      • Curcumol Induces Apoptosis in SPC-A-1 Human Lung Adenocarcinoma Cells and Displays Anti-neoplastic Effects in Tumor Bearing Mice

        Tang, Qi-Ling,Guo, Ji-Quan,Wang, Qi-You,Lin, Hai-Shu,Yang, Zhou-Ping,Peng, Tong,Pan, Xue-Diao,Liu, Bing,Wang, Su-Jun,Zang, Lin-Quan Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.6

Curcumol is a sesquiterpene originally isolated from curcuma rhizomes, a component of herbal remedies commonly used in oriental medicine. Its beneficial pharmacological activities have attract significant interest recently. In this study, anti-cancer activity of curcumol was examined with both in vitro and in vivo models. It was found that curcumol exhibited time- and concentration-dependent anti-proliferative effects in SPC-A-1 human lung adenocarcinoma cells with cell cycle arrest in the G0/G1 phase while apoptosis-induction was also confirmed with flow cytometry and morphological analyses. Interestingly, curcumol did not display growth inhibition in MRC-5 human embryonic lung fibroblasts, suggesting the anti-proliferative effects of curcumol were specific to cancer cells. Anti-neoplastic effects of curcumol were also confirmed in tumor bearing mice. Curcumol (60 mg/ kg daily) significantly reduced tumor size without causing notable toxicity. In conclusion, curcumol appears a favorable anti-cancer candidate for further development.

Dynamic changes of bacterial communities and nitrite character during northeastern Chinese sauerkraut fermentation

        Qi Zhou,Shizhu Zang,Zinan Zhao,Xinli Li 한국식품과학회 2018 Food Science and Biotechnology Vol.27 No.1

Northeastern Chinese sauerkraut is a wellknown traditional fermented vegetable in China. Incomplete identification of the microorganisms’ (bacteria in spontaneous fermentation) diversity and accumulation of nitrite make it difficult to normalize the fermentation process and product qualities of northeastern Chinese sauerkraut. Conventional culturing and polymerase chain reaction-denaturing gradient gel electrophoresis methods were combined to describe microbial structure and diversity. Lactobacillus, Leuconostoc, Enterobacter, Accumulibacter, Thermotoga, Pseudomonas, Clostridium, Rahnella and Citrobacter were predominant microorganisms in different fermentation periods. The pH value and nitrite concentration presented a certain relevance to the amount of lactic acid bacteria. Lactobacillus and Leuconostoc had the ability to decrease nitrite by inhibiting nitrate-reducing bacteria such as Enterobacter. Therefore, Northeastern Chinese sauerkraut should not be eaten until 4 weeks of fermentation for the safety and quality of fermented foods. Northeastern Chinese sauerkraut is rich in lactic acid bacteria, which demonstrate its ability as an excellent probiotic for applications in functional foods.

      • A Probabilistic Framework for Mapping Hazards of Coseimic Landslides

        ( Mingdong Zang ),( Shengwen Qi ) 대한지질공학회 2019 대한지질공학회 학술발표회논문집 Vol.2019 No.2

Coseismic landslides have been responsible for destroyed buildings and tens of thousands of deaths. Recent years, coseismic landslides occur frequently, and researches on coseismic landslides have become a global hot spot. Occurrences of earthquakes are random and difficult to predict accurately. Therefore, before an earthquake occurred, predicting the spatial distribution of coseismic landslides is one of key scientific issues needing to be solved in earthquake preventions. We have collected all of the data sets needed to conduct a regional probabilistic hazard analysis of coseismic slope instability, including: (1) 1:500000-scale geological map of the region, (2) extensive data on engineering properties of the geological units from geological surveying and field investigation, (3) historical seismic data of PGA from 1900 to 2018, and (4) high-resolution digital elevation models of the topography from ASTER. All of these data sets are digitized and rasterized at 30m grid spacing using ArcGIS and combined in a dynamic slope model based on Newmark permanent-deformation analysis modified by Barton model. The coseismic landslide displacements are estimated in each grid cell from a probabilistic framework through calculating a seismic hazard level of 10% probability of exceedance in 50 years, and the coseismic landslide hazard map is then generated based on the classification of the coseismic landslide displacement. The map can be applied to predict the hazard zone of the region and provide a reference for making decisions regarding infrastructure development and pre-earthquake construction.

        Zhu, Qiang,Zang, Qi,Jiang, Zhong-Min,Wang, Wei,Cao, Ming,Su, Gong-Zhang,Zhen, Tian-Chang,Zhang, Xiao-Tian,Sun, Ning-Bo,Zhao, Cheng Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.9

Objective: To explore the clinical application of recombinant human endostatin (Endostar) in the treatment of patients with non-small cell lung cancer (NSCLC) in Chinese mainland. Materials and Methods: A total of 75 patients diagnosed as NSCLC were randomly divided into control group (37 cases) and treatment group (38 cases). Control group was treated with postoperative complementary chemotherapy containing two-agent platinum protocol on postoperative d21, 3 weeks as a cycle, for totally 4~6 cycles. On this basis, treatment group was added with Endostar $7.5mg/m^2$ on postoperative d8~9, 3~4 h/time, qd, 14 weeks as a cycle, for totally 4 cycles. The interval between every two cycles was 7 d. The 5-year progression-free survival (PFS), 5-year survival time and complications in both groups were observed. Results: Compared with control group, the average PFS increased evidently in treatment group by 9.8 months (41.6 months vs. 31.8 months), and there was significant difference (P<0.05). And the median PFS was 42.5 months in treatment group, obviously longer than that in control group (33.7 months) by 8.8 months (P<0.05). Additionally, the 5-year overall survival rate (OS), average survival time and median survival time (MST) were 47.4%, 50.1 months and 59.3 months in treatment group, significantly higher than the 29.7%, 42.1 months and 43.5 months in control group (P<0.05). Only 1 patient showed poor healing of surgical wound in treatment group, but no surgery-associated complication was found in control group. Moreover, the postoperative complementary therapy-connected complication rates were 63.2% (24/38) and 59.5% (22/37) in treatment group and control group respectively, but there was no significant difference (P>0.05). Conclusions: The application of Endostar combined with sensitive platinum-contained chemotherapeutic agents in the postoperative complementary chemotherapy can be widely used in clinic because it can significantly prolong the long-term survival time of patients with NSCLC.

Analysis of the failure mechanism and support technology for the Dongtan deep coal roadway

        Chen, Miao,Yang, Sheng-Qi,Zhang, Yuan-Chao,Zang, Chuan-Wei Techno-Press 2016 Geomechanics & engineering Vol.11 No.3

The stability of deep coal roadways with large sections and thick top coal is a typical challenge in many coal mines in China. The innovative Universal Discrete Element Code (UDEC) trigon block is adopted to create a numerical model based on a case study at the Dongtan coal mine in China to better understand the failure mechanism and stability control mechanism of this kind of roadway. The failure process of an unsupported roadway is simulated, and the results suggest that the deformation of the roof is more serious than that of the sides and floor, especially in the center of the roof. The radial stress that is released is more intense than the tangential stress, while a large zone of relaxation appears around the roadway. The failure process begins from partial failure at roadway corners, and then propagates deeper into the roof and sides, finally resulting in large deformation in the roadway. A combined support system is proposed to support roadways based on an analysis of the simulation results. The numerical simulation and field monitoring suggest that the availability of this support method is feasible both in theory and practice, which can provide helpful references for research on the failure mechanisms and scientific support designing of engineering in deep coal mines.

      • Expression and Function of GSTA1 in Lung Cancer Cells

        Pan, Xue-Diao,Yang, Zhou-Ping,Tang, Qi-Ling,Peng, Tong,Zhang, Zheng-Bing,Zhou, Si-Gui,Wang, Gui-Xiang,He, Bing,Zang, Lin-Quan Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.20

Glutathione S-transferase A1 (GSTA1) appears to be primarily involved in detoxification processes, but possible roles in lung cancer remain unclear. The objective of this study was to investigate the expression and function of GSTA1 in lung cancer cells. Real-time PCR and Western blotting were performed to assess expression in cancer cell lines and the normal lung cells, then verify the A549 cells line with stable overexpression. Localization of GSTA1 proteins was assessed by cytoimmunofluorescence. Three double-strand DNA oligoRNAs (SiRNAs) were synthesized prior to being transfected into A549 cells with Lipofectamine 2000, and then the most efficient SiRNA was selected. Expression of the GSTA1 gene in the transfected cells was determined by real-time PCR and Western blotting. The viability of the transfected cells were assessed by MTT. Results showed that the mRNA and protein expression of A549 cancer cells was higher than in MRC-5 normal cells. Cytoimmunofluorescence demonstrated GSTA1 localization in the cell cytoplasm and/or membranes. Transfection into A549 cells demonstrated that down-regulated expression could inhibit cell viability. Our data indicated that GSTA1 expression may be a target molecule in early diagnosis and treatment of lung cancer.

A Fiber Spool’s Vibration Sensitivity Optimization Based on Orthogonal Experimental Design

        Jing Gao,Linbo Zhang,Dongdong Jiao,Guanjun Xu,Xue Deng,Qi Zang,Honglei Yang,Ruifang Dong,Tao Liu,Shougang Zhang 한국광학회 2024 Current Optics and Photonics Vol.8 No.1

A fiber spool with ultra-low vibration sensitivity has been demonstrated for the ultra-narrowlinewidth fiber-stabilized laser by the multi-object orthogonal experimental design method, which can achieve the optimization object and analysis of influence levels without extensive computation. According to a test of 4 levels and 4 factors, an L 16 (4 4 ) orthogonal table is established to design orthogonal experiments. The vibration sensitivities along the axial and radial directions and the normalized sums of the vibration sensitivities are determined as single objects and comprehensive objects, respectively. We adopt the range analysis of object values to obtain the influence levels of the four design parameters on the single objects and the comprehensive object. The optimal parameter combinations are determined by both methods of comprehensive balance and evaluation. Based on the corresponding fractional frequency stability of ultra-narrow-linewidth fiber-stabilized lasers, we obtain the final optimal parameter combination A3B1C2D1, which can achieve the fiber spool with vibration sensitivities of 10 −12 /g magnitude. This work is the first time to use an orthogonal experimental design method to optimize the vibration sensitivities of fiber spools, providing an approach to design the fiber spool with ultra-low vibration sensitivity.
