        Estimation of Rumen Microbial Protein Supply Using Urinary Purine Derivatives Excretion in Crossbred Calves Fed at Different Levels of Feed Intake

        A study was carried out to study the response of total purine derivatives (PD) excretion in urine to determine microbial N (MN) supply at four fixed levels of feed intake (namely 95, 80, 60 and 40% of voluntary intake). The crossbred (CB) calves were allocated according to a $4{\times}4$ Latin Square Design and fed wheat straw and concentrate (1:1). The rate of PD excretion (mmol/d) as a linear function of feed intake was 15.85/kg DMI and 20.12/kg DOMI. Based on the endogenous and PD excretion rates obtained in this study, a relationship between daily urinary PD excretion (Y, mmol) and daily microbial protein supply (X, mmol) was developed for crossbred calves as Y = 0.83X+0.296 kg $W^{0.75}$. The derived microbial N values using this equation differed (p<0.001) among the 4 groups and was the highest in L-95 followed by L-80, L-60 and L-40. The relationship between urinary nitrogen loss (Y, g/d) and DOMI (X, kg/d) was established as: Y = 6.038X+21.753 ($r^2$ = 0.663, p<0.01). When urinary excretion of PD (Y, mmol/d) was plotted against intake of DM and DOM (X, kg/d), the equations obtained were: Y = 7.1711X+8.674 ($r^2$ = 0.889, p<0.01) and Y = 12.434X+7.683 ($r^2$ = 0.896, p<0.01), respectively. The proportional contribution of allantoin and uric acid to total PD remained stable irrespective of level of feed intake. Similarly, urinary excretion of creatinine did not differ (p>0.05) between animals fed at different levels. The MN supply was the highest to animals at intake levels L-95, and decreased linearly with corresponding decrease in feed intake. However, the MN supply when expressed per kg DOMI remained statistically (p>0.05) similar irrespective of level of intake. The results revealed that the excretion of urinary purine derivatives were positively correlated with the level of feed intake as well as rumen microbial supply and thus it could be a good indicator for measuring the microbial protein supply and nutritional status of animals.

        Effects of Phytase Supplementation on the Performance of Broiler Chickens Fed Maize and Wheat Based Diets with Different Levels of Non-phytate Phosphorus

        An experiment was conducted to evaluate the effects of phytase supplementation on the growth performance, nutrients utilization and bone mineralization in broiler chickens. Day-old broiler chicks (n=480) were equally devided into eight treatment groups and fed maize or wheat based isocaloric, isonitrogenous and isocalcium diets having two non phytate phosphorus (NPP) concentrations (0.50% and 0.30%) and two phytase levels (0 and 500 phytase units/kg diet) in a 42 days growth trial. Maize based dietary treatments were MC (NPP 0.50%, MN (NPP 0.30%), MNP (MN+500 units of phytase) and MCP (MC+500 units of phytase), whereas wheat based experimental diets were WC (NPP 0.50%), WN (NPP 0.30%), WNP (WN+500 units of phytase) and WCP (WC+500 units of phytase). The NPP levels were maintained by dicalcium phosphate. Reduction in dietary NPP depressed live weight gain and feed intake and increased feed conversion ratio (FCR). Phytase supplementation to low NPP (0.30%) diets significantly (p<0.05) improved the growth performances of broilers. The supplementation to low NPP diets allowed complete, safe and economic replacement of dietary inorganic P (dicalcium phosphate) to reduce feed cost per kg live weight gain of broilers. Reduction in dietary NPP did not affect retention of nutrients except phosphorus (P) but had a significant (p<0.05) depression in tibia ash and minerals (Ca, P) concentration in serum and tibia ash. Phytase supplementation at low NPP level was effective (p<0.05) in improving the retention of dry matter, Ca and P and Ca and P concentration in serum and tibia ash. However, the supplementation was not effective at high level of NPP (0.50%). There were no significant (p>0.05) differences in carcass quality among dietary treatments. The response of phytase was greater in low NPP and maize based diets as compared with high NPP and wheat based diets, respectively. The results show that phytase supplementation to low NPP (0.30%) diets improved the growth performance, relative retention of nutrients (N, Ca and P) and minerals (Ca, P) status of blood and bone in broiler chickens, with a better efficacy in maize based diets.

        Accuracy of Ultrasonography in Early Pregnancy Diagnosis in Doe

        The present study was undertaken to evaluate the accuracy of ultrasonography in early pregnancy diagnosis in goats. Ultrasonographic scanning with real time B-mode ultrasound machine having 5 MHz linear array transducer was performed on gravid uterus (n=24) obtained from slaughterhouse (Group I). Crown rump length (CRL) measured by ultrasound was found significantly different (p<0.05) with actual CRL measured after dissection in early pregnancy. However, age predicted by ultrasound through the measurement of CRL was found highly correlated (r=0.92) with age measured after dissection through CRL and the weight of fetus. Ages predicted by ultrasound through the measurement of trunk diameter (TD) and uterine diameter (UD) and ages measured after dissection were found highly and equally correlated (r=0.98) and did not differ significantly. Data from six does synchronized (Group II) with PGF2$\alpha$ (Estrumate) at 11 days apart were collected through ultrasound from 17 to 42 days post breding. The correlation between CRL and gestational age was high (r=0.97) in day 30 to 42 post breeding. A high coefficient of correlation (r=0.98) was also observed between predicated age by ultrasound and actual age calculated after kidding. The correlation between CRL and gestational age by the formula Y=(a+bX) i.e. Y=24.42+0.39 X where Y=gestational age and X=CRL, was recorded very high (r=0.99). Accuracy of ultrasonography was lowest on day 17 to 19 (66%) and reached 100% on day 34. Data from 30 does (group III) randomly subjected to only one time ultrasounds scanning to assess the accuracy of pregnancy diagnosis were also obtained. Ages predicted by TD and UD measurements were observed to be non-significantly different with actual age obtained after kidding and correlation between ages predicted by TD and UD measurement with actual age after kidding was found equally and highly correlated (r=0.98). The operator's accuracy in the whole experiment including all three groups was found to be 92%. The sensitivity was 93% and specificity was 86%. From the present study, it was observed that CRL was the most reliable parameter to find out gestational age in early pregnancy and the new formula derived was found very accurate to find out gestational age. TD and UD were also found to be equally reliable parameter to find out gestational age in mid and late stage of pregnancy through ultrasonography. It was concluded that ultrasonography by real time B mode with 5 MHz transrectal transducer was found to be reliable, safe and accurate and practicable means in diagnosing early pregnancy diagnosis as early as 25 days post breeding.

        Effect of Sources of Supplementary Protein on Intake, Digestion and Efficiency of Energy Utilization in Buffaloes Fed Wheat Straw Based Diets

        Sixteen adult male buffaloes (average body weight $443{\pm}14kg$) were equally distributed into four groups in an experiment to study the effect of supplementary protein sources on energy utilization efficiency in buffaloes fed a wheat straw-based diet. The animals in the control group were offered a basal diet composed of 700 g deoiled ground nut cake and ad libitum wheat straw. Animals of other groups were offered 1.8 kg of soyabean meal (SBM), linseed meal (LSM) or mustard cake (MC) along with the basal diet. Protein supplementation increased the digestibility of DM (p<0.01), OM (p<0.01) CP (p<0.01) and CF (p<0.05). Maximum CP digestibility was observed on SBM, followed by LSM and MC when compared to the control. Total DMI and DOMI was significantly (p<0.01) higher in protein supplemented groups with no differences between treatment groups. Digestible crude protein (DCP) intake and N balance were significantly (p<0.01) different between the groups; maximum response was obtained with SBM supplementation, followed by LSM and MC. Faecal energy was significantly (p<0.01) lower in SBM and LSM groups in comparison to other groups. Methane production (% DEI) was significantly (p<0.05) lower on the SBM treatment. Metabolizable energy (ME) intake increased significantly due to protein supplementation. Metabolizable energy intake (MEI) of animals in the MC group was less than LSM and SBM. Energy balance was increased significantly (p<0.01) due to protein supplementation and within supplement variation was also significant with maximum balance in SBM followed by LSM and MC groups. Protein supplementation significantly (p<0.05) increased the digestibility and metabolizability of energy from whole ration. Metabolizable energy (ME) content (Mcal/kg DM) of SBM, LSM and MC was 4.49, 3.56 and 2.56, respectively. It was concluded that protein supplementation of wheat straw increased intake, digestibility and metabolizability of energy and maximum response could be obtained when soybean meal was used as a supplement.

      • A sensitive fluorescent probe for the polar solvation dynamics at protein-surfactant interfaces

        <P>Relaxation dynamics at the surface of biologically important macromolecules is important taking into account their functionality in molecular recognition. Over the years it has been shown that the solvation dynamics of a fluorescent probe at biomolecular surfaces and interfaces account for the relaxation dynamics of polar residues and associated water molecules. However, the sensitivity of the dynamics depends largely on the localization and exposure of the probe. For noncovalent fluorescent probes, localization at the region of interest in addition to surface exposure is an added challenge compared to the covalently attached probes at the biological interfaces. Here we have used a synthesized donor-acceptor type dipolar fluorophore, 6-acetyl-(2-((4-hydroxycyclohexyl)(methyl) amino) naphthalene) (ACYMAN), for the investigation of the solvation dynamics of a model protein-surfactant interface. A significant structural rearrangement of a model histone protein (H1) upon interaction with anionic surfactant sodium dodecyl sulphate (SDS) as revealed from the circular dichroism (CD) studies is nicely corroborated in the solvation dynamics of the probe at the interface. The polarization gated fluorescence anisotropy of the probe compared to that at the SDS micellar surface clearly reveals the localization of the probe at the protein-surfactant interface. We have also compared the sensitivity of ACYMAN with other solvation probes including coumarin 500 (C500) and 4-(dicyanomethylene)-2-methyl-6-(p-dimethylamino-styryl)-4H-pyran (DCM). In comparison to ACYMAN, both C500 and DCM fail to probe the interfacial solvation dynamics of a model protein-surfactant interface. While C500 is found to be delocalized from the protein-surfactant interface, DCM becomes destabilized upon the formation of the interface (protein-surfactant complex). The timescales obtained from this novel probe have also been compared with other femtosecond resolved studies and molecular dynamics simulations.</P>

        Cytokines Expression and Nitric Oxide Production under Induced Infection to Salmonella Typhimurium in Chicken Lines Divergently Selected for Cutaneous Hypersensitivity

        In the present study, the impact of Salmonella Typhimurium on cell-mediated immunity (CMI) was investigated in 5 week-old immuno divergent broiler lines selected for the high and low response to phytohemagglutinin-P. The immune response was assessed in peripheral-blood mononuclear cells (PBMCs) induced with Salmonella Typhimurium at different time intervals (0 h, 0.5 h, 2 h, 4 h, 6 h, 12 h and 24 h). The differential mRNA expression patterns of IFN-${\gamma}$, IL-2 and iNOS were evaluated by quantitative real time PCR. In-vitro production of nitric oxide (NO) was also estimated in the culture supernatant and correlated with iNOS mRNA expression. Present study showed higher production of NO in the high cell-mediated line (HCMI) as compared to the low cell-mediated line (LCMI) upon stimulation with Salmonella Typhimurium. Correspondingly, higher mRNA expression of iNOS and IFN-${\gamma}$ were observed in high response birds (HCMI); but IL-2 was down regulated in this line compared to the low response birds (LCMI). Significantly (p<0.05) higher expression of iNOS, IFN-${\gamma}$ and higher production of NO in high line indicated that the selection for PHA-P response might be employed for increasing the immune competence against Salmonella Typhimurium in chicken flocks.

        Monocrotophos Induces the Expression of Xenobiotic Metabolizing Cytochrome P450s (CYP2C8 and CYP3A4) and Neurotoxicity in Human Brain Cells

        <P>Expression of various cytochrome P450s (CYPs) in mammalian brain cells is well documented. However, such studies are hampered in neural/glial cells of human origin due to nonavailability of human brain cells. To address this issue, we investigated the expression and inducibility of CYP2C8 and CYP3A4 and their responsiveness against cyclophosphamide (CPA) and organophosphorus pesticide monocrotophos (MCP), a known developmental neurotoxicant in human neural (SH-SY5Y) and glial (U373-MG) cell lines. CPA induced significant expression of CYP2C8 and CYP3A4 in both types of cells in a time-dependent manner. Neural cell line exhibited relatively higher constitutive and inducible expression of CYPs than the glial cell line. MCP exposure alone could not induce the significant expression of CYPs, whereas the cells preexposed to CPA showed a significant response to MCP. Similar to the case of CPA induced expressions, neural cells were found to be more vulnerable than glial cells. Our data indicate differential expressions of CYPs in cultured human neural and glial cell lines. The findings were synchronized with protein ligand docking studies, which showed a significant modulatory capacity of MCP by strong interaction with CYP regulators-CAR and PXR. Similarly, the known CYP inducer CPA has also shown significant high docking scores with the two studied CYP regulators. We also observed a significant induction in reactive oxygen species (ROS), lipid peroxides (LPO), micronucleus (MN), chromosomal aberration (CA), and reduction in reduced glutathione (GSH) and catalase following the exposure of MCP. Moreover, the expressions of apoptotic markers such as caspase-3, caspase-9, Bax, and p53 were significantly upregulated, whereas the levels of antiapoptotic marker, Bcl2, was downregulated after the exposure of MCP in both cell lines. These findings confirm the involvement of ROS-mediated oxidative stress, which subsequently triggers apoptosis pathways in both human neural (SH-SY5Y) and glial (U373-MG) cell lines following the exposure of MCP.</P>

        Serum and CSF Mineral Profile of Himalayan Yak (Bas grunniens) in their Natural Habitat

        Mineral profile of serum and cerebrospinal fluid (CSF) of 15 Himalayan Yak (adult female, n=8; adult male n=4 and young male,n=3) was studied in their natural habitat at an altitude of 3300 meters above mean sea level at Sangla in north western Himalayas. The macro and micro minerals estimated in serum and CSF were; Sodium, Potassium, Calcium and Magnesium and Zinc, Copper and Iron respectively. The values recorded among different Yak groups did not significantly differ from each other except serum iron and haemoglobin which were significantly higher (p<0.05 ) in young male Yaks compared to the adult male and female Yaks. An observation of great significance was considerably higher potassium and lower sodium level in Yaks compared to other bovine species of plains. The serum potassium values in some adult female Yaks were recorded as high as 10.4 mEq/l and the values varied between 6.6 to 9.8 mEq/l in young male Yaks. The serum Sodium values and Na:K ratios in Yak serum ranged between 117.5 to 122.6 mEq/l and 13.7 to 15.3 respectively. The possible relationship of high serum Potassium value with hypoxic conditions and hostile mountain environment has been discussed.

        Status of Lipid Peroxidation in Normal Cycling and α-Tocopherol Supplemented Anestrus Buffalo Heifers (Bubalus bubalis)

        The present study was undertaken to analyze the status of erythrocytic malonyl dialdehyde (MDA) as an index of lipid peroxidation in normal cycling and ${\alpha}$-tocopherol supplemented anestrus buffalo heifers. Before supplementation, the blood samples were collected at weekly interval in anestrus and normal cycling buffalo heifers and data from four samples was pooled to establish pre-supplementation base line. Thereafter, the blood samples from anestrus and ${\alpha}$-tocopherol supplemented anestrus buffalo heifers were taken at fortnightly interval for 12 weeks during supplementation and for four weeks during the post-supplementation period. The pre-supplementation endogenous erythrocytic MDA, 0.3% $H_2O_2$ induced-erythrocytic MDA and 1.5% $H_2O_2$ induced-erythrocytic MDA levels (nmol/g Hb) increased significantly (p<0.05) in anestrus heifers ($372.3{\pm}5.0$, $564.4{\pm}5.7$, $862.4{\pm}8.5$) when compared to normal cycling animals ($289.6{\pm}3.0$, $508.2{\pm}6.0$, $777.7{\pm}4.9$). Oral supplementation of 3,000 mg of ${\alpha}$-tocopherol per week per animal in anestrus heifers resulted in a progressive and significant (p<0.01) decline in erythrocytic MDA levels. These observations implied that supplementation of ${\alpha}$-tocopherol to anestrus buffalo heifers ameliorated their antioxidant status by alleviating the effects of oxidative stress.
