Glucagon-like peptide-1 (GLP-1) was a potential therapeutic drug for type II diabetes, mainly because of the stimulatory effect on insulin secretion under condition of high blood glucose. We used PCR to obtain a recombination gene, GGH, in which two GLP-1 (GLP-1A2G) mutants were connected in series and then fused to the N terminal of human serum albumin. The fusion gene was inserted into pGAPZaA plasmid with Saccharomyces cerevisiae α- factor secretion signal sequence, and was expressed by the glyceraldehyde-3-phosphate dehydrogenase (GAP) promoter. The engineered strain was constructed by integrating the recombinant plasmid pGAPZαA/GGH into the genome of Pichia pastoris GS115. Genome PCR and western blot showed that the recombinant P. pastoris successfully expressed the fusion protein GGH. The yield of GGH reached 78 mg/L after 72 h fermentation in a flask, using glucose as the optimal carbon source. Fed-batch fermentation was investigated in a 5 L bioreactor, and the expression level of GGH reached 246 mg/L in 52 h. The fusion protein GGH was purified in four steps, and the final purity was 96.1%. The in vitro bioactivity of GGH was the same as that expressed in P. pastoris by the AOX1 promoter. This study described an efficient way to express GGH fusion protein in P. pastoris using GAP promoter, fermentation was easier to control without carbon source change and fermentation time was 20 h less than AOX1 promotercontrolled GGH fermentation.
Research on the Flame Simulation Method Based on the Particle System

In reality, flame is a common fluid scene. In this paper, we simulated flame by using improved particle system and detail the properties and changes of particles in the model. We realized a realistic color change and dynamic flickering flame characteristics. We used texture mapping method to render the flame and used the hierarchical programming structure to provide a simple and practical interactive flame simulation. We also designed and implemented demonstration system of flame simulation. Experiments show that using the flame image generated in this paper can not only satisfy the visual effect but also meet the real-time requirements.

A new prenylated dihydrobenzofuran derivative (1), was isolated from the rhizomes of Atractylodes lancea DC (Asteraceae), along with ten known compounds, including atractylenolide II (2), $\varphi$-taraxasteryl acetate (3), taraxerol acetate (4), $\beta$-sitosterol (5), stigmasterol (6), $\beta$-eudesmol (7), atractylenolide III (8), atractylenolide IV (9), daucosterol (10), and stigmasterol 3-O-$\beta$-D-glucopyranoside (11). The structure of the new compound (1) was elucidated as trans-2-hydroxyisoxypropyl-3-hydroxy-7-isopentene-2,3-dihydrobenzofuran-5-carboxylic acid by the combination of 1D, 2D NMR analysis and mass spectrometry, and it was the first reported 2,3-dihydrobenzofuran derivative having a carboxyl residue at C-5 and an isopentene moiety at C-7 contemporaneously. In addition, compound 1 exhibited significant cytotoxicity against cancer cell lines HCT-116 and MKN-45.

Zinc(II) tetra-(2-chloropropionylamido) phthalocyanine (TAPcCl) was synthesized as the initiator for atom transfer radical polymerization (ATRP). Using CuBr/tris(2-dimethylaminoethyl)amine as the catalyst system,ATRP of N-isopropylacrylamide (NIPAM) was performed to create a new star-shaped poly(N-isopropylacrylamide)(PNIPAM) with a zinc phthalocyanine core and PNIPAM arms (TAPc-PAM). The structures of the initiator and the polymers were characterized by means of Fourier transform infrared spectroscopy and proton nuclear magnetic resonance. The polydispersity index obtained by gel permeation chromatography indicated that the molecular weight distribution was narrow. The lower critical solution temperatures (LCST) for the TAPc-PAM aqueous solutions measured using the turbidimetry method were increased due to incorporation of the phthalocyanine core and decreased as molecular weight increased. TAPc-PAM possessed photocatalytic activity, a finding that was verified by Rhodamine B degradation in the presence of hydrogen peroxide under visible light. Moreover, the catalytic efficiency was higher at its LCST, which encouraged reuse of the photocatalyst.

To evaluate the behavior of the advanced unbonded pre-stressed concrete containment vessel (UPCCV) for one typical China nuclear power plant under Japan’s March 11 earthquake, five nonlinear time history analysis and a nonlinear static analysis of a 1:10 scale UPCCV structure have been carried out with MSC.MARC finite element program. Comparisons between the analytical and experimental results demonstrated that the developed finite element model can predict the earthquake behavior of the UPCCV with fair accuracy. The responses of the 1:10 scale UPCCV subjected to the 11 March 2011 Japan earthquakes recorded at the MYG003 station with the peak ground acceleration (PGA) of 781 gal and at the MYG013 station with the PGA of 982 gal were predicted by the dynamic analysis. Finally, a static analysis was performed to seek the ultimate load carrying capacity for the 1:10 scale UPCCV.

<P>Graphene is moving beyond the realm of simple electronic devices toward areas such as advanced biochemical sensing. The infrared (IR) response of graphene, characterized by collective long-lived charge-carrier oscillations, could be applied in IR-absorption spectroscopy, typically used for bio-chemical analysis. However, direct light absorption by propagating plasmons in graphene is forbidden due to the large momentum mismatch. Proposed methods to overcome this bottleneck come at a cost, the use of noble metal particles on graphene reduces the spectral bandwidth and nano-structuring graphene is expensive. Here, we propose a simple and cheap method to fabricate large scale ultra-sensitive graphene based mid-IR biosensors, by introducing dielectric beads to excite mid-IR range plasmons. Interference from waves scattered by the beads excite surface plasmon polaritons, which propagate several micrometers in graphene and enhance the interaction between the molecules and mid-IR light. The efficacy of Trichogramma against pests basically depends on the dispersal capacity of the wasps. A releaserecapture method was used in this study to evaluate the effect of relative wind speed (RWS) during the nighttime and the daytime on the dispersal capacity of postrelease population of Trichogramma dendrolimi Matsumura. Two repeated measurements were carried out, using sentinel cards with the host eggs and yellow sticky card traps on the third and the sixth day after wasp release. The results showed that: (1) both the number of recaptured wasps and parasitized eggs increased with the RWS in the daytime during the first three days. The number of parasitized eggs also increased with the RWS in the daytime over the latter three days. The distances that encompassed 98% recaptured T. dendrolimi adults (X 98 ) at the first three days was quadratically increased by the RWS of the daytime. (2) The number of wasps recaptured decreased with the distance from the releasing point during the first three days, but it was not influenced by the distance from the releasing point during the latter three days. (3) The average dispersal radius of the wasps during the first three days was shorter than during the latter three days. The present results will be helpful to improve release techniques based on the wind effects at different times.

Ultra performance liquid chromatography/quadrupole-time-of-flight mass spectrometry (UPLC/Q-TOF MS)technique combined with MetabolynxTM software was usedfor analysis of the metabolites of quercitrin by the isolatedhuman intestinal bacteria from the human feces. Four metabolitesof quercitrin were detected and tentatively identifiedbased on the characteristics of their protonated ions. Themetabolites were metabolized by four main metabolic pathwaysincluding hydroxylation, demethylation, deglycosylationand ring-cleavage. Quercitrin was metabolized to the hydroxyquercitrinand desmethylquercitrin by themajority of theisolated intestinal bacteria such as Bacteroides sp. 54, and wasdegraded to the deglycosylated product quercetin by rhamnosidaseand further ring-cleavage metabolite 3,4-dihydroxybenzoicacid by the minority of the isolated bacteria such asBacteroides sp. 45. The metabolic pathways and most of themetabolites of quercitrin were reported for the first time.
