      • Comparison of impact of insomnia on depression and quality of life in restless legs syndrome/Willis-Ekbom disease and primary insomnia patients

        Background: Although insomnia is common among people with restless legs syndrome (RLS), its impact on the daily suffering of those with RLS remains unclear. This study aimed to compare the differential impact of clinical insomnia on depression and quality of life (QoL) among people with RLS, primary insomnia, and healthy controls. Methods: A total of 148 people with RLS, 115 with primary insomnia, and 117 healthy controls were enrolled into this cross-sectional study. Participants completed sleep, depression, and QoL questionnaires. Clinical insomnia was defined as Korean version of the Insomnia Severity Index (K-ISI)@?≥@?15. Correlation coefficients between sleep measures and both depression and QoL were calculated. Multivariate regression was used to identify the clinical factors that were most closely associated with depression and QoL among people with RLS and primary insomnia. Results: Participants with RLS had insomnia and sleep quality at intermediate levels between the healthy controls and primary insomnia subjects, but those with clinical insomnia had equivalent depression and QoL scores regardless of RLS diagnosis. Insomnia severity correlated with depression and QoL in RLS and primary insomnia. Multivariate regression, however, revealed that RLS severity was the most overall predictive factor for depression and QoL among those with RLS. Insomnia severity was the strongest predictor in primary insomnia. Conclusion: Insomnia was more closely associated with depression and QoL among people with primary insomnia than those with RLS, but clinical insomnia may have a significant impact in RLS as well. Future RLS studies should account for sleep quality in addition to RLS symptom severity when investigating mood and QoL.

        Munchausen Stridor–A Strong False Alarm of Anaphylaxis

        The diagnosis of anaphylaxis is often based on reported symptoms which may not be accurate and lead to major psychosocial and financial impacts. We describe two adult patients who were diagnosed as having recurrent anaphylaxis witnessed by multiple physicians based on recurrent laryngeal symptoms. The claimed cause was foods in one and drugs in the other. We questioned the diagnosis because of absent documentation of objective findings to support anaphylaxis, and the symptoms occurred during skin testing though the test sites were not reactive. Our initial skin testing with placebos reproduced the symptoms without objective findings. Subsequent skin tests with the suspected allergens were negative yet reproduced the symptoms without objective findings. Disclosing the test results markedly displeased one patient but reassured the other who subsequently tolerated the suspected allergen. In conclusion, these 2 patients’ symptoms and evaluation were not supportive of their initial diagnosis of recurrent anaphylaxis. The compatible diagnosis was Munchausen stridor which requires psychiatric evaluation and behavior modification, but often rejected by patients.

        Water Sorption/Desorption Characteristics of Eutectic LiCl-KCl Salt-Occluded Zeolites

        Molten salt consisting primarily of eutectic LiCl-KCl is currently being used in electrorefiners in the Fuel Conditioning Facility at Idaho National Laboratory. Options are currently being evaluated for storing this salt outside of the argon atmosphere hot cell. The hygroscopic nature of eutectic LiCl-KCl makes is susceptible to deliquescence in air followed by extreme corrosion of metallic cannisters. In this study, the effect of occluding the salt into a zeolite on water sorption/desorption was tested. Two zeolites were investigated: Na-Y and zeolite 4A. Na-Y was ineffective at occluding a high percentage of the salt at either 10 or 20wt% loading. Zeolite-4A was effective at occluding the salt with high efficiency at both loading levels. Weight gain in salt occluded zeolite-4A (SOZ) from water sorption at 20% relative humidity and 40℃ was 17wt% for 10% SOZ and 10wt% for 20% SOZ. In both cases, neither deliquescence nor corrosion occurred over a period of 31 days. After hydration, most of the water could be driven off by heating the hydrated salt occluded zeolite to 530℃. However, some HCl forms during dehydration due to salt hydrolysis. Over a wide range of temperatures (320–700℃) and ramp rates (5, 10, and 20℃ min−1), HCl formation was no more than 0.6% of the Cl− in the original salt.

        EFFECT OF EXERCISE ON MILK YIELD, MILK COMPOSITION AND BLOOD METABOLITE CONCENTRATIONS IN HEREFORD × FRIESIAN CATTLE

        Three experiments were carried out in which lactating Hereford ${\times}$ Friesian cattle walked up to ten kilometers a day for three periods of five days with two non-walking days between each walking period and in which the animals were fed different diets. Measurements were made of milk yield, milk constituent yields and concentrations and blood metabolite concentrations. Exercise caused significant reductions in milk yield and in the yields of lactose and milk protein. Milk fat yield was not reduced when animals were exercised. During exercise the concentrations of ${\beta}-OH$ butyrate and free fatty acids increased, whereas the concentrations of glucose, magnesium and inorganic phosphorus decreased. Diet influenced the effect of exercise on some blood metabolite concentrations.

        The Helix-Loop-Helix Factors Id3 and E47 Are Novel Regulators of Adiponectin

        <P>Adiponectin is an adipocyte-derived cytokine with beneficial effects on insulin sensitivity and the development of atherosclerosis. Id3 is a helix-loop-helix factor that binds to E-proteins such as E47 and inhibits their binding to DNA. Although the helix-loop-helix factor sterol regulatory element binding protein (SREBP)-1c is a known activator of adiponectin transcription, this study provides the first evidence of a role for Id3 and E47 in adiponectin expression. Decreased Id3 in differentiating adipocytes correlates with increased adiponectin expression and forced expression of Id3 inhibits adiponectin expression. Moreover, Id3-null mice have increased adiponectin expression in visceral fat tissue and in serum. We demonstrate that E47 potentiates SREBP-1c-mediated adiponectin promoter activation and that Id3 can dose-dependently inhibit this action via interaction with E47. Mutation of a consensus E47 binding site results in nearly complete loss of promoter activation. Furthermore, we demonstrate E47 binding to the endogenous adiponectin promoter both in vitro and in vivo by chromatin immunoprecipitation analysis. Binding is not detected in undifferentiated cells which express Id3 but peaks during differentiation in parallel with Id3 decline. This promoter binding can be completely abolished by the overexpression of Id3 and is enhanced in adipose tissue null for Id3. These data establish Id3 and E47 as novel regulators of SREBP-1c-mediated adiponectin expression in differentiating adipocytes and provide evidence that Id3 regulates adiponectin expression in vivo.</P>

        On-chip capacitive sensing and tilting motion estimation of a micro-stage for <i>in situ</i> MEMS gyroscope calibration

        <P><B>Abstract</B></P> <P>In this paper, we introduce a novel on-chip capacitive sensor design and a motion estimation method for measuring multi-axis out-of-plane tilting motion of a micro-stage that is used for <I>in situ</I> calibration of an integrated MEMS gyroscope. On-chip sensing and motion estimation is critical for this application, since precision in measuring high frequency tilting motion of the micro-stage determines how accurately an on-stage MEMS gyroscope can be calibrated in the field with respect to this motion. To obtain decoupled sensing signals from multi-axis out-of-plane tilting motion, a capacitive sensing layout with high off-axis signal rejection capability is introduced. With the proposed design specifications and range of tilting motion, a dual axis capacitive sensor model is developed, and simulation results show that the sensor is able to reject more than 99.90% of the off-axis signal. The sensor also permits detection of perturbations to stage geometry due to ambient temperature variation through the effects of nonlinear behavior on signal timing. To measure tilting motion with the obtained sensing signal, a motion estimator with extended Kalman Filter (EKF) is also developed. With assistance of a laser Doppler vibrometer (LDV), experiments are performed to identify the parameters of the motion estimator and validate its performance. The experimental results demonstrate a 286 ppm error in scale factor calibration of a commercial MEMS gyroscope by the micro-stage with moderate temperature variation.</P>

      • Integration of Genome-wide Approaches Identifies lncRNAs of Adult Neural Stem Cells and Their Progeny In Vivo

        Long noncoding RNAs (lncRNAs) have been described in cell lines and various whole tissues, but lncRNA analysis of development in vivo is limited. Here, we comprehensively analyze lncRNA expression for the adult mouse subventricular zone neural stem cell lineage. We utilize complementary genome-wide techniques including RNA-seq, RNA CaptureSeq, and ChIP-seq to associate specific lncRNAs with neural cell types, developmental processes, and human disease states. By integrating data from chromatin state maps, custom microarrays, and FACS purification of the subventricular zone lineage, we stringently identify lncRNAs with potential roles in adult neurogenesis. shRNA-mediated knockdown of two such lncRNAs, Six3os and Dlx1as, indicate roles for lncRNAs in the glial-neuronal lineage specification of multipotent adult stem cells. Our data and workflow thus provide a uniquely coherent in vivo lncRNA analysis and form the foundation of a user-friendly online resource for the study of lncRNAs in development and disease.
