      • Agrobacterium tumefaciens의 Ti plasmid 재조합에 관한 연구

        Crown gall의 병원체로 알려진 Agrobacterium tumefaciens가 함유하고 있는 Tumor inducing(Ti) plasmid가 식물 hormone과 opine의 합성원인이라는 것이 밝혀지고 식물 세포의 crown gall에는 Ti plasmid가 존재하며 Ti plasmid의 일부가 식물세포의 핵 genome에 삽입되어 형질 변화가 일어남이 보고 되었고 Ti plasmid의 식물체로의 gene도입 vector로서의 연구가 최근 활발히 진행되고 있으며 현재 PLGV vector와 PMON vector 등이 개발되고 있으나 조작이 복잡하고 아직은 초보적 단계인 것으로 알려져 있다. 본 연구에서는 원하는 식물 gene을 식물체에 도입하는 유용한 vector개발의 초보단계로서 Ti plasmid의 T-DNA의 일부인 nos gene을 분리하여 pBR322와의 재조합을 기도한 것이다. pBR 322를 포함하는 E. coli로 부터 Horwitz(1979)의 방법으로 pBR 322를 분리하였고 A. tumefaciens C58에서 Kado(1979)의 방법으로 Ti plasmid를 분리한 후 Ti plasmid 내의 nos gene을 분리하였다. 분리된 pBR 322를 Hind Ⅲ로 절단한 후 CIP로 처리하고 T_4 DNA ligase를 이용하여 Hind Ⅲ로 처리한 nos gene fragment와 재조합하였다. 이와같이 하여 얻은 recombinant DNA로 인한 E. coli HB101의 형질 변화를 Mandel과 Higa의 방법에 따라 조사한 결과 pBR 322의 tetracyclin gene 사이에 nos gene이 삽입된 것으로 확인되었다. 즉 ampicillin 배지에서는 92개의 colony가 형성되었는데 그중 59개는 tetracyclin 배지에서 colony를 형성하지 못하였다. 이러한 tetracyclin 감수성이면서 ampicillin 저항성인 colony를 배양하여 얻은 recombinant plasmid를 전기이동법으로 분리한 결과 예상대로 크기가 7.5kb 정도였고 Hind Ⅲ로 절단하였을 때 2개의 band가 확인되었다. 본 연구에서 얻은 nos gene-pBR 322 hybrid plasmid는 원하는 식물 gene을 이것에 삽입하여 A. tumefaciens 내에서 homologous recombination을 행한 후 식물체에 감염시켜 원하는 gene의 발현여부를 검출하는데 이용될 것으로 기대된다. It is mow realized that the tumor-inducing(Ti) plasmid in Agrobacterium tumefaciens is responsible for the induction of crown gall tumors in dicotyledonous plants and it has been demonstrated that a segment of the Ti plasmid, so called T-DNA, is stably integrated into and expressed in the genome of transformed plant cells. Recently, the use of A. tumefaciens Ti plasmid as a vector to introduce a foreign gene has been intensively studied and several vectors such as PLGV vector and PMON vector has been reported. However, their practical use is still at a primitive stage. It was attempted in the present study, therefore, to conduct the recombination of nos gene of T-DNA from A. tumefaciens Ti plasmid and E. coli pBR 322 as a vector it introduce foreign plant gene into dicotyledon plant cells. pBR 322 was isolated from E. coli containing pBR 322 accdrding to Horowitz(1979) and nos gene of T-DNA in Ti plasmid from A. tumefaciens C58 was obtained according to Kado(1979). The isolated pBR 322 was cut using Hind Ⅲ followed by the addition of CIP·pBR 322 fragments and Hind Ⅲ treated nos gene fragments were than recombined using T_4 DNA ligase and the E. coli cells were then transformed by recombinant DNA according to Mandel and Higa(1970). It was confirmed that nos gene was inserted into tetracyclin gene of pBR 322 by the finding that when the transformed cells were incubated in the ampicillin medium, 92 colonies were formed, in which 59 were tetracyclin sensitive. The above tetracyclin sensitive but ampicillin resistant cells were then cultured and the recombinant pBR-nos gene was isolated and subjected to electrophoresis. It was found that size of the recombinant DNA was approximately 7.5 Kb as expected and two bands were appeared on electrophoretogram when the recombinant DNA was treated with Hind Ⅲ. It might be possible to insert the foreign plant genes to the pBR-nos gene hybrid plasmid obtained in the present study and the resultant DNA could be then homolgously recombined with Ti plasmid of A. tumefaciens and the final recombined Ti plasmid could be used for the introduction of the foreign plant genes into dicotyledon plant cells.

        중생 고품질 내병 다수성 벼 ‘영보(嶺寶)’

        ‘Yeongbo’ is a variety of japonica rice (Oryza sativa L.) with good eating quality, resistance to rice stripe virus, rice dwarf virus, and bacterial blight disease. It was developed by the rice breeding team of the Yeongdeok Branch, National Institute Crop Science, Rural Development Administration in 2014. This variety was derived from a cross between ‘Yeongdeog35’ with good eating quality and ‘Saekyehwa’ with good plant type in the 2002 summer season. A promising line selected through the pedigree breeding method, YR24269-1-2-1-2-3-3, was designated as ‘Yeongdeog57’ in 2011. After a local adaptability test at nine locations from 2012 to 2014, ‘Yeongdeog57’ was released under the name of ‘Yeongbo’ in 2014. ‘Yeongbo’ had a short culm length of 66 cm and medium growth duration. This variety was resistant to K1, K2, and K3 races of bacterial blight, rice stripe virus, and dwarf virus, and moderately resistant to leaf blast disease. ‘Yeongbo’ had translucent and clear milled rice kernel without white core and belly, and good eating quality according to a panel test. The yield potential of ‘Yeongbo’ in milled rice was approximately 5.75 MT/ha at an ordinary fertilizer level in the local adaptability test. This cultivar would be adaptable to middle plain area, south plain area, Yeongnam plain area, east costal area, and south mid-mountainous area. (Registration No.6804)

        K-IFRS 적용 확정급여형 퇴직급여 회계정보의 가치관련성

        본 연구에서는 K-IFRS 제1019호의 도입 이후 확정급여형 퇴직연금제도를 채택한 기업을 대상으로 퇴직연금요소의 가치관련성에 대해 조사하였다. 이를 위하여 K-IFRS를 전면 도입한 2011년부터 2013년까지 3년간 확정급여형 퇴직연금제도를 채택한 상장기업 중 제조업을 대상으로 기업-연도 관찰치를 수집하였다. 분석 결과, 퇴직급여와 관련된 회계정보는 가치관련성을 가짐을 확인하였다. 구체적으로 살펴보면 퇴직연금의 적립수준을 나타내는 순확정급여부채는 주가와 유의한 음의 관계를 보였으며, 순확정급여부채의 구성요소인 확정급여채무와 사외적립자산은 각각 주가와 유의한 음과 양의 관계를 가짐을 확인하였다. 또한 주석에 공시되는 퇴직급여원가의 주요구성항목들도 검증한 결과, 주가와 가치관련성을 가지나 일부 항목은 일관된 결과가 보이지 않았다. 추가분석에서는 이들 퇴직급여원가의 구성 항목들이 주가와 비선형관계를 가질 가능성이 있음을 확인하였다. 본 연구는 국내 연구에서 거의 다루지 않은 퇴직급여와 관련된 회계정보의 가치관련성에 대해 체계적으로 검증하였다는 점에서 공헌점을 갖는다. 또한 퇴직급여와 관련하여 주석에 공시되는 세부정보에 대한 가치관련성을 검증함으로써 퇴직급여와 관련하여 주석에 공시되는 정보들도 정보효과를 가짐을 확인하였다. In this paper, we examine value relevance of accounting numbers pertinent to post-employment benefits under K-IFRS No.1019. K-IFRS No.1019 requires companies to use the projected unit credit method to estimate pension costs and pension liabilities, and to disclose separately several components of pension costs. In order to find out whether pension accounting numbers are valued by the stock market, we use a sample of KOSPI-listed manufacturing firms with defined benefit pension plan from 2011 to 2013. The major findings are : (1) We find, as expected, that pension liability(the present value of the defined benefit obligation) and pension assets(plan assets) are value relevant. (2) Pension expenses(defined benefit cost) are value-relevant, that is, estimation for the regression coefficient turns out to be significantly negative. Additionally we disaggregate the pension expenses into current service cost, interest expense and expected return on plan assets and so on. Contrary to expectations, the coefficients on interest expense and expected return on plan assets are significantly positive and negative respectively. This puzzling result, that has also turned up in additional analysis, could be attributable to nonlinear relationship between the pension cost components and stock price. Another possibility is that information users may discount or ignore information disclosed on the footnote. Overall, the results suggest that information on the post-retirement benefits is value relevant. This study highlights the importance of considering information on post-employment benefits in the valuation.

        MAS를 이용한 줄무늬잎마름병 저항성 조생종 벼 ‘해담쌀’ 개발

        ‘Haedamssal’ is an early maturing and rice stripe virus disease-resistant cultivar adaptable for early-transplanting cultivation that was developed by the rice breeding team of the Department of Southern Crop, NICS, RDA, in 2014. This cultivar was derived from the cross YR25869 (YR21247-B-B-B-49-1/Sasanishiki BL4//Koshihikari) and YR25868 (Unkwang//YR21247-B-B-B-49-1/Sasanishiki BL4) made in the 2005/2006 winter season and was advanced to the F5 generation by a bulk breeding method using rapid generation advance. To incorporate rice stripe virus resistance, marker-assisted selection on the RSV gene was conducted in 3-way and 6-way cross F1 generation using the tightly linked marker RM6897. From testing in the replicated yield trial in 2011, a promising line YR26258-B-B-B-33-3 was selected and it was designated as ‘Milyang276’. A local adaptability test of ‘Milyang276’ was performed at three locations from 2012 to 2014 and it was named as ‘Haedamssal’, which was a good eating quality variety. The culm length was 67 cm in yield trials, which was 4 cm shorter than ‘Jopyeong’. The number of spikelets per panicle was lower than ‘Jopyeong’, whereas the number of tillers per hill was higher. This variety was resistant to RSV disease, bacterial blight, and leaf blast disease. The milled rice yield of ‘Haedamssal’ was 5.48 MT per ha at the early transplanting in the local adaptability test. ‘Haedamssal’ is well adapted to early transplanting cultivation in the southern plain area (Registration No. 6811).

        연구논문 : 이중 변화형태 모형을 적용한 초기 청소년의 집단따돌림 가해경험과 피해경험의 분류 및 예측

        본 연구의 목적은 초기 청소년의 집단따돌림 가해경험과 피해경험의 변화형태에 따른 잠재계층을 동시에 추정하고 두 변수에 영향을 미치는 영향요인들과의 관련성을 밝히는 것이다. 이를 위해 한국청소년패널 초4패널 1차년도부터 5차년도까지의 종단 자료를 이용하였다. 가해경험과 피해경험의 잠재계층을 동시에 도출하기 위해 준모수적 집단중심 접근방법의 확장모형인 이중 변화형태 모형을 적용하였다. 분석 결과, 집단따돌림 가해경험과 피해경험 각각 2개의 잠재계층들로 분류되었고, 변화형태에 따라 가해 저수준 집단과 가해 중간감소 집단, 피해 무경험 집단과 피해 지속 집단으로 명명하였다. 영향요인들의 영향력을 검증한 결과, 가해경험 잠재계층에는 성별, 공격성, 친구의 폭력이, 피해경험 잠재계층에는 성별, 우울 및 불안, 공격성, 부모와 친구의 폭력, 부모와 친구로부터 오는 스트레스가 유의한 영향을 미치는 것으로 나타났다. 본 연구는 실제적인 측면에서는 집단따돌림을 경험하는 초기 청소년들을 위한 예방 대책을 마련하는 데 도움을 줄 것이며, 연구방법론적인 측면에서는 국내에서 거의 사용되지 않은 이중 변화형태 모형을 적용한 연구라는 점에서 의의가 있다. The purpose of this study is to identify the latent classes depending on trajectories of early adolescents` bullying and victimized experiences and to test the effects of eco-system variables on determining the latent classes. For these research goals, we used the Korean Youth Panel Study, which is 5-year longitudinal data from 4th graders of elementary school to 2nd graders of middle school. To estimate the latent classes in bullying and victimized experience simultaneously, we applied dual trajectory model by extending semi-parametric group-based approach. Results showed that bullying and victimized experience, respectively, have two latent classes. The latent classes were defined as low-level bullying group, midium-decreasing bullying group, no-victimized group, and continuous victimized group according to their trajectory patterns. Gender, aggression, and friends` violence have a significant effect on both the bullying and victimized latent classes. However, depression/anxiety, parents` violence, and stress from parents or friends affected siginificantly the victimized latent classes only. This study can be useful for developing prevention programs for adolescents who experience bullying and victimization. The present study also contributed methodologically by applying an advanced, novel statistical model, dual trajectory model, to substantive issues.

        In this paper, Coplanar Waveguide(CPW) low-pass filter with the spurious-reduced characteristic is designed by using Defected Ground Structures(DGS). Two different LPF unit cells and DGS Pattern are Combined to reduce the spurious. In comparison to a conventional step impedance LPF, the size of proposed LPF as 17.2 mm × 6.8 mm is serveral times smaller. The simulation and measurements confirm that spurious characteristics is presented less than -30dB(S21) at the three times range of cut off frequency.

        CFRP 복합적층판의 적층배향·계면수에 따른 저속충격특성

        In this paper, this study aims at the evaluation on the characteristics of CFRP laminate plates using a falling weight impact tester. The experiment was conducted on several laminates of different orientation. A system was built for measuring the impact strength of CFRP laminates in consideration of stress wave propagation theory using a falling weight impact tester. Delamination areas of impacted specimens for the different ply orientation were measured with ultrasonic C-scanner to find correlation between impact energy and delamination area. Absorbed energy of quasi-isotropic specimen having four interfaces was higher than that of orthotropic laminates with two interfaces. The more interfaces, the greater the energy absorbed. The absorbed energy of a hybrid specimen containing a GFRP layer was higher than that of normal specimens.

        A biological assessment of water quality was carried out in Giran stream from May 203 to January 2004. The benthic macroinvertebrates collected seasonally from 5 field sites were identified mostly up to species level. The benthic macroinvertebrates collected from the surveyed sites were composed of 88 species, 68 genera,42 families, 14 orders, 5 class and 4 phyla. The dominance index (DI) showed the high-est as 0.802 at site 5 and the lowest as 1.643 at site 3. Ecological score of benthic macroinvertebrate (ESB) showed the highest as 103 at site 2 and the lowest as 63 at site 3. All surveyed sites were Oligosaprobic that the water quality estimated by ecological score of benthic macroinvertebratc community.

      • 균 spheroplast 및 large unilamellar vesicle의 식물 protoplast로의 도입

        A. tumefaciens의 식물 protoplast로의 도입을 확인하는 방법으로 방사성 인산(Na_2H^32PO_4)을 함유한 0.8% nutrient broth에서 증식한 Agrobacterium tumefaciens C58를 사용하여 방사성 spheroplast와 방사성 Ti plasmid encapsulated liposome을 만들어 Danus carota protoplast로의 도입을 시도한 결과 다음과 같은 실험 성적을 얻었다. 1. A. tumefaciens에서 추출한 Ti plasmid를 Reverse Phase Evaporation 법으로 phosphatidyl-choline, stearylamine과 함께 Ti plasmid encapsulated liposome을 제조한 결과 17%의 Ti plasmid가 liposome안으로 편입되었다. 2. 방사성 Ti plasmid encapsulated liposome과 Dacus carota root protoplast를 polyethyleneglycol-Ca^2+법으로 처리한 결과 introduction frequency는 38%이었다. 3. A. tumefaciens C58의 spheroplast와 Dacus carota root protoplast를 polyethyleneglycol Ca^2+ 법으로 처리한 결과 introduction frequency가 47.6%였다. 4. 본 연구에서 얻은 실험결과로 부터 liposome법, spheroplast법 모두 Ti plasmid의 식물 protoplast로의 도입은 용이하게 일어나는 것으로 생각된다. It was attempted in the present study to prepare radioactive Ti plasmid to identify the introduction frequency of Ti plasmid encapsulated liposome into Dacus carota root protoplast. Agrobcterium tumefaciens C58 was cultured in 0.8% nutrient broth containing radioactive phosphoric acid(H_3^32PO_4) and the radioactive Ti plasmid wad isolated. Radioactive Ti plasmid was encapsulated into phosphatidyl choline-stearylamine liposomes by reverse phase evaporation technique and the encapsulation frequency was found to be about 17%. Radioactive Ti plasmid encapsulated liposome and Dacus carota root protoplast was treated by polyethyleneglycol Ca^2+ procedure. The introduction frequency was 38%. The spheroplast of A. tumefaciens C38 was prepared from the bacteria cultured in 0.8% nutrient broth containing radioactive phosphoric acid using lysozyme. As much as 47% of the spheroplast were found to be introduced into Dacus carota protoplast.
