        Probiotics in Drinking Water Alleviate Stress of Induced Molting in Feed-deprived Laying Hens

        An experiment was conducted to evaluate the physiological changes of laying hens subjected to feed removal during induced molting while received probiotics in the drinking water. Post-molt performance and egg quality criteria were also studied. Ninety 78-week-old Hy-line W36 laying hens were divided into two treatment groups according to equal body weight and subjected to induced molting by continuous feed removal until around 30% BW reduction. The experiment lasted 12 wks consisting of 4-wk molting and 8-wk post-molt periods. Treatment 1 received no probiotics and was considered as the control. Treatment 2 was similar to the control except that hens received probiotics in the drinking water at 400 mg/L during feed deprivation. The results indicated that hens in both groups went out of production by Day 5. However, hens received probiotics reached 5 and 50% egg production sooner than the control (30 and 52 days vs. 31 and 54 days). Starvation during molting increased heterophil to lymphocyte (H/L) ratio, hematocrit and plasma T4 and $Na^+$ levels while plasma T3 and Cl- levels were decreased. Probiotics had no significant impact on BW reduction during molt. Post-molt egg production and egg mass were higher in hens which previously received probiotics, but these responses were not significant. However, feed conversion ratio was significantly better in hens which received probiotics. Hematocrit, plasma thyroid hormone concentrations (T3 and T4) and plasma $Na^+$, $K^+$ and Cl- levels during molting were not significantly influenced by supplementation of probiotics. However, H/L ratio showed a significant (p<0.05) reduction in birds which received probiotics suggesting beneficial effects of this product for feed-deprived laying hens. No significant difference was observed in post-molt egg quality criteria.

        Evaluation of a Dietary Organic Selenium Supplement at Different Dietary Protein Concentrations on Growth Performance, Body Composition and Antioxidative Status of Broilers Reared under Heat Stress

        Three hundred chicks were randomly assigned among four treatments to evaluate the effect of an organic selenium supplement at various levels of dietary protein. Two levels of supplemental selenium (0 and 0.3 mg/kg) from zinc-L-selenomethionine were tested at two levels of dietary protein (normal and reduced) in a completely randomized design with $2{\times}2$ factorial layout. The experiment lasted up to 49 d of age during which all birds were exposed to $31{\pm}1^{\circ}C$. The effects of selenium or its interaction with CP on growth performance and carcass characteristics were not significant. However, feeding the reduced-CP diet decreased weight gain in the starting period and increased liver and abdominal fat weights relative to body weight. Ferric reducing ability of plasma (FRAP) was not significantly affected by dietary CP and Se or their interaction though FRAP values were numerically higher in the Se-supplemented group. Dietary CP content did not affect the activity of plasma glutathione peroxidase (GSHPx), though Se significantly elevated plasma GSHPx activity. The interaction of CP and Se was not significant for FRAP and plasma GSHPx activity.

        Adsorptive Immobilization of Acetylcholine Esterase on Octadecyl Substituted Porous Silica: Optical Bio-analysis of Carbaryl

        A sensory element against carbaryl, as a widely used pesticide was prepared based on adsorbed acetylcholine esterase (AChE) from Torpedo california. Octadecyl was substituted on macro-porous silica, confirmed by infra-red (IR) spectroscopy and quantitatively estimated through thermo-gravimetric analysis (TGA). Immobilization of the enzyme was achieved by adsorption on this support. Activity of the immobilization product was measured as a function of the loaded enzyme concentration, and maximum binding capacity of the support was estimated to be 43.18 nmol.mg-1. The immobilized preparations were stable for more than two months at storage conditions and showed consistency in continuous operations. Possible application of the immobilized AChE for quantitative analysis of carbaryl is proposed in this study.

        The Bacillus subtilis and Lactic Acid Bacteria Probiotics Influences Intestinal Mucin Gene Expression, Histomorphology and Growth Performance in Broilers

        The aim of the present study was to evaluate the effect of commercial monostrain and multistrain probiotics in diets on growth performance, intestinal morphology and mucin gene (MUC2) expression in broiler chicks. Three hundred seventy-eight 1-d-old male Arian broiler chicks were allocated in 3 experimental groups for 6 wk. The birds were fed on a corn-soybean based diet and depending on the addition were labeled as follows: control-unsupplemented (C), birds supplemented with Bacillus subtilis (BS) and lactic acid bacteria (LAB) based probiotics. Each treatment had 6 replicates of 21 broilers each. Treatment effects on body weight, feed intake, feed conversion ratio and biomarkers such as intestinal goblet cell density, villus length, villus width, and mucin gene expression were determined. Total feed intake did not differ significantly between control birds and those fed a diet with probiotics (p>0.05). However, significant differences in growth performance were found. Final body weight at 42 d of age was higher in birds fed a diet with probiotics compared to those fed a diet without probiotic (p<0.05). Inclusion of Bacillus subtilis based probiotic in the diets also significantly affected feed conversion rate (FCR) compared with control birds (p<0.05). No differences in growth performance were observed in birds fed different types of probiotic supplemented diets. Inclusion of lactic acid bacteria based probiotic in the diets significantly increased goblet cell number and villus length (p<0.05). Furthermore, diets with Bacillus subtilis based probiotics significantly increased gene expression (p<0.05), with higher intestinal MUC2 mRNA in birds fed diet with probiotics compared to those fed the control diet. In BS and LAB probiotic fed chicks, higher growth performance may be related to higher expression of the MUC2 gene in goblet cells and/or morphological change of small intestinal tract. The higher synthesis of the mucin gene after probiotic administration may positively affect bacterial interactions in the intestinal digestive tract, intestinal mucosal cell proliferation and consequently efficient nutrient absorption.
