Studies on the impact of Fine dust (FD) on the skin as a causal factor of chronic skin disorders including skin barrier abnormalities, pruritus, and immunological dysregulations have recently received an increased interest. Effects of FD collected from Beijing, China on HaCaT keratinocytes were herein investigated. Based on dose and time response analysis, FD treatment-induced intracellular ROS generation and caused cell damage. Intracellular ROS were evaluated by fluorescence microscopic and FACS analysis. FD exposure-induced mitochondrial depolarization and apoptosis as evidenced by apoptotic body formation, and accumulation of sub-G1 hypodiploid cells. The events proceeded via mitochondria and ER stress-mediated apoptosis pathways. Further, FD elevated inflammatory responses as elicited by the activation of key mediators in the NF-κB pathway. Moreover, FD disrupted skin barrier functions involved in the formation of tight junctions and moisturization. A total of 90 one-day-old male broilers (Ross 308) were randomly assigned to one of the three dietary treatments, each consisting of six replicates (5 broilers/cage). The dietary treatments were 1) control (CON: fresh clean water with no supplement); 2) low dose [LD: CON + 1.56% extractions from the wooden chips (EWC)] and 3) high dose (HD: CON + 12.5% EWC). Drinking water supplemented with EWC was provided using specifically designed individual nipple drinker units. Average daily water intake (ADWI), average daily gain (ADG), average daily feed intake (ADFI), and feed conversion ratio (FCR) were measured weekly for 21 days. One broiler from each cage was euthanized for measuring the visceral organ weights and collecting ileal tissue samples for ileal architecture analysis on day 21. Broilers assigned to the LD and HD watery groups showed higher ADWI than that in broilers consumed CON on day 7 (P<0.05). The broilers subjected to HD treatment showed a deeper crypt depth (P<0.05) than that in broilers subjected to LD and CON on day 21. Therefore, broilers consumed HD showed a lower (P<0.05) villus height:crypt depth ratio than that broilers consumed CON on day 21. Broilers provided drinking water containing any of the tested concentrations of EWC showed no effect (P > 0.05) on growth performance, ileal villus height, and visceral organ weights as compared with those in the CON from hatch to 21 days. In conclusion, broilers fed HD showed reduction in villus height:crypt depth ratio without impairing growth performance and visceral organ weights for the experimental period.

Recent studies demonstrate that the dysregulated production of reactive oxygen species (ROS) and the resulting oxidative stress are implicated in the pathogenesis of numerous detrimental cellular responses caused by numerous factors including, xenobiotics, exposure to ionizing radiation, and disease conditions. At present, a special interest is paid towards research on antioxidants for the prevention of oxidative stress and associated health issues. Moringa oleifera is a well-known source of biocompatible antioxidants. The present study discloses the evaluation of the antioxidant activity of M. oleifera hot water extract (MOH) on H2O2 induced oxidative stress in Vero cells. Per analysis, MOH indicated DPPH and ABTS<sup>+</sup> radical scavenging with IC values of 102.52 and 122.55 µg/ml. MOH dose-dependently reduced intracellular ROS generation in H2O2 stimulated Vero cells while increasing the cell viability with the best bioactivity at 125 µg/ml. Further analysis using fluorescence microscopy and flowcytometry supported the above findings. H2O2-induced mitochondrial depolarization and apoptosis were dose-dependently suppressed by MOH. The suppression of apoptosis was mediated via the mitochondrial apoptosis pathway. Further analysis of MOH bioactivity and safety would potentiate its applications in manufacturing functional food.
