A novel semi-interpenetrating polymer network (s-IPN) based on the entrapment of a thermosensitive polymer, the poly(vinyl methyl ether) (PVME), within a crosslinked 3D structure of poly(vinyl alcohol) (PVA) and poly(vinyl methyl etheralt- maleic acid) (COP) was synthesized by an autoclaving process. The preparation method avoids the use of toxic crosslinkers and allows the simultaneous sterilization of material. The PVA/COP/PVME hydrogel were characterized by Fourier transform infrared spectroscopy, thermal techniques, swelling kinetic measurements, scanning electron microscopy, and rheological analysis. The entrapment of PVME within the hydrated polymer framework significantly modified its transition temperature at pH 7.4 and pH 3 conditions. The swelling kinetics of the s-IPN were dependent on pH (7.4, 3 and 1), and temperature (25 and 37℃). The interpenetrated polymer chains reduced the internal pore sizes of crosslinked network without altering its elastic, solid-like behavior. The loading and in vitro release of 5-fluorouracil, a chemotherapeutic agent, from hydrogel systems were studied at different temperature and pH values. The hydrogels showed a sustained drug release up to 5 h at 37℃, in different pH media. The s-IPN exhibited a promising performance for a range of biomedical applications, in particular, for the controlled drug delivery in response to the pH and temperature conditions.

ZnS films were grown on (001) GaAs substrates at different temperatures by RF magnetron sputtering. The ZnS chemical stoichiometry was determined by Energy-dispersive X-ray spectroscopy (EDS), besides it allowed to find the residual impurities, mainly oxygen. The X-ray diffraction (XRD) analysis and Raman scattering reveal that ZnS deposited thin films showed hexagonal wurtzite crystalline phase. The films average crystallite size range was from 8.15 to 31.95 nm, which was determined using the Debye eScherrer equation for the peak W(101). Besides an experimental study of first- and second-order Raman scattering of ZnS films is made. An energy level diagram involving oxygen traps and interstitial sulphur ions is used to explain the origin of the radiative transitions observed in the room temperature photoluminescence (PL) spectra.

Cathepsin activities are responsible for mediating various pathways involved in immune response, including the apoptosis pathway, toll-like receptor (TLR) signaling, cytokine induction and activation of granule serine proteases. In the present study, we investigated cathepsin responses in the kidneys of olive flounder infected with Streptococcus parauberis, analyzing cathepsin expression using a label-free, quantitative proteomic approach in conjunction with quantitative real-time polymerase chain reaction (qRT-PCR). In proteomic analyses, we detected cathepsin B, D, L and S proteins, noting significant decreases and increases in cathepsins B and L, respectively, with infection. Taken together with an evaluation of cathepsin B, D, F, K, L, S and X gene expression in normal and infected kidneys by qRT-PCR, our results indicate that cathepsins B, D, L and S are the dominant lysosomal proteases in the immune system of the teleostei, olive flounder. Cathepsins F, K and X were regarded as minor cathepsins.

We present the structural and optical characterization of cadmium selenide sulphur (CdSe1-ySy) deposited by chemical bath deposition (CBD) technique at low-temperature (20 ± 2 ℃). The sulphur molar fraction is varied from 0 to 42.13%. The chemical stoichiometry is estimated by energy-dispersive X-ray spectroscopy (EDS). The CdSe1-ySy shows hexagonal wurtzite crystalline phase, which was found by X-ray diffraction (XRD) analysis and it was confirmed by Raman spectroscopy. The average grain size of the CdSe1-ySy films was ranged from 1.20 to 1.68 nm that was determined by Debye-Scherrer equation from W(002) direction and it was confirmed by high resolution transmission electron microscopy (HRTEM). This average grain size indicates a high quantum confinement because of it is smaller than the Bohr radii of CdS (2.8 nm) and CdSe (4.9 nm). Raman spectra show two dominant vibrational bands about 208 and 415 cm-1 associated at CdSe-1LO-like and CdSe-2LO-like. By transmittance measurements at room temperature are found that the optical band gap energies vary from 1.86 to 2.16 eV in the range of investigated sulphur molar fraction. Room temperature photoluminescence shows radiative bands in the visible range and a dominant band in the UV range, approximately 3.0 eV, which can be associated with a radiative transition, bound exciton to donor impurity.

Soft tunic syndrome of Halocynthia roretzi manifests as soft, weak, and rupturable tunics, causing mass mortality. Utilizing liquid chromatography-tandem mass spectrometry (LC-MS/MS), innate immune response was established by comparing hemolymph protein profiles of ascidians with healthy or softened tunics. Of 100 proteins in each individual ascidian, 59 proteins from healthy and 56 proteins from diseased ascidians were functionally classified. Proteins found only in diseased individuals included trypsin inhibitor and Hr-29, and with high exponentially modified protein abundance index (emPAI) values. From 41 proteins identified to be common to both healthy and diseased ascidians, 15 were associated with innate immune response. Ficolin 3, a component of the lectin-complement system, was significantly decreased in diseased ascidians, but a cell surface protein, type II transmembrane serine protease-1 (TTSP), was considerably elevated. These results suggest that trypsin inhibitor, ficolin 3, and TTSP are probably involved in the innate immune response related to this tunic disease. Beside, Hr-29 could be suggested as a biomarker for soft tunic syndrome.

The epidermal secretion of fish contains various non-specific immune substances that act as the first line of defense against invading pathogens. The present study investigated the level of mucosal antibodies, the activities of hemagglutinin and protease, and other enzymes in the skin mucus of farm reared olive flounder (Paralichthys olivaceus) for 1year, in order to gain an insight into the relationship between these mucosal immune substances and their seasonal variation. These levels varied significantly during different months of sample collection. The present study showed a positive correlation between water temperature and the level of mucosal antibodies, and an inverse relationship between the level of mucosal antibodies and the activity of mucosal hemagglutinin and protease, but no relationship between lysozyme activity and other innate immune substances. This relationship is thought to be a compensatory response in olive flounder to protect itself against pathogenic microorganisms which are inherently present in the aquatic environment.

Interferon gamma (IFN-γ) is a cytokine that plays a very important role in defining Th1 immune response in all vertebrates. In this study, recombinant IFN-γ (rIFN-γ) from the olive flounder (Paralichthys olivaceus) was produced in an Escherichia coli system using a pET expression vector. Stimulation of whole kidney leukocytes (immune-related cells) in vitro with the resulting rIFN-γ significantly induced the gene expression of interleukin-1β (IL-1β), signal transducer and activator of transcription 1 (STAT1), CXCL13-like chemokine (CXCL13), and IFN-γ. rIFN-γ also weakly induced the expression of IL-1β, tumor necrosis factor-α (TNF-α), CXCL13, and IFN-γ in olive flounder-derived HINAE (non-immune) cells. The effects of rIFN-γ against Edwardsiella tarda infection in vivo were assessed by intraperitoneally injecting a mixture of rIFN-γ (100 ng) and E. tarda (1 x 105 CFU/ml) into the olive flounder. The survival rate in the rIFN-γ-injected group was 60% compared to 0% in the group treated with E. tarda only, demonstrating that olive flounder IFN-γ is effective in reinforcing immune responses and preventing against edwardsiellosis.
