Let-7c Inhibits NSCLC Cell Proliferation by Targeting HOXA1

Objective: The aim of the present study was to explore mechanisms by which let-7c suppresses NSCLC cell proliferation. Methods: The expression level of let-7c was quantified by qRT-PCR. A549 and H1299 cells were transfected with let-7c mimics to restore the expression of let-7c. The effects of let-7c were then assessed by cell proliferation, colony formation and cell cycle assay. Mouse experiments were used to confirm the effect of let-7c on tumorigenicity in vivo. Luciferase reporter assays and Western blotting were performed to identify target genes for let-7c. Results: HOXA1 was identified as a novel target of let-7c. MTS, colony formation and flow cytometry assays demonstrated that forced expression of let-7c inhibited NSCLC cell proliferation by inducing G1 arrest in vitro, consistent with inhibitory effects induced by knockdown of HOXA1. Mouse experiments demonstrated that let-7c expression suppressed tumorigenesis. Furthermore, we found that let-7c could regulate the expression of HOXA1 downstream effectors CCND1, CDC25A and CDK2. Conclusions: Collectively, these results demonstrate let-7c inhibits NSCLC cell proliferation and tumorigenesis by partial direct targeting of the HOXA1 pathway, which suggests that restoration of let-7c expression may thus offer a potential therapeutic intervention strategy for NSCLC.

The APETALA2/ethylene-responsive elementbinding factors (AP2/ERF) play central roles in the stressresponse in plants. In this study, we identified and isolateda novel salt stress-related gene, LcERF080, that encodes anAP2/ERF protein in Lotus corniculatus cultivar Leo. LcERF080 was classified into the B-4 group of the ERFsubfamily based on multiple sequence alignment andphylogenetic characterization. Expression of LcERF080was strongly induced by salt, abscisic acid, 1-aminocyclopropane-1-carboxylic acid, methyl jasmonate, and salicylicacid stresses. Subcellular localization assay confirmedthat LcERF080 is a nuclear protein. LcERF080 overexpressionin Arabidopsis resulted in pleiotropic phenotypeswith a higher seed germination rate and transgenic plantswith enhanced tolerance to salt stress. Further, under stressconditions, the transgenic lines exhibited elevated levels ofsoluble sugars and proline as well as relative moisturecontents but a lower malondialdehyde content than incontrol plants. The expression levels of hyperosmoticsalinity response genes COR15A, RD22, and P5CS1 werefound to be elevated in the LcERF080-overexpressingArabidopsis plants compared to the wild-type plants. Theseresults reveal that LcERF080 is involved in the responsesof plants to salt stress.
Strong Tracking Tobit Kalman Filter with Model Uncertainties

The Tobit Kalman filter (TKF) is a good choice for applications of discrete time-varying systems withcensored measurements. The traditional TKF is usually designed under the assumption of exactly knowing systemstate and measurement functions, which can not guarantee the good performance and is even unsuitable for systemwith model mismatch. To ensure the performance, this paper proposes a novel TKF algorithm in the presenceof both censored measurements and model uncertainties. Our proposed algorithm, called strong tracking TKF(STTKF), adopts the orthogonal principle as an additional criterion to overcome model mismatch problem. Byintroducing fading factor into a priori error covariance, STTKF adaptively adjust gain matrix according to differentmodel mismatch degree. Firstly the recursive fading factor formulation is deduced based on orthogonal principle. Then the designed STTKF process is given in a recursive manner. Finally the effectiveness of STTKF is verified bytwo examples of oscillator system and unmanned aerial vehicle (UAV) transmitter.
Effect of Silver Doping on Transport Properties of Bi2Se3: AgxBi2Se3 and Bi2−xAgxSe3

Ag-doped Bi2Se3with the formula AgxBi2Se3and Bi2−xAgxSe3 were prepared and their electrical and magnetic transportproperties have been investigated to study the influence of silver doping on transport properties of Bi2Se3with differentAg-doped method. All samples exhibited metallic resistivity and the resistivity increased with increasing Ag concentration. The lattice parameter c of Ag-substituted and Ag-intercalated samples displays a contrary change as the Ag concentrationincreased. For the Ag-intercalated samples, both the resistance upturn were observed in the curves of temperature dependentof resistivity and temperature dependent of magnetoresistance, respectively, indicating that the enhanced surface effectwas obtained in those samples. Monotonously, field-induced MR peaks around 200 K were also observed in those samples. Similar behaviors were not observed in the Ag-substituted samples.
Testing serial correlation for partially nonlinear models

Partially nonlinear models, as extensions of partially linear models are extensively used in statistical modeling. This paper considers the spline empirical log-likelihood ratio for testing serial correlation in partially nonlinear models. It is shown that the proposed empirical log-likelihood ratio converges to the standard chi-square distribution under the null hypothesis of no serial correlation. Some simulations are conducted to estimate the rejection probabilities under the null hypothesis and serial correlation. An example of application is also illustrated.
