        The present study was designed to isolate and characterize novel chemical constituents of the stem bark of Juglans mandshurica Maxim. (Juglandaceae).The chemical constituents were isolated and purified by various chromatographic techniques. The structures of the compounds were elucidated on the basis of spectral data (1D and 2D NMR, HR-ESI-MS, CD, UV, and IR) and by the comparisons of spectroscopic data with the reported values in the literatures. Two long chain polyunsaturated fatty acids (1 and 2) were obtained and identified as (S)-(8E, 10E)-12-hydroxy-7-oxo-8, 10-octadecadienoic acid (1) and (S)-(8E, 10E)-12-hydroxy-7-oxo-8,10-octadecadienoic acid methyl ester (2). To the best of our knowledge, this is the first report on the isolation and structural elucidation of the two new conjugated ketonic fatty acids from this genus.

      • Lentivirus-mediated shRNA Interference Targeting SLUG Inhibits Lung Cancer Growth and Metastasis

        Objective: Lung cancer is a deadly cancer, whose kills more people worldwide than any other malignancy. SLUG (SNAI2, Snail2) is involved in the epithelial mesenchymal transition in physiological and in pathological contexts and is implicated in the development and progression of lung cancer. Methods: We constructed a lentivirus vector with SLUG shRNA (LV-shSLUG). LV-shSLUG and a control lentivirus were infected into the non-small cell lung cancer cell A549 and real-time PCR, Western blot and IHC were applied to assess expression of the SLUG gene. Cell proliferation and migration were detected using MTT and clony formation methods. Results: Real-time PCR, Western Blot and IHC results confirmed down-regulation of SLUG expression by its shRNA by about 80%~90% at both the mRNA and protein levels. Knockdown of SLUG significantly suppressed lung cancer cell proliferation. Furthermore, knockdown of SLUG significantly inhibited lung cancer cell invasion and metastasis. Finally, knockdown of SLUG induced the down-regulation of Bcl-2 and up-regulation of E-cadherin. Conclusion: These results indicate that SLUG is a newly identified gene associated with lung cancer growth and metastasis. SLUG may serve as a new therapeutic target for the treatment of lung cancer in the future.

        Chemical constituents from the leaves of Juglans mandshurica

        Two new (1 and 3) and two known diarylheptanoids(2 and 4), along with two tetralones (5 and 6), onenaphthoquinone (7), four phenylpropanoids (8–11), andone phenol (12) were isolated from the leaves of Juglansmandshurica. Their structures were elucidated on the basisof spectral and chemical data. Compounds 2 and 10 arefirstly isolated from this plant and 8 and 12 were isolatedfrom the Juglans genus for the first time. Among thesecompounds, only 7 exhibited moderate cytotoxicitiesagainst cultured MGC-803, A549, K562, and HeLa tumorcell lines with IC50 values of 25.90, 28.60, 39.06,44.90 lM, respectively.

        Two new (1 and 3) two known diarylhep-tanoids (2 and 4), along with two tetralones (5 and 6), one naphthoquinone (7), four phenylpropanoids (8-11), and phenol (12) were isolated from the leaves of Juglans mandshurica. Their structures were elucidated on the basis of spectral and chemical data. Compounds 2 and 10 are firstly isolated from this plant and 8 and 12 were isolated from the Juglans genus for the first time. Among these compounds, only 7 cxhibitcd moderate cytotoxicities against cultured Mgc-803, A549, K562, and Hela tumor cell lines with IC50 values of 25.90, 28.60, 39.06, 44.90 LLM, respectively.

        Evaluation of the efficacy of unipolar and bipolar spinal dorsal root ganglion radiofrequency thermocoagulation in the treatment of postherpetic neuralgia

        Background: Different views have been proposed on the radiofrequency treatment modes and parameters of radiofrequency thermocoagulation of the spinal dorsal root ganglion for the treatment of postherpetic neuralgia (PHN). It is urgent to identify a more effective therapy for patients with PHN. Methods: Patients who underwent radiofrequency thermocoagulation therapy for PHN were retrospectively reviewed and were divided into a radiofrequency thermocoagulation (CRF) and double neddles radiofrequency thermocoagulation (DCRF). The pain scores (numerical rating scale, NRS) were evaluated at the following time points: before the operation, 1 day, 3 months, 6 months, 1 year, and 2 years after operation. The incidence of complications and the degree of pain relief were evaluated. The in vitro ovalbumin experiment was used to indicate the effects of radiofrequency thermocoagulation. Results: Compared with the preoperative NRS scores, the postoperative NRS scores decreased significantly; the NRS scores of the DCRF group was lower than that of the CRF group at all time points from 6 months to 2 years following the operation. The total effective rate of the DCRF group was significantly higher than that of the CRF group at 2 years following the operation. The incidence of numbness in the DCRF group was higher than that noted in the CRF group. The ovalbumin experiments in vitro indicated that the effects of radiofrequency thermocoagulation were optimal when the distance between the two needles was 5 mm. Conclusions: DCRF with a 5 mm spacing exhibits a longer duration and higher effective rate in the treatment of PHN and is worth promoting.

        Lifetime Prediction and Aging Behaviors of Nitrile Butadiene Rubber under Operating Environment of Transformer

        Based on the actual operating environment of transformer, the aging tests of nitrile butadiene rubber (NBR) were conducted systematically under four conditions: in air, in transform oil, under compression in air and under compression in transform oil to studythe effect of high temperature, transform oil and compression stress simultaneously on the thermal aging behaviors of nitrile butadiene rubber and predict the lifetime. The effects of liquid media and compression stress simultaneously on the thermal aging behaviors of nitrile butadiene rubber were studied by using characterization methods such as IR spectrosc-opy, thermogravimetric measurements, Differential Scanning Calorimetry (DSC) measurements and mechanical property measurements. The changes in physical properties during the aging process were analyzed and compared. Different aging conditions yielded materials with different properties. Aging at 70°C under compression stress in oil, the change in elongation at break was lower than that aging in oil, but larger than that aging under compression in air. The compression set or elongation at break as evaluation indexes, 50% as critical value, the lifetime of NBR at 25°C was predicted and compared. When aging under compression in oil, the prediction lifetime was lower than in air and under compression in air, and in oil. It was clear that when predicting the service lifetime of NBR in oil sealing application, compression and media liquid should be involved simultaneously. Under compression in oil, compression set as the evaluation index, the prediction lifetime of NBR was shorter than that of elongation at break as the evaluation index. For the life prediction of NBR, we should take into account of the performance trends of NBR under actual operating conditions to select the appropriate evaluation index.

        Cytotoxic anthraquinone dimers from Melandrium firmum

        Two new anthraquinone dimers, melrubiellin A(1) and melrubiellin B (2), were isolated from the aerialpart of Melandrium firmum Rohrbach, along with sevenknown compounds (3–9). The structures of these compoundswere elucidated by spectral analyses, including 1Dand 2D NMR (COSY, HMQC, HMBC and NOESY)experiments. Compound 1 and 2 exhibited significantcytotoxicity towards HeLa, NCI-H460, Hep G2, Hep 3Band MKN-28 cell lines with IC50 values ranging from 5.26to 81.16 lM.

        Chemical constituents from the aerial parts of Melandrium firmum

        Two new anthraquinones, melrubiellin C (1) andmelrubiellin D (2), were isolated from the aerial parts of Melandriumfirmum Rohrbach, together with eight known compounds(3–10). The structures of these compounds wereelucidated using 1Dand 2DNMR(COSY,HMQC,HMBCandNOESY) experiments. All isolated compounds were tested fortheir cytotoxicity against NCI-H460, Hep G2, MKN-28 andA-549 cells.Of these 10 compounds,1 and 2 exhibitedmoderatecytotoxicity with IC50 values ranging from 9.54 to 32.41 lM.
