Purpose: The universal organic solvent dimethyl sulfoxide (DMSO)can be used as a differentiation inducer of many cancer cells andhas been widely used as a solvent in laboratories. However, itseffects on breast cancer cells are not well understood. The aimof this study is to investigate the effect and associated mechanismsof DMSO on mouse breast cancer. Methods: We appliedDMSO to observe the effect on tumors in a mouse breast cancermodel. Tumor-associated macrophages (TAMs) were tested byflow cytometry. Ex vivo tumor microenvironment was imitated by4T1 cultured cell conditioned medium. Enzyme-linked immunosorbentassays were performed to detect interleukin (IL)-10 andIL-12 expression in medium. To investigate the cytotoxicity ofDMSO on TAMs, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide (MTT) assays were performed. Results: We foundthat DMSO produced tumor retardation when injected into mouseperitoneal cavities in a certain concentration range (0.5–1.0 mg/g). Furthermore, as detected by flow cytometry, TAM subtypeswere found to be transformed. We further imitated a tumor microenvironmentin vitro by using 4T1 cultured cell conditionedmedium. Similarly, by using low concentration DMSO (1.0%–2.0% v/v), TAMs were induced to polarize to the classically activatedmacrophage (M1-type) and inhibited from polarizing intothe alternatively activated macrophage (M2-type) in the conditionedmedium. IL-10 expression in tumors was reduced, whileIL-12 was increased compared with the control. Furthermore, wereported that 2.0% (v/v) DMSO could lead to cytotoxicity in peritonealmacrophages after 48 hours in MTT assays. Conclusion:Our findings suggest that DMSO could exert antitumor effects in4T1 cancer-bearing mice by reversing TAM orientation and polarizationfrom M2- to M1-type TAMs. These data may providenovel insight into studying breast cancer immunotherapy.

Bioassay-guided fractionation of CHCl3 extract from the fermentation broth of a sponge Mycale plumose-derived actinomycete Saccharopolyspora sp. nov., led to the isolation of two known prodigiosin analogs - metacycloprodigiosin (1) and undecylprodigiosin (2). These compounds exhibited significant cytotoxic activities against five cancer cell lines: P388, HL60, A-549, BEL- 7402, and SPCA4. This is the first report on the significant cytotoxicity of metacycloprodigiosin (1) against human cancer cell lines.

The shallow-donor impurity states in cylindrical zinc-blende (ZB) InxGa1−xN/GaN quantum dots (QDs) have been theoretically investigated, considering the combined effects of an intense laser field (ILF), an external electric field, and hydrostatic pressure. The numerical results show that for an on-center impurity in ZB InxGa1−xN/GaN QD, (1) the ground-state binding energy of the donor impurity is a decreasing function of the laser-dressing parameter and/or the QD’s height; (2) as the QD’s radius decreases, the binding energy of the donor impurity increases at first, reaches a maximum value, and then drops rapidly; (3) the binding energy of the donor impurity is a decreasing function of the external electric field due to the Stark effect; (4) the binding energy of the donor impurity increases as the applied hydrostatic pressure becomes large. In addition, the position of the impurity ion was also found to have an important influence on the binding energy of the donor impurity. The physical reasons have been analyzed in detail.

Phospholipid hydroperoxide glutathione peroxidase (PHGPx) is an unique antioxidant enzyme that directly reduces lipid hydroperoxides in biomembranes. In the present work, the entire encoding region for Oryza sativa PHGPx was expressed in Escherichia coli M15, and the purified fusion protein showed a single band with 21.0 kD and pI = 8.5 on SDS- and IFE-PAGE, respectively. Judging from CD and fluorescence spectroscopy, this protein is considered to have a well-ordered structure with 12.2% $\alpha$-helix, 30.7%$\beta$-sheet, 18.5% $\delta$-turn, and 38.5% random coil. The optimum pH and temperature of the enzyme activity were pH 9.3 and 27$^{\circ}C$. The enzyme exhibited the highest affinity and catalytical efficiency to phospholipid hydroperoxide employing GSH or Trx as electron donor. Moreover, the protein displayed higher GSH-dependent activity towards t-Butyl-OOH and $H_2O_2$. These results show that OsPHGPx is an enzyme with broad specificity for hydroperoxide substrates and yielded significant insight into the physicochemical properties and the dynamics of OsPHGPx.

The Wnt signaling pathway is well known to play major roles in skeletal development and homeostasis. In certain aspects, fracture repair mimics the process of bone embryonic development. Thus, the importance of Wnt signaling in fracture healing has become more apparent in recent years. Here, we summarize recent research progress in the area, which may be conducive to the development of Wnt-based therapeutic strategies for bone repair.

This paper investigates the distributed fixed-time attitude synchronization control problem for multiplerigid spacecraft system with external disturbances. Based on sliding-mode estimators, the authors remove therequirement of neighbours’ input control information. Using the fixed-time-based terminal sliding mode, the distributedadaptive control laws are developed to guarantee the attitude tracking errors converge to the regions in fixedtime independent of initial conditions, and adaptive laws are employed to deal with external disturbances. Finally,numerical simulations are presented to illustrate the performance of the proposed controllers.
