        Importance of Heat Shock Proteins in Maize

        Abiotic and biotic stress conditions cause extensive losses to maize production, mainly due to protein dysfunction in these condi-tions. In higher plants, the occurrence of heat-shock proteins (HSPs) in response to different environmental stresses is a universal phenomenon and has been well documented. Many studies have demonstrated that most HSPs are involved in many regulatory path-ways, act as molecular chaperones for other cell proteins, and have strong cytoprotective effects. Although many functional roles for HSPs are known, the mechanisms for these multiple functions are not entirely understood. Here we reviewed the correlation among HSP genes/proteins and plant tolerance, especially maize, in different environmental stresses. Due to the low availability of informa-tion regarding the expression of HSP genes in response to different stresses in maize, we decided to mine databases in order to gener-ate ew insights related to this topic.

        Differential Expression of Genes Associated with Endomembrane Transport and Cell Wall Metabolism during Ripening of ‘Chimarrita’ and ‘Granada’ Peach

        Expression of genes involved in endomembrane transport and cell wall metabolism, particularly in synthesis and degradation of cell wall macromolecules were compared during the ripening of a white melting flesh cultivar, Chimarrita and a yellow non-melting flesh cultivar, Granada in order to evaluate their association with fruit softening. In both, most genes were up-regulated as ripening evolved. In ‘Chimarrita’, overall higher transcript accumulation occurred at 95 days after anthesis, while in ‘Granada’ higher relative transcription was detected at 115 days after anthesis. Although transcript accumulation was correlated with fruit softening, the process does not seem to be totally dependent on the set of studied genes, since the peak of RNA abundance occurred when flesh firmness had already decreased.

      • Study of Counter Diffusion in Isostatic Permeameters

        The counter-diffusion of two gaseous substances permeating a polymeric membrane has been investigated both experimentally and theoretically. The aim of the study was to find mutual effects, if any, that could influence the permeability and diffusivity data. The experimental data were obtained with an isostatic permeameter operating at ambient pressure and 303 K: helium, nitrogen, carbon dioxide methane were used as permeating gas at different partial pressure; helium or nitrogen as equilibrating or carrier gas. No evident mutual effect of the counter-diffusing gas was observed. The theoretical analysis gave some insight into the phenomena and it was concluded that at near-atmospheric pressures, and in the absence of swelling phenomena no mutual interaction exists. On a theoretical basis any mutual interaction between diffusing and counter-diffusing gases could only occur: i) at high pressures , when the free movement of permeating gas molecules within the polymer is hindered by the counter-diffusing gas; ii) when a large part of the free volume fraction is occupied by the counter--diffusing gas; iii) swelling phenomena modify the structure and free volume fraction of the polymer.

        free volume in polymers is defined as the difference of the specific volume and the volume which is not available for the particular molecular motion which is responsible or the process that is considered . Relations between free volume and viscosity free volume and diffusion coefficient are pre-sented both in the case of simple low molecular weight liquids and in the case of polymers. Molecular models and free volume models are reminded starting from the equilibrium state equation of Simha and Somcynski. The non equilibrium situations of specific volume of glass polymers below Tg are shown introducing different relaxation volume equations which involve different material's parameters and con-cept of the fictitious temperature. The diffusivity equations of Vrentas and Duda are introduced both for the glassy and rubbery states. The possibility of introducing time relaxation functions is also suggested. The importance of finding experimental evidences of the free volume is stressed. highlights of the free volume measurement methods are given in particular as to dilatometry photocromy fluorescence electron spin resonance small angle X-ray scattering positron annihilation spectroscopy.

      • Direct Observation of Wet Biological Samples by Graphene Liquid Cell Transmission Electron Microscopy

        <P>Recent development of liquid phase transmission electron microscopy (TEM) enables the study of specimens in wet ambient conditions within a liquid cell; however, direct structural observation of biological samples in their native solution using TEM is challenging since low-mass biomaterials embedded in a thick liquid layer of the host cell demonstrate low contrast. Furthermore, the integrity of delicate wet samples is easily compromised during typical sample preparation and TEM imaging. To overcome these limitations, we introduce a graphene liquid cell (GLC) using multilayer graphene sheets to reliably encapsulate and preserve biological samples in a liquid for TEM observation. We achieve nanometer scale spatial resolution with high contrast using low-dose TEM at room temperature, and we use the GLC to directly observe the structure of influenza viruses in their native buffer solution at room temperature. The GLC is further extended to investigate whole cells in wet conditions using TEM.         Safety Culture: A Retrospective Analysis of Occupational Health and Safety Mining Reports

        Background: In the mining industry, various methods of accident analysis have utilized official accident investigations to try and establish broader causation mechanisms. An emerging area of interest is identifying the extent to which cultural influences, such as safety culture, are acting as drivers in the reoccurrence of accidents. Thus, the overall objective of this study was to analyze occupational health and safety (OHS) reports in mining to investigate if/how safety culture has historically been framed in the mining industry, as it relates to accident causation. Methods: Using a computer-assisted qualitative data analysis software, 34 definitions of safety culture were analyzed to highlight key terms. Based on word count and contextual relevance, 26 key terms were captured. Ten OHS reports were then analyzed via an inductive thematic analysis, using the key terms. This analysis provided a concept map representing the 50-year data set and facilitated the use of text framing to highlight safety culture in the selected OHS mining reports. Results: Overall, 954 references and six themes, safety culture, attitude, competence, belief, patterns, and norms, were identified in the data set. Of the 26 key terms originally identified, 24 of them were captured within the text. The results made evident two distinct frames in which to interpret the data: the role of the individual and the role of the organization, in safety culture. Conclusion: Unless efforts are made to understand and alter cultural drivers and share these findings within and across industries, the same accidents are likely to continue to occur.

        Genetic variability and heritability of agronomic traits in a wheat collection used in southern Brazil

        Wheat is a staple food for a large part of the world's population and faces a continuous demand for increased productivity. The presence of genetic variability allows the success in developing cultivars with higher yield. The understanding of the genetic variability available is crucial for their efective use in breeding programs. Inheritance of wheat yield components is complex, due to polygenic control and the strong infuence of the environment. Therefore, an accurate heritability estimation can accelerate the selection gains towards increasing wheat yield. Thus, this study aimed to characterize agronomic traits related to yield in a collection of 99 wheat accessions used in Brazil. In addition, the heritability of these traits was estimated. The studied accessions showed variability for yield, which can be explained by the presence of variability in most yield components. Cluster analysis showed that there is variability in wheat genotypes, however, the introduction of new sources of variability in crossing blocks or the induction of mutations is suggested. The broad-sense heritability for grain yield was considered high, as well as for most of the studied agronomic traits, suggesting that it is possible to obtain genetic gain with selection in this environment. However, for greater accuracy, genetic gain values should be estimated and considered when designing selection strategies. Hectoliter weight showed low heritability, demonstrating the complexity of selection for this trait. In summary, this study demonstrates the presence of genetic variability and high heritability for agronomic traits, enabling genetic gain in breeding programs using these wheat accessions.
