Despite intensive research efforts in recent decades, cancer remains a leading cause of death worldwide. The chalcone family is a promising group of phytochemicals for therapeutic use against cancer development. Naturally-occurring chalcones, as well as synthetic chalcone analogues, have shown many beneficial biological properties, including anti-inflammatory, antioxidant, and anti-cancer activities. In this report, trans-chalcone (TChal) was found to increase cell death in breast cancer cells, assessed using high content screening. Subsequently, using antibody array analysis, TChal was found to increase heme oxygenase-1 (HO-1) expression in TChal-treated breast cancer cells. Blocking of HO-1 by siRNA in breast cancer cells diminished the effect of TChal on cell growth inhibition. TChal-fed mice also showed less tumor growth compared to vehicle-fed mice. Overall, we found that TChal increases HO-1 expression in breast cancer cells, thereby enhancing anti-tumorigenesis. Our results suggest that HO-1 expression could be a potential new target of TChal for anti-tumorigenesis in breast cancer.
Effect of a professional soccer match in skin temperature of the lower limbs : a case study

The purpose of this study is to analyze the relationship between lower limbs skin temperature (Tsk) recorded by infrared thermography and creatine kinase (CK) response following an official soccer game. A pro-fessional soccer player, from the first division Brazilian League, defender, 27 years and 183 cm of height was analyzed. Measures of Tsk with a thermal camera and blood CK concentrations were obtained 24 hr be-fore (24H-PRE), 24 (24H-POST) and 48 hr (48H-POST) after an official match. The results showed that CK values were 193 U/L 24 hr before, ris-ing to 1,083 U/L 24 hr after the game and 414 U/L 48 hr after the game. Anterior thigh Tsk was 31.5ºC, 33.8ºC, and 31.8ºC in the moments 24H-PRE, 24H-POST, and 48H-POST, respectively. Similarly, anterior leg presented temperature of 31.2ºC, 33.3ºC, and 31.5ºC at the same moments. Qualita-tive analysis of the thermograms showed that whole lower limbs are much warmer 24 hr after the match and certain areas as the right anteri-or thigh, the left anterior leg, both anterior ankles, and both posterior thighs have not fully recovered their initial Tsk 48 hr after the match. The results of this study indicate that participation in a professional soccer match can lead to significant increases in Tsk values measured by IRT 24 hr after the match. Considering this variable as an indicator of muscu-lar damage, it could help in the process of training control, being part of an injury prevention program in professional soccer clubs.
Production system influences color stability and lipid oxidation in gluteus medius muscle

Objective: We aimed to evaluate the color and oxidative stability of beef gluteus medius (GM) from cattle raised in organic and non-organic production systems. Methods: The GM samples (n = 10) were obtained from organic (ORG; n = 5) or nonorganic (NORG; n = 5) beef samples, sliced into 2.54-cm steaks, packaged in aerobic conditions, and stored for nine days at 4°C. ORG and NORG steaks were compared regarding myoglobin concentration, pH, instrumental color, delta E (ΔE), metmyoglobin reducing activity (MRA), and lipid oxidation on days 0, 5, and 9. Results: Feeding system did not influence (p>0.05) the myoglobin concentration. ORG steaks exhibited greater (p<0.05) meat pH, yellowness, and MRA, whereas NORG steaks exhibited greater (p<0.05) redness, chroma, R630/580, delta E, and lipid oxidation. ORG and NORG steaks exhibited similar (p>0.05) lightness and hue angle. During storage, ORG and NORG exhibited an increase in muscle pH, hue angle, and lipid oxidation; and a decrease (p<0.05) in redness, yellowness, chroma, and color stability (R630/580). Both samples exhibited a stable (p>0.05) pattern for lightness and MRA. Conclusion: Therefore, the production system can affect beef color and lipid stability during storage.

<P>Curcumin is a potential anticancer drug with poor bioavailability, which limits its clinical use as a therapeutic agent. The aim of this study was a preliminary evaluation of the curcumin analogue CH-5 as a cytotoxic agent in human osteosarcoma cell lines U2OS, MG-63, and Saos-2. CH-5 inhibited cell viability at lower concentrations than curcumin, leading to the induction of apoptosis. The cellular levels of the transcription factors p53 and Sp1 affect the expression of cellular pathways that lead to apoptosis. CH-5 increased p53 protein levels in U2OS cells and reduced Sp1 levels, with a consequent effect on the expression of their target genes DNA methyltransferase 1 (<I>DNMT1)</I> and growth arrest and DNA damage-inducible 45 alpha gene (<I>Gadd45a)</I>. CH-5 repressed <I>DNMT1</I> and increased <I>Gadd45a</I> mRNA expression, which was dependent on p53, as this effect was only observed in the colorectal cancer cell line HCT116 with active p53, but not in the isogenic p53-deficient HCT116 cells. CH-5 also reduced the protein levels of DNMT1, which led to the upregulation of <I>Gadd45a</I>. These results suggest that CH-5 has potentially higher anticancer activity than curcumin, which is associated with the expression of apoptosis-associated genes regulated by the transcription factors Sp1 and p53. Future work on CH-5 will define the therapeutic potential of this compound in vivo.</P>

<P>Osteosarcoma (OS) is a primary malignant bone tumor that mainly affects children, adolescents, and young adults. The inhibition of metastasis is a main strategy of OS therapy since the development of metastatic disease due to drug resistance remains the most important cause of death from this cancer. Considering the severe side effects of current OS chemotherapy, the identification of anti-metastatic drugs with reduced toxicity is of great interest. Chalcones are polyphenols with a basic structure consisting of an α-, β-unsaturated carbonyl system linking two aryl rings. These compounds exhibit anticancer activity against a variety of tumor cell lines through multiple mechanisms, including the regulation of the tumor-suppressor protein p53 and its target genes. An important process regulated by p53 is epithelial-mesenchymal transition (EMT), which facilitates tumor metastasis by conferring migratory and invasive properties to cancer cells. The activation of p53 can revert EMT and reduce migration and invasion. This study aimed to examine the inhibitory effects of two 4′-aminochalcones on the migration/invasion of the U2OS (p53+/+) and SAOS-2 (p53−/−) OS cell lines as well as the underlying molecular mechanisms. Cell viability was examined by MTT assay. Transwell assays were used to evaluate the migratory and invasive ability of the cells. The two 4′-aminochalcones showed low capacity to inhibit the viability of OS cells independent of p53 status, but preferentially suppressed the migration of U2OS cells and of a SAOS-2 cell line expressing p53. Invasion was strongly inhibited by both chalcones independent of p53 status. RT-PCR, zymography, and Western blot were used to study the expression of matrix metalloproteinases and EMT markers after treatment with the chalcones. The results indicated that the 4′-aminochalcone-induced antimigratory and anti-invasive effects are potentially associated with the inhibition of extracellular matrix (ECM) enzymatic degradation in OS cells and with the modulation of EMT genes. These effects probably result from the induced increase of p53 protein expression by the two chalcones. In conclusion, chalcones D14 and D15 have potential anti-metastatic activity mediated by p53 that can be exploited for OS treatment.</P>
Effect of microtemperatures for micropolar thermoelastic bodies

In this paper we investigate the theory of micropolar thermoelastic bodies whose micro-particles possess microtemperatures. We transform the mixed initial boundary value problem into a temporally evolutionary equation on a Hilbert space and after that we prove the existence and uniqueness of the solution. We also approach the study of the continuous dependence of solution upon initial data and loads.

A New CAD Environment for Benchmarks of Analog Circuit Sizing Tools and Algorithms

This paper presents a new open environment for the design of analog circuits exchanging information from existing tools. The main features of these tools have been extended in CONNAN by means of a netlist analyzer, the interaction with a commercial CAD tool and a transistor model manager that solves some common problems of sizing tools based on electrical simulators. As an initial test CONNAN is applied to the sizing of analog IC based on electrical simulators and simulated annealing algorithms.

Abortive infection systems (Abi) are phage resistance systems that can be prophage-encoded. Here, two genes encoding an Abi system were identified on a prophage sequence contained by the chromosome of the Levilactobacillus brevis strain UCCLBBS124. This Abi system is similar to the two-component AbiL system encoded by Lactococcus lactis biovar. diacetylactis LD10-1. The UCCLBBS124 prophage-derived Abi system (designated here as AbiL124) was shown to exhibit specific activity against phages infecting L. brevis and L. lactis strains. Expression of the AbiL124 system was shown to cause reduction in the efficiency of plaquing and cell lysis delay for phages of both species.
