<P>We performed chloroplast genome sequencing and comparative analysis of two Rutaceae species, <I>Zanthoxylum schinifolium</I> (Korean pepper tree) and <I>Z. piperitum</I> (Japanese pepper tree), which are medicinal and culinary crops in Asia. We identified more than 837 single nucleotide polymorphisms and 103 insertions/deletions (InDels) based on a comparison of the two chloroplast genomes and developed seven DNA markers derived from five tandem repeats and two InDel variations that discriminated between Korean <I>Zanthoxylum</I> species. Metabolite profile analysis pointed to three metabolic groups, one with Korean <I>Z. piperitum</I> samples, one with Korean <I>Z. schinifolium</I> samples, and the last containing all the tested Chinese <I>Zanthoxylum</I> species samples, which are considered to be <I>Z. bungeanum</I> based on our results. Two markers were capable of distinguishing among these three groups. Hypermethylation of PDX1, EN2, and MSX1 predicts the prognosis of colorectal cancer

Despite numerous observations regarding the relationship between DNA methylation changes and cancer progression, only a few genes have been verified as diagnostic biomarkers of colorectal cancer (CRC). To more practically detect methylation changes, we performed targeted bisulfite sequencing. Through co-analysis of RNA-seq, we identified cohort-specific DNA methylation markers: CpG islands of the intragenic regions of PDX1, EN2, and MSX1. We validated that these genes have oncogenic features in CRC and that their expression levels are increased in correlation with the hypermethylation of intragenic regions. The reliable depth of the targeted bisulfite sequencing data enabled us to design highly optimized quantitative methylation-specific PCR primer sets that can successfully detect subtle changes in the methylation levels of candidate regions. Furthermore, these methylation levels can divide CRC patients into two groups denoting good and poor prognoses. In this study, we present a streamlined workflow for screening clinically significant differentially methylated regions. Our discovery of methylation markers in the PDX1, EN2, and MSX1 genes suggests their promising performance as prognostic markers and their clinical application in CRC patients.

Purpose: Vascular endothelial growth factor (VEGF) contributes to tumor angiogenesis including lung cancer. The role of VEGF single nucleotide polymorphisms (SNPs) in susceptibility and prognosis of lung cancer remains inconclusive and controversial. We hypothesized that VEGF polymorphisms may affect survival outcome among early stage non- small cell lung cancer (NSCLC) in Korean population. Method: We genotyped three potentially functional VEGF SNPs [rs833061 T>C, rs2010963 G>C, and rs3025039 C>T] and evaluated the relationship between these VEGF variants and survival outcome among early stage NSCLC patients, who were treated with surgical resection at Kyungpook National University Hospital. Analyses were performed using Kaplan-Meier log-rank test, and Cox proportional hazard models. Result: Although all three polymorphisms were not significantly associated with overall survival (OS) and disease-free survival (DFS) in NSCLC as whole, there were significant differences in OS and DFS by rs833061 T>C polymorphism when separated by tumor histology. Conclusion : rs833061 T>C polymorphism may be a prognostic marker of NSCLC and affect differently depending on tumor histology.

Purpose: This study was conducted to investigate whether polymorphisms of genes involved in immune checkpoints can predict the clinical outcomes of patients with advanced stage non-small cell lung cancer (NSCLC) after 1st line paclitaxel-cisplatinchemotherapy. Experimental Design: A total of 379 patients with advanced stage NSCLC were enrolled. Twelve single nucleotide polymorphisms (SNPs) of PD-1, PD-L1, and CTLA-4 genes were selected and genotyped. The associations of SNPs with chemotherapy response and overall survival (OS) were analyzed. Results: Among the 12 SNPs investigated, PD-L1 rs2297136T>C and rs4143815C>G were significantly associated with clinical outcomes after chemotherapy. The rs2297136T>C was significantly associated with both better chemotherapy response and better OS (adjusted odds ratio [aOR] = 1.82, 95% CI = 1.19-2.77, P = 0.006; adjusted hazard ratio [aHR] = 0.76, 95% CI = 0.61-0.94, P = 0.01), and the rs4143815C>G had a significantly better response to chemotherapy (aOR = 1.42, 95% CI = 1.05-1.93, P = 0.02) under additive model for the variant alleles. Consistent with the individual genotype analyses, rs2297136C-rs4143815G haplotype harboring variant alleles at both loci was significantly associated with better chemotherapy response and OS compared with combined other haplotypes (aOR = 1.92, 95% CI = 1.27-2.91, P = 0.002; aHR = 0.76, 95% CI = 0.61-0.94, P = 0.01, respectively). Conclusions: PD-L1 rs2297136T>C and rs4143815C>G polymorphisms may be useful for the prediction of clinical outcome of 1st line paclitaxel-cisplatin chemotherapy in NSCLC.

Utilization of catalytic hydrolysis of ethyl acetate for solvent removal during microencapsulation

<P>The objective of this study was to apply the specific acid-catalysed hydrolysis of ethyl acetate to completing solvent extraction during an emulsion-based microencapsulation process. The dispersed phase consisting of poly-<SMALL>D,L</SMALL>-lactide-<I>co</I>-glycolide and ethyl acetate was emulsified in an acid catalyst containing aqueous phase. Catalytic hydrolysis of ethyl acetate led to its continual leaching from the dispersed phase of the emulsion, thereby triggering microsphere hardening with high efficiency. Ketoprofen was successfully encapsulated into microspheres via this technique, and liquid chromatography-mass spectrometry showed that its structural integrity was preserved during microencapsulation. Compared to typical solvent extraction approaches, the acid-catalysis technique helped minimize the consumption of a quench liquid. Also, the resultant microspheres displayed excellent dispersibility and decreased propensity for aggregation. Furthermore, the new method provided better drug encapsulation efficiency and lower levels of residual ethyl acetate in microspheres. In conclusion, the acid-catalysis approach had great potential for the preparation of versatile microspheres and nanoparticles.</P>

In this paper, a water thermal seat is proposed to increase thermal comfort in automobiles. In order to confirm the applicability of a water thermal seat, a numerical study was conducted on the driver's thermal comfort according to the use of the basic seat and water thermal seats (hot and cold water seats) in the car cabin under summer and winter conditions. As a result, after an elapsed cooling time of 30 min under summer condition, the predicted percentage of dissatisfaction (PPD) using the cold water seat was reduced by 23.4 % compared with that when using the basic seat. In winter, a slight dissatisfaction with the PPD (10.1 %) was presented for the basic seat, compared to 7.8 % in the initial 5 min when the hot water seat was used. Therefore, it was confirmed that the simultaneous use of HVAC and water thermal seats during summer and winter could significantly reduce the discomfort of the driver at the beginning of driving.
