        The effect of mesenchymal stem cell use on intraabdominal adhesions in a rat model

        Purpose: Intra-abdominal adhesions (IAA) are among the most frequently seen pathologies in general surgery practice with an increased morbidity and mortality. In the present study, we investigated the effect of locally applied mesenchymal stem cells (MSCs) on IAA. Methods: Twenty-four Wistar Albino rats were used in the study. The rats were divided into three groups including: Sham, control, and MSCs group. On day 0, cecum was reached under anesthesia in all groups, except the Sham group. Scraping with a sponge was performed until petechial bleeding occurred. The control group received no treatment. In the stem cell group, MSCs were applied topically immediately after surgery on adhesions. The rats were sacrificed on day 10 and colon tissues and blood samples were collected for macroscopic, histopathological, and biochemical analysis. Results: In our study, E-selectin, P-selectin, TNF-α and IL-1 levels were statistically significantly lower in the MSC group than the control group, while the sham group has the lowest levels. In both the macroscopic and histopathological analyses (Zühlke’s scale), the least amount of adhesion was observed in the Sham group. In addition, although there was less adhesion in the MSC group than the control group, the difference did not reach statistical significance. Conclusion: Topical MSC application immediately after surgery suppresses the inflammatory process. However it was found to be ineffective in histopathological and macroscopic examinations performed on the 10th day.

        Effect of ankaferd blood stopper in experimental peritoneal adhesion model

        Purpose: Ankaferd Blood Stopper (ABS) is an herbal extract attained from 5 different plants. It has the therapeutic potential to be used for the management of external hemorrhage and controlling gastrointestinal bleedings. To date, the safety of ABS for intraperitoneal usage is not clear. In this study, we investigated the effectiveness and safety of using intraperitoneal ABS in an experimental peritoneal adhesion model. Methods: Twenty-four male Wistar Albino rats were used in the study. The rats were randomly divided into 3 groups: saline, ABS, and control. On the 10th day, all rats were euthanized. The adhesions were evaluated by Nair’s macroscopic adhesion classification, and pathologically evaluated with Zühlke’s microscopic adhesion classification. Results: A macroscopic and microscopic comparison between the ABS and saline groups did not show any differences but both the ABS and saline groups were superior when compared to the control group. Conclusion: ABS was found equally effective with saline on the abdominal adhesions and to no effect on postoperative adhesion formation.

        Effectiveness of thymoquinone, zeolite, and platelet-rich plasma in model of corrosive oesophagitis induced in rats

        Purpose: The effectiveness of platelet-rich plasma (PRP), thymoquinone, and zeolite in corrosive esophageal burns was investigated in a rat model. Methods: Four groups were comprised as containing 10 rats in each group. For group I, oesophagitis was induced and no other procedure was performed (control group). For group II, oesophagitis was induced and thymoquinone was administered for 1 week via oral gavage once a day (thymoquinone group). For group III, oesophagitis was induced for 1 week via oral gavage once a day (PRP group). For group IV, oesophagitis was induced and zeolite was administered for 1 week via oral gavage once a day (zeolite group). On the 10th day, the rats were sacrificed under anaesthesia and venous blood sampling was performed from the vena portae. The oesophaguses were totally excised. Biochemically, interleukin (IL)-1B, IL-6, TNF-α, and MCP-1 were examined from venous blood. Inflammation score was evaluated histopathologically in oesophageal tissue that was collected. Results: There was a statistically significant difference among groups in terms of IL-1, IL-6, MCP levels, compared to the control group; median IL-1, IL-6, MCP levels of thymoquinone, PRP, and zeolite groups were statistically significantly lower. There was a statistically significant difference among groups in terms of inflammation scores, compared to group I; median inflammation scores of groups II, III and IV were statistically significantly lower thymoquinone. Conclusion: PRP, and zeolite exhibited positive effect on recovery in oesophagitis by reducing inflammation in the involved segment.

        DNA Methylation Pattern of Gene Promoters of MB-COMT, DRD2, and NR3C1 in Turkish Patients Diagnosed with Schizophrenia

        Objective: We aim to evaluate the methylation status of membrane-bound catechol-O-methyltransferase (MB-COMT ) promotor, dopamine receptor D2 (DRD2 ), and nuclear receptor subfamily 3 group C member 1 (NR3C1 ) gene in patients with SCZ by comparing healthy controls. Methods: A sample of 110 patients with SCZ and 100 age- and sex-matched healthy volunteers was included in the study. The interview was started by filling out data forms that included sociodemographic and clinical information. The Structured Clinical Interview for DSM-IV Axis I Disorders was used to confirming the diagnosis according to DSM-IV-TR criteria. Then the patients were evaluated with the Positive and Negative Symptoms Scale in terms of symptom severity. Methylation-specific polymerase chain reaction was used to determine the methylation status of MB-COMT promotor, DRD2 , and NR3C1 gene from DNA material. Results: When we compared the percentages of MB-COMT promotor, DRD2 , and NR3C1 gene methylation status in SCZ patients with the healthy control group, the percentages of MB-COMT promotor (OR: 0.466; 95% CI: 0.268− 0.809; p = 0.006), DRD2 (OR: 0.439; 95% CI: 0.375−0.514; p < 0.001), and NR3C1 (OR: 0.003; 95% CI: 0.001− 0.011; p < 0.001) gene methylation status of SCZ was found to be significantly different from the control group. Whereas unmethylation of MB-COMT promotor and NR3C1 genes were associated with SCZ, the partial methylation of the DRD2 gene was related to the SCZ. Conclusion: The MB-COMT promotor, DRD2 , and NR3C1 gene methylation status may be associated with the SCZ in the Turkish population.

        Effect of shearing on some physiological and hormonal parameters in Akkaraman sheep

        Objective: Shearing is one of the practices that is applied periodically to fiber producing animals, which can also alter resistance of animals to high temperatures in especially summer months. This study aimed to investigate effects of shearing on some physiological and hormonal parameters in Akkaraman sheep during summer season. Methods: This study was carried out on 39 non-pregnant Akkaraman ewes (aged 1.5 years at the beginning of experiment). The 39 ewes were chosen randomly from the flock belonging to the Erciyes University and they were assigned to two groups as follows: i) group A (n = 20) designed as the control group, they were shorn and group B (n = 19) designed as the experimental group, they were unshorn. Prior to the shearing (-1 day) and on days 1, 7, 15, 30, 45, 60, 75, and 90 following the shearing, blood samples were taken from the vena jugularis of each sheep. Cortisol, β-endorphin, growth hormone (GH), thyroxine (T<sub>4</sub>), triiodothyronine (T<sub>3</sub>), and heat shock protein 70 (HSP-70) concentrations were determined using the enzyme immunoassay method. Body weight (BW), rectal temperature (RT), pulse rate (PR), and respiratory rate (RR) of each sheep were recorded at the same time. The data obtained were analyzed using two-way repeated measures analysis of variance. Results: Statistical analysis showed a significant effect of shearing×period interaction (p<0.01) and a significant effect of period (p<0.01) on BW, HSP-70, cortisol, T<sub>4</sub> and RT, PR, GH, β-endorphin, T<sub>3</sub>, respectively. Also these analysis showed no significant effect of shearing×period interaction or period on RR. Conclusion: The results showed that the thermoregulation abilities of sheep were affected by shearing treatment and the shorn ewes were less affected by heat stress. In conclusion, based on the data of this study, shearing can be considered as a necessary management practice that requires protection for sheep from the effect of heat stress.

        Frequency analysis of GPS data for structural health monitoring observations

        In this study, low- and high-frequency structure behaviors were identified and a systematic analysis procedure was proposed using noisy GPS data from a 165-m-high tower in ${\dot{I}}stanbul$, Turkey. The raw GPS data contained long- and short-periodic position changes and noisy signals at different frequencies. To extract the significant results from this complex dataset, the general structure and components of the GPS signal were modeled and analyzed in the time and frequency domains. Uncontrolled jumps and deviations involving the signal in the time domain were pre-filtered. Then, the signal was converted to the frequency domain after applying low- and high-pass filters, and the frequency and periodic component values were calculated. The spectrum of the tower motion obtained from the filtered GPS data had dominant peaks at a low frequency of $1.15572{\times}10-4Hz$ and a high frequency of 0.16624 Hz, consistent with two equivalent GPS datasets. Then, the signal was reconstructed using inverse Fourier transform with the dominant low frequency values to obtain filtered and interpretable clean signals. With the proposed sequence, processing of noisy data collected from the GPS receivers mounted very close to the structure is effective in revealing the basic behaviors and features of buildings.

        Usability of inclinometers as a complementary measurement tool in structural monitoring

        In the last few years, many structural monitoring studies have been performed using different techniques to measure structures of different scales such as buildings, dams or bridges. One of the mostly used tools are GPS instruments, which have been utilized in various combinations with accelerometers and some other conventional sensors. In the current study, observation series were recorded for 8 hours with GPS receivers (NovAtel) and Inclination Measurement Sensors mounted on a television tower in Istanbul, Turkey. Each series of observations collected from two different sensors were transformed into a single coordinate system (Local Topocentric Coordinates System). The positional changes of the tower were calculated from the GPS and the inclination data. These changes were plotted in two dimensions (2D) on the same graphic. Thus, the possibility of comparison and analysis were found using the data from both the GPS and the Inclinometer complement each other, in the real test area. The positional changes of the tower were modeled for further examination. As a result, the movement of the tower within an area of 1×1 cm2 was observed. Based on the results, it can be concluded that inclinometers can be used for monitoring the structural behavior of the tower.
