      • 산삼 체세포배양근의 에탄올 농도 및 추출 시간에 따른 최적 추출조건

        Background : From 2000 years ago, Panax ginseng is identified as precious pharmaceutical plant. Depend on growing environment, the name would be vary. For instance, it is called "mountain cultured ginseng (jangnoesam)" which is artificially grown ginseng, "Cultured ginseng (jaebaesam)" which refer to the ginseng grown in the forest, and lastly "Wild ginseng (sansam)" which inhabits in deep mountain. The main active compounds in the Panax ginseng is called ginsenoside and many researches have been performing in biological field. However, most studies focus on functional ability of ginseng. In this study, to seek the suitable extraction condition and antioxidant activity, cell cultured Panax ginseng was extracted according to different ethanol concentration and extraction time. Methods and Results : To establish the optimal extraction condition, the sample was pulverized into 500 μm and added 10% (v/v), 30% (v/v), 50% (v/v), 70% (v/v) and, 90% (v/v) EtOH. After that, the samples are extracted in different time by ultrasonic bath (Power sonic 520, Hwashin Co., Korea). The extracts was filtered by Whatman No. 2 filtering paper. Eventually, the saponin was separated by n-butanol as the ginsenoside, the combination of terpenoid and sugar. The extraction yield of 90% cell cultured panax ginseng EtOH extract was 7.36±0.33%, which was the lowest extraction yield and simultaneously, 10% EtOH extract showed 1.8 times more yield that of 90% EtOH extract. The saponin extraction yield revealed 10% and 70% EtOH extract showed 1.64±0.06% and 3.13±0.08%, respectively. Conclusion : The suitable extraction yield in cell cultured panax ginseng and saponin were evaluated by different extraction condition such as ethanol concentration and extraction time. As a result, when 10% EtOH was applied as solvent, the yield was doubles of 90% EtOH extract. As ethanol became high concentrations, the extraction yield was gradually increased. Among them, crude saponin, the main active compounds in Panax ginseng was extracted the most by 70% EtOH and that value was 3.13±0.08%.

        Association between Osteoporosis and Skeletal Muscle Mass in Men

        Study Design: Cross-sectional study.Purpose: This cross-sectional study aimed to investigate the risk factors for osteoporosis in men by assessing bone mineral density (BMD), skeletal muscle mass, body fat mass, grip strength, and advanced glycation end products (AGEs).Overview of Literature: Fewer studies have reported the correlation between BMD and skeletal muscle mass in women. Moreover, a few studies have examined the relationship between osteoporosis and skeletal muscle mass.Methods: This study included 99 men (mean age, 74.9 years; range, 28–93 years) who visited Qiball Clinic for BMD and body composition examinations. The osteoporosis group consisted of 24 patients (mean age, 72.5 years; range, 44–92 years), and the control group consisted of 75 individuals (mean age, 74.9 years; range, 28–93 years). Whole-body skeletal muscle mass was measured using a bioelectrical impedance analyzer. BMD was measured by dual X-ray absorptiometry. Skin autofluorescence (SAF), a marker of dermal AGE accumulation, was measured using a spectroscope. Osteoporosis was defined as a bone density T score of –2.5 or less. Physical findings, skeletal muscle mass, BMD, grip strength, and SAF were compared between the osteoporosis and control groups.Results: The osteoporosis group had significantly lower trunk muscle mass (23.1 kg vs. 24.9 kg), lower leg muscle mass (14.4 kg vs. 13.0 kg), and skeletal mass index (7.1 kg/m<sup>2</sup> vs. 6.7 kg/m<sup>2</sup>) than the control group (all <i>p</i><0.05). Lower limb muscle mass was identified as a risk factor for osteoporosis in men (odds ratio, 0.64; <i>p</i>=0.03).Conclusions: Conservative treatment of osteoporosis in men will require an effective approach that facilitates the maintenance or strengthening of skeletal muscle mass, including exercise therapy with a focus on lower extremities and nutritional supplementation.

        에탄올 농도별 당유자 잎의 최적추출조건 및 생리활성 평가

        The Citrus grandis Osbeck is a special product in the Jeju island. The product has been as a remedy for liver damage and hang over. This study demonstrates how to investigate and compare the antioxidant, phenol content, tyrosinase and α-glucosidase inhibitory activity, antimicrobial, and alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activity with the C. grandis leaves extracted in different ethanol concentrations. From the yield, a 20% ethanol extract demonstrated the highest results among the other extracts. The distilled water extract showed the most abundant in a total phenol content and highest ABTS radical scavenging activity and reducing power assay. In the DPPH radical scavenging activity, α-glucosidase and tyrosinase inhibitory assay (used L-tyrosine as substrate), the 80% ethanol extract exhibited a higher value than other extracts. The 60% ethanol extract showed prominent activities in the tyrosinase inhibitory (used L-dopa as substrate), ADH and ALDH activity assay. In the minimum inhibitory concentration (MIC) assay, 60% and 80% ethanol extracts inhibited the bacterial growth almost similarly. Moreover, the gram-positive bacteria was more restrained than the gram-negative bacteria. The results revealed that the distilled water and 80% ethanol extract showed a relatively higher antioxidant activity compared to other extracts. The 60 ~ 80% ethanol extracts demonstrated potential tyrosinase, α -glucosidase inhibitory, antimicrobial, ADH and ALDH activities. Therefore, the C. grandis is suggested to be considered as a functional material for various proposes.

        에탄올 농도별 당유자 잎의 최적추출조건 및 생리활성 평가

        제주도 특산물로 알려진 당유자의 잎을 이용한 연구는 미흡한 실정이며, 특히 용매 농도별로 다양한 생리활성을 측정한 연구는 지금까지 보고되어 있지 않았다. 이에 따라 본 연구에서는 에탄올 농도별로 추출된 당유자 잎에 대한 생리활성을 측정하였다. 수율과 총 페놀 함량은 각각 20% 에탄올, 물 추출물이 가장높은 함량을 가진 것으로 나타났으며 가장 낮은 수율을 보인 100% 에탄올 추출물에서 비교적 높은 페놀 함량이 나타나 수율에 비하여 많은 페놀 함량이 확인되었다. 환원력과 ABTS radical 소거능 실험 결과 물 추출물이 가장 높은 활성을 나타냈으며 80% 에탄올 추출물이 DPPH radical 소거능, ${\alpha}$-glucosidase, ${\text\tiny{L}}$-tyrosine을 기질로 한 tyrosinase 저해 활성에서 가장 우수한 활성을 가진 것으로 나타났다. 또한 ${\text\tiny{L}}$-dopa를 기질로 한 tyrosinase 저해 활성과 지금까지 보고된 바 없는 ADH 및 ALDH 활성에서는 60% 에탄올 추출물이 가장뛰어난 활성을 촉진시켰고 미생물 생육을 억제하는 능력이 60%, 80% 에탄올에서 거의 동일하게 나타났다. 이 결과에서 항산화 효과를 얻고자 할때는 물이나 80% 에탄올로, 그 외 생리활성(${\alpha}$-glucosidase 저해활성, tyrosinase 저해활성, 항미생물 활성, ADH 및 ALDH 활성)에서는 60~80% 에탄올로 추출하는 것이 가장 효과적일 것으로 사료된다. 본 실험의 결과는 당유자 잎을 신소재 개발 원료로서 사용하는데 하나의 기초 자료가 될 것으로 기대된다.       • Transaplantation of neural stem cells into the spinal cord after injury

        Thanks to advances in the stem cell biology of the central nervous system (CNS), the previously inconceivable regeneration of the damaged CNS is approaching reality. The availability of signals to induce the appropriate differentiation of the transplanted and/or endogenous neural stem cells(NSCs) as well as the timing of the transplantation are important for successful functional recovery of the damaged CNS. Because the immediately post-traumatic microenvironment of the spinal cord is in an acute inflammatory stage, it is not favorable for the survival and differentiation of NSC transplants. On the other hand, in the chronic stage after injury, glial scars form in the injured site that inhibit the regeneration of neuronal axons. Thus, we believe that the optimal timing of transplantation is 1-2 weeks after injury.

      • 메탄올 농도에 따른 당유자 잎의 항산화, α-Glucosidase 억제활성 측정

        Backgrounds : Pomelo (Citrus grandis Osbeck) is the kind of citrus fruit which is Dicotyledoneae belongs to Rutaceae and special product in Jeju island. According to the previous researches, coumarin, eliocitrin, naringin are identified and these kinds of constituents revealed to be effective as anticancer, antioxidant, and antidiabetic. Until recently, there are many investigations about its functional properties were reported, but investigation about biological activities depend on extraction conditions are not sufficient. Methods and Results : 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was performed by the method of Blois with minor modification. After adding DPPH radical to each sample solutions, the mixtures were incubated in 30 minutes at aphotic place. Then, the degree of scavenging activity was recorded by microplate reader at 490 nm. The scavenging activity was expressed by RC50, which is the concentration of sample solution necessitated to scavenge 50% DPPH radical against negative control. For α-glucosidase inhibitory activity, the method using p-nitrophenyl-α-D-glucopyranoside (pNPG) was applied. To each samples, α-glucosidase and pNPG were mixed and incubated, consequently, Na2CO3 was added to terminate the reaction. Finally, absorbance was read at 450 nm. The same as DPPH radical scavenging activity, the inhibition was explicitly expressed by the amount of sample solution to inhibit 50% α-glucosidase, IC50. The 80% MeOH extract demonstrated the highest radical scavenging activity with 74.32±8.45 μg/ml. α-Glucosidase inhibitory activity was carried out to evaluate the inhibitory activity of sugar digestion and absorption and 60, 80% MeOH extract exhibited 416.35±11.07 μg/ml and 336.57±2.03 μg/ml, respectively. Conclusion : The DPPH radical scavenging and α-glucosidase inhibitory activity were evaluated in this study. The highest DPPH radical scavenging activity was seen in 80% Citrus grandis Osbeck MeOH extract, following 60% MeOH extract exhibited the second highest scavenging activity. Also, 80% MeOH extract showed 336.57±2.03 μg/ml, which was the highest α-glucosidase inhibition among all extracts.
