft type fixed bed Gasifier attached with 10 kW duel fuel diesel engine. The main objective of the study was to use wood as the biomass fuel for downdraft Gasifier and to evaluate the operating parameter of gasifier unit to predict its performance in terms of gas yield and cold gas efficiency. The influence of different biomass on fuel consumption rate, gas yield and cold gas efficiency was studied. Composition of producer gas was also detected for measuring the lower heating value of producer gas to select the feed stock so that optimum performance in the existing gasifier unit can be achieved. Under the experimental conditions, Lower heating value, of producer gas, cold gas efficiency and gas yields, using wood as a feed stock, are $4.85MJ/m^3$, 46.57% and $0.519m^3/kg$.

        Monocrotophos Induces the Expression of Xenobiotic Metabolizing Cytochrome P450s (CYP2C8 and CYP3A4) and Neurotoxicity in Human Brain Cells

        <P>Expression of various cytochrome P450s (CYPs) in mammalian brain cells is well documented. However, such studies are hampered in neural/glial cells of human origin due to nonavailability of human brain cells. To address this issue, we investigated the expression and inducibility of CYP2C8 and CYP3A4 and their responsiveness against cyclophosphamide (CPA) and organophosphorus pesticide monocrotophos (MCP), a known developmental neurotoxicant in human neural (SH-SY5Y) and glial (U373-MG) cell lines. CPA induced significant expression of CYP2C8 and CYP3A4 in both types of cells in a time-dependent manner. Neural cell line exhibited relatively higher constitutive and inducible expression of CYPs than the glial cell line. MCP exposure alone could not induce the significant expression of CYPs, whereas the cells preexposed to CPA showed a significant response to MCP. Similar to the case of CPA induced expressions, neural cells were found to be more vulnerable than glial cells. Our data indicate differential expressions of CYPs in cultured human neural and glial cell lines. The findings were synchronized with protein ligand docking studies, which showed a significant modulatory capacity of MCP by strong interaction with CYP regulators-CAR and PXR. Similarly, the known CYP inducer CPA has also shown significant high docking scores with the two studied CYP regulators. We also observed a significant induction in reactive oxygen species (ROS), lipid peroxides (LPO), micronucleus (MN), chromosomal aberration (CA), and reduction in reduced glutathione (GSH) and catalase following the exposure of MCP. Moreover, the expressions of apoptotic markers such as caspase-3, caspase-9, Bax, and p53 were significantly upregulated, whereas the levels of antiapoptotic marker, Bcl2, was downregulated after the exposure of MCP in both cell lines. These findings confirm the involvement of ROS-mediated oxidative stress, which subsequently triggers apoptosis pathways in both human neural (SH-SY5Y) and glial (U373-MG) cell lines following the exposure of MCP.</P>

      • Effect of ZnO nanoparticles aggregation on the toxicity in RAW 264.7 murine macrophage

        Nanostructured zinc oxide (ZnO) has received much attention due to its biological and medical applications, where detailed knowledge about particle sizing, aggregation propensity and its related hazards are crucial. Herein, the aggregation propensity and dissolution behavior of ZnO nanoparticles in aqueous medium (PBS) were studied as a function of concentration and further correlated with its toxicity in RAW 264.7 murine macrophages. Fast formation of smaller aggregates having high dissolution rate was observed at low concentration ZnO (LC-ZnO). Compared to high concentration ZnO (HC-ZnO) aggregates, the LC-ZnO aggregates were highly pronounced in terms of reactive oxygen species generation and exerting cell apoptosis, ascribed to the secondary size effect, size-dependent cellular uptake and ion solubility. This study outlines the nanoparticle concentration as a key factor in scaling its aggregation, dissolution tendency and also emphasizes the accounting of ingested nanomaterials long-term fate inside the cells.
