      • Experimental and numerical flow analysis in hydraulic vane pump

        The purpose of this study is to conduct a three dimensional flow analysis of a hydraulic vane pump by using computational fluid dynamics (CFD), to evaluate an accuracy of volumetric flow rate in CFD compared to experimental value and to examine a influence of the evaporation coefficient and condensation coefficient in Zwart–Gerber–Belamri cavitation model to pressure in a vane chamber. In this study, standard k-ε turbulent model with the Zwart–Gerber–Belamri cavitation model is used to estimate the cavitating flow in vane pump. In CFD, two notch models are used. From results of experiments and CFD analysis of volumetric flow rate, there is little difference between the experiments and CFD results in the low speed range. However, for high speed range, in a case with no cavitation model, the CFD results of volumetric flow rate are different from the experimental ones. On the other hand, in the case with cavitation model, those agree with the experimental values. In this case, the error of volumetric flow rate is less than 1% in the low speed range and less than 2.5% in the high speed range. From CFD results of pressure fluctuation in a vane chamber when the evaporation coefficient and condensation coefficient in the cavitation model are varied, the surge pressure can be seen in a vane chamber. In conclusion, it could be considered that the CFD calculation with cavitation model is sufficiently accurate in volumetric flow rate. In addition, as the evaporation coefficient becomes large, amplitude of the pressure fluctuation in a vane chamber after a surge pressure becomes large, as the condensation coefficient becomes large, the surge pressure in a vane chamber becomes low.

        Long-term safety and effectiveness of adalimumab in Japanese patients with Crohn’s disease: 3-year results from a real-world study

        Background/Aims: Crohn’s disease is a chronic disorder; therefore, it is essential to investigate long-term safety and efficacy of treatments. This study assessed the safety and effectiveness of adalimumab for up to 3 years in Japanese patients with Crohn’s disease in real-world settings. Methods: This was a multicenter, single-cohort, observational study of patients with Crohn’s disease. Safety assessments included incidence of adverse drug reactions. Effectiveness assessments included clinical remis-sion, mucosal healing, and Work Productivity and Activity Impairment (WPAI). Results: The safety and effectiveness analysis populations comprised 389 and 310 patients, respectively. Mean (standard deviation) exposure to adalimumab in the safety analysis population was 793.4 (402.8) days, with a 58.1% retention rate. A total of 105 patients (27.0%) and 43 patients (11.1%) experienced adverse drug reactions and serious adverse drug reactions, respectively, with no patient reporting tuberculosis or hepatitis B. Infections and serious infections were reported in 37 patients (9.5%) and 17 patients (4.4%), respectively. Malig-nancy was reported as an adverse drug reaction in 2 patients (0.5%). Remission rate increased from 37.8% (98/259) at baseline to 73.9% (167/226) at week 4 and remained >70% over 3 years. Proportion of patients without mucosal ulcerations increased from 2.7% (2/73) at baseline to 42.3% (11/26) between years >2 to ≤3. WPAI improvement started at 4 weeks, with the overall work impairment score improving from 42.7 (n=102) at baseline to 26.9 (n=84) at 4 weeks. Conclusions: Results from this study confirm the long-term safety and effectiveness of adalimumab treatment in Japanese patients with Crohn’s disease in the real-world setting.         Reduced Coenzyme Q10 Decreases Urinary 8-Oxo-7,8-Dihydro-20-Deoxyguanosine Concentrations in Healthy Young Female Subjects

        It remains unclear whether dietary supplementation with coenzyme Q10 (CoQ10) provides beneficial effects for healthy individuals, especially young subjects. This study investigated the effects of dietary supplementation with CoQ10 on oxidative stress in healthy young females. We performed a placebo-controlled trial using a crossover design (n = 28) with 100 mg/day CoQ10 in reduced form or placebo, each lasting 2 weeks with a 2-week interval. The urinary levels of 8-oxo-7,8-dihydro-20-deoxyguanosine (8-oxodG), a biomarker of oxidative DNA damage, were determined by high-performance liquid chromatography (HPLC) coupled to an electrochemical detector. Levels of malondialdehyde (MDA), a biomarker of lipid peroxidation, and antioxidant vitamin C in urine were also measured using a thiobarbituric acid-reactive substance method with a commercial kit and by the 2,4-dinitrophenylhydrazine method with HPLC, respectively. Urinary 8-oxodG levels during supplementation with reduced form of CoQ10 (median [first and third quartiles]: 1.76 [1.24–2.08] nmol/mmol creatinine) were significantly lower than those with placebo (2.00 [1.34–2.49] nmol/mmol creatinine, P = .031 by Student’s paired t-test using the logarithmically transformed values). In contrast, the urinary levels of MDA and vitamin C were not significantly affected (P = .094 and P = .247 by Student’s paired t-test, respectively). There was no evidence of any side effects. Supplementation with CoQ10 in the reduced form showed a slightly protective effect against oxidative DNA damage even in healthy young subjects.
