        Cytogenotoxic study of aqueous fruit extract of Morinda citrifolia in Wistar albino rats

        The use of herbal products and/or their derivatives as an alternative or complementary medicine becomes globally popular. To date, various biological activities have been reported in Morinda citrifolia Linn, especially antioxidants, antimicrobial, anti-infammatory and antitumor efects. This study aimed to investigate the genotoxicity and DNA repair capacity of the aqueous fruit extract of M. citrifolia (AEMC). Additionally, we checked the mutagenic capacity of AEMC by the comet assay and micronucleus tests in rats. For this, rats (n=10; 5 males and 5 females) were divided into fve groups as: negative control (distilled water); positive control (25 mg/kg of cyclophosphamide, i.p.); and three groups for AEMC (2.5, 5 and 10 mg/kg, i.p.). The results suggest that, the AEMC induced genotoxicity in white blood cells, but a DNA repair capacity was also observed after 72 h of AEMC exposure. AEMC also showed cytotoxicity and mutagenicity in liver and kidney cells of rats at 5 and 10 mg/kg doses. In conclusion, AEMC exhibited a dose-dependent cytotoxicity, genotoxicity and mutagenicity along with a time-dependent DNA repair capacity in rats. Further studies are necessary regarding the safe use of this medicinal plant for human consumption.

        A microfluidic device for partial cell separation and deformability assessment

        Blood flow in microcirculation shows several interesting phenomena that can be used to develop microfluidic devices for blood separation and analysis in continuous flow. In this study we present a novel continuous microfluidic device for partial extraction of red blood cells (RBCs) and subsequent measurement of RBC deformability. For this purpose, we use polydimethylsiloxane (PDMS) microchannels having different constrictions (25%, 50% and 75%) to investigate their effect on the cell-free layer (CFL)thickness and separation efficiency. By using a combination of image analysis techniques we are able to automatically measure the CFL width before and after an artificial constriction. The results suggest that the CFL width increases with enhancement of the constriction and contributes to partial cell separation. The subsequent measurements of RBCs deformation index reveal that the degree of deformation depends on the constriction geometries and hematocrit after the cell separation module. The proposed microfluidic device can be easily transformed into a simple, inexpensive and convenient clinical tool able to perform both RBC separation and deformability analysis in one single device. This would eliminate the need for external sample handling and thus reducing associated labor costs and potential human errors.

        Effectiveness and safety of rotary and reciprocating kinematics for retreatment of curved root canals: a systematic review of in vitro studies

        Objectives This systematic review (register-osf.io/wg7ba) compared the efficacy and safety of rotary and reciprocating kinematics in the removal of filling material from curved root canals. Materials and Methods Only in vitro studies evaluating both kinematics during retreatment were included. A systematic search (PubMed/MEDLINE, Scopus, and other databases, until January 2021), data extraction, and risk of bias analysis (Joanna Briggs Institute checklist) were performed. Efficacy in filling removal was the primary outcome. Results The search resulted in 2,795 studies, of which 15 were included. Efficacy was measured in terms of the remaining filling material and the time required for this. Nine studies evaluated filling material removal, of which 7 found no significant differences between rotary and reciprocating kinematics. Regarding the time for filling removal, 5 studies showed no difference between both kinematics, 2 studies showed faster results with rotary systems, and other 2 showed the opposite. No significant differences were found in apical transportation, centering ability, instrument failure, dentin removed and extruded debris. A low risk of bias was observed. Conclusions This review suggests that the choice of rotary or reciprocating kinematics does not influence the efficacy of filling removal from curved root canals. Further studies are needed to compare the kinematics safety in curved root canals.         Public Investment, Economic Performance and Budgetary Consolidation : VAR Evidence for the First 12 Euro Countries

        In a period of heightened concern about fiscal consolidation in the euro area, a politically expedient way of controlling the public budget is to cut public investment. A critical question, however, is whether or not political expediency comes at a cost, in terms of both long-term economic performance and future budgetary contention efforts. First, common wisdom suggests that public investments have positive effects on economic performance although the empirical evidence is less clear. Second, it is conceivable that public investment has such strong effects on output that over time it generates enough additional tax revenues to pay for itself. Obviously, it is equally plausible that the effects on output although positive are not strong enough for the public investment to pay for itself. In this paper, we investigate these issues empirically for the first twelve countries in the euro area using a vector auto-regressive approach. We conclude that the euro countries can be gathered in four groups according to the nature of the economic and budgetary impact of public investment. The first group includes Austria, Belgium, Luxembourg, and Netherlands, where the economic effects are either negative or positive but very small and, therefore, cuts will be harmless for the economy and effective from a budgetary perspective. The second group includes Finland, Portugal, and Spain, where public investment does not pay for itself and, therefore, cuts are an effective tool of budgetary consolidation although they are harmful for the economy. The third group includes France, Greece, and Ireland where public investment just pays for itself and therefore cuts are not an effective way of achieving long-term budgetary consolidation and are harmful for the economy. Finally, the fourth group includes Germany and Italy, where public investment more than pays for itself and, therefore, cuts are not only harmful for the economy but also counterproductive from a budgetary perspective.

        Applied element method simulation of experimental failure modes in RC shear walls

        With the continuous evolution of the numerical methods and the availability of advanced constitutive models, it became a common practice to use complex physical and geometrical nonlinear numerical analyses to estimate the structural behavior of reinforced concrete elements. Such simulations may yield the complete time history of the structural behavior, from the first moment the load is applied until the total collapse of the structure. However, the evolution of the cracking pattern in geometrical discontinuous zones of reinforced concrete elements and the associated failure modes are relatively complex phenomena and their numerical simulation is considerably challenging. The objective of the present paper is to assess the applicability of the Applied Element Method in simulating the development of distinct failure modes in reinforced concrete walls subjected to monotonic loading obtained in experimental tests. A pushover test was simulated numerically on three distinct RC shear walls, all presenting an opening that guarantee a geometrical discontinuity zone and, consequently, a relatively complex cracking pattern. The presence of different reinforcement solutions in each wall enables the assessment of the reliability of the computational model for distinct failure modes. Comparison with available experimental tests allows concluding on the advantages and the limitations of the Applied Element Method when used to estimate the behavior of reinforced concrete elements subjected to monotonic loading.

        Influence of mating and feeding on reproduction pattern of haematophagous bug Triatoma williami Galvão, Souza & Lima, 1965 (Hemiptera, Triatominae)

        The bloodsucking bug Triatoma williami is the main vector of Trypanosoma cruzi, the etiological agent of Chagas disease in State of Mato Grosso, Brazil. It occurs in sylvatic areas of Brazilian savanna with often records of adult specimens in domestic and peridomestic habitats. We describe, for the first time, the reproductive parameters of T. williami females such as fecundity, fertility and lifespan. We also evaluate whether mating and nourishment status of females of T. williami may modulate the reproductive parameters. As a result, the presence of male corroborated to increase female lifespan from 5.14 ± 1,77 to 34.57 ± 10,86 weeks. The oviposition rate in mated female increased 275% when we compared to the fecundity values in unmated females. Finally, our results indicate that mating is crucial to improve lifespan and fecundity of T. williami females. This characteristic may be related to the environmental conditions under T. williami develops, because the sylvatic ecotopes form small scattered colonies with infrequent access to blood sources.

        A review of hemorheology: Measuring techniques and recent advances

        Significant progress has been made over the years on the topic of hemorheology, not only in terms of the development of more accurate and sophisticated techniques, but also in terms of understanding the phenomena associated with blood components, their interactions and impact upon blood properties. The rheological properties of blood are strongly dependent on the interactions and mechanical properties of red blood cells, and a variation of these properties can bring further insight into the human health state and can be an important parameter in clinical diagnosis. In this article, we provide both a reference for hemorheological research and a resource regarding the fundamental concepts in hemorheology. This review is aimed at those starting in the field of hemodynamics, where blood rheology plays a significant role, but also at those in search of the most up-to-date findings (both qualitative and quantitative) in hemorheological measurements and novel techniques used in this context, including technical advances under more extreme conditions such as in large amplitude oscillatory shear flow or under extensional flow, which impose large deformations comparable to those found in the microcirculatory system and in diseased vessels. Given the impressive rate of increase in the available knowledge on blood flow, this review is also intended to identify areas where current knowledge is still incomplete, and which have the potential for new, exciting and useful research. We also discuss the most important parameters that can lead to an alteration of blood rheology, and which as a consequence can have a significant impact on the normal physiological behavior of blood.
