Background/Aims Patients with long-standing diabetes often demonstrate intestinal dysfunction and abdominal pain. However, the pathophysiology of abdominal pain in diabetic patients remains elusive. The purpose of study was to determine roles of voltage-gated sodium channels in dorsal root ganglion (DRG) in colonic hypersensitivity of rats with diabetes. Methods Diabetic models were induced by a single intraperitoneal injection of streptozotocin (STZ; 65 mg/kg) in adult female rats, while the control rats received citrate buffer only. Behavioral responses to colorectal distention were used to determine colonic sensitivity in rats. Colon projection DRG neurons labeled with DiI were acutely dissociated for measuring excitability and sodium channel currents by whole-cell patch clamp recordings. Western blot analysis was employed to measure the expression of NaV1.7 and NaV1.8 of colon DRGs. Results STZ injection produced a significantly lower distention threshold than control rats in responding to colorectal distention. STZ injection also depolarized the resting membrane potentials, hyperpolarized action potential threshold, decreased rheobase and increased frequency of action potentials evoked by 2 and 3 times rheobase and ramp current stimulation. Furthermore, STZ injection enhanced neuronal sodium current densities of DRG neurons innervating the colon. STZ injection also led to a significant upregulation of NaV1.7 and NaV1.8 expression in colon DRGs compared with age and sex-matched control rats. Conclusions Our results suggest that enhanced neuronal excitability following STZ injection, which may be mediated by upregulation of NaV1.7 and NaV1.8 expression in DRGs, may play an important role in colonic hypersensitivity in rats with diabetes. This paper presents a dual-algorithm search method (GWO-GSO) combining grey wolf optimization (GWO) and golden-section optimization (GSO) to realize maximum power point tracking (MPPT) for photovoltaic (PV) systems. First, a modified grey wolf optimization (MGWO) is activated for the global search. In conventional GWO, wolf leaders possess the same impact on decision-making. In this paper, the decision weights of wolf leaders are automatically adjusted with hunting progression, which is conducive to accelerating hunting. At the later stage, the algorithm is switched to GSO for the local search, which play a critical role in avoiding unnecessary search and reducing the tracking time. Additionally, a novel restart judgment based on the quasi-slope of the power-voltage curve is introduced to enhance the reliability of MPPT systems. Simulation and experiment results demonstrate that the proposed algorithm can track the global maximum power point (MPP) swiftly and reliably with higher accuracy under various conditions.

TIM-1 (also known as KIM-1 and HAVcr-1) is a type I transmembrane glycoprotein member of the TIM family that may play important roles in innate and adaptive immune responses. The overexpression of proteins associated with membrane proteins is a major obstacle to overcome in studies of membrane protein structures and functions. In this study, we successfully coupled the overexpression of the TIM-1 protein with a C-terminal enhanced green fluorescent protein (GFP) tag in Escherichia coli. To the best of our knowledge, this report is the first to describe the overexpression of human TIM-1 in E. coli. The purified TIM-1-EGFP fusion protein recognized and bound directly to apoptotic cells and did not to bind to viable cells. Furthermore, we confirmed that the interactions of TIM-1-EGFP with apoptotic cells were blocked by TIM-1-Fc fusion proteins. This fusion protein represents a readily obtainable source of biologically active TIM-1 that may prove useful in future studies of human TIM-1.

Conventional maximum power point tracking (MPPT) methods are ineffective under partially shaded conditions because multiple local maximum can be exhibited on power–voltage characteristic curve. This study proposes an improved cuckoo search (ICS) MPPT method after investigating the cuckoo search (CS) algorithm applied in solving multiple MPPT. The algorithm eliminates the random step in the original CS algorithm, and the conception of low-power, high-power, normal and marked zones are introduced. The adaptive step adjustment is also realized according to the different stages of the nest position. This algorithm adopts the large step in low-power and marked zones to reduce search time, and a small step in high-power zone is used to improve search accuracy. Finally, simulation and experiment results indicate that the promoted ICS algorithm can immediately and accurately track the global maximum under partially shaded conditions, and the array output efficiency can be improved.

Under partial shading conditions (PSCs), multiple maximums may be exhibited on the P-U curve of string inverter photovoltaic (PV) systems. Under such conditions, heuristic methods are invalid for extracting a global maximum power point (GMPP); intelligent algorithms are time-consuming; and model-based methods are complex and costly. To overcome these shortcomings, a novel hybrid MPPT (MPF-IP&O) based on a model-based peak forecasting (MPF) method and an improved perturbation and observation (IP&O) method is proposed. The MPF considers the influence of temperature and does not require solar radiation measurements. In addition, it can forecast all of the peak values of the PV string without complex computation under PSCs, and it can determine the candidate GMPP after a comparison. Hence, the MPF narrows the searching range tremendously and accelerates the convergence to the GMPP. Additionally, the IP&O with a successive approximation strategy searches for the real GMPP in the neighborhood of the candidate one, which can significantly enhance the tracking efficiency. Finally, simulation and experiment results show that the proposed method has a higher tracking speed and accuracy than the perturbation and observation (P&O) and particle swarm optimization (PSO) methods under PSCs.

The Multi-Purpose Overload Robot (CMOR) is a key subsystem of China Fusion Engineering Test Reactor (CFETR) remote handling system. Due to the long cantilever and large loads of the CMOR, it has a large rigid-flexible coupling deformation that results in a poor position accuracy of the end-effector. In this study, based on the Levenberg-Marquardt algorithm, the spatial grid, and the linearized variable load principle, a variable parameter compensation model was designed to identify the parameters of the CMOR's kinematics models under different loads and at different poses so as to improve the trajectory tracking accuracy. Finally, through Adams-MATLAB/Simulink, the trajectory tracking accuracy of the CMOR's rigid-flexible coupling model was analyzed, and the end position error exceeded 0.1 m. After the variable parameter compensation model, the average position error of the end-effector became less than 0.02 m, which provides a reference for CMOR error compensation.
miR-19a Promotes Cell Growth and Tumorigenesis through Targeting SOCS1 in Gastric Cancer

Accumulating evidence has shown that microRNAs are involved in cancer development and progression. However, it remains unknown about the potential role of miR-19a in the pathogenesis of gastric cancer. Here, we report that suppressor of cytokine signaling 1 (SOCS1) is a novel target of miR-19a in gastric cancer cells and that miR-19a expression is inversely correlated with SOCS1 expression in gastric cancer cells and a subset of gastric cancer tissues. Ectopic expression of miR-19a dramatically promoted proliferation and tumorigenicity of gastric cancer cells both in vitro and in vivo. Moreover, we showed that silencing of SOCS1 promoted cell growth and colony formation resembling that of miR-19a overexpression, whereas re-introduction of SOCS1 (without the 3'-UTR) attenuated the pro-tumorigenic functions. Taken together, our findings suggest that the SOCS1 gene is a direct target of miR-19a, which functions as an oncogenic miRNA in gastric cancer by repressing the expression of tumor suppressor SOCS1.
