        Effect of Acibenzolar-S-methyl and Rahnella aquatilis (Ra39) on Chitinase and β-1, 3-glucanase Activities and Disease Resistance of Apple Plants

        The effect of Acibenzolar-S-methyl (ASM) and Rahnella aquatilis Ra39 against apple fire blight disease caused by Erwinia amylovora were tested as a possible alternative to streptomycin. In vitro studies, no inhibition effect against the pathogen was found when ASM was tested. Under greenhouse conditions, application of R. aquatilis Ra39 with the highly susceptible M26 rootstock resulted in a marked disease suppression. Application of ASM and strain Ra39 caused a high decrease of the disease, 82% and 58% respectively; this was correlated with a reduction of the growth of the pathogen within host plants up to 64% and 49.5% respectively. Further studies in the field under artificial infection condition during full bloom revealed that application of ASM and R. aquatilis Ra39 with Gala variety resulted in a control effect up to 21 and 29% respectively. In physiological studies, enhanced activities of PR-proteins (chitinase and β-1, 3-glucanase) were detected, which are well known as biochemical markers for systemic acquired resistance. Application of ASM to apple shoots caused the highest chitinase activity followed by strain Ra39. The enzyme activity was increased after 2, 4 and 6 days from application. In addition, ASM-treatment caused the higher β-1, 3-glucanase activity than strain Ra39. Maximum enzyme activity was recorded after 6 days from application and then decreased after 8 and 10 days from application.

        Selection of Sahiwal Cattle Bulls on Pedigree and Progeny

        The objective of the study was to compare ranking of Sahiwal bulls selected on the basis of highest lactation milk yield of their dams with their estimated breeding values (EBVs) using an animal model. Data on 23,761 lactation milk yield records of 5,936 cows from five main Livestock Experiment Stations in Punjab province of Pakistan (1964-2004) were used for the study. At present the young A.I bulls are required to be from A-category bull-dams. Dams were categorized as A, B, C and D if they had highest lactation milk yield of ${\geq}$2,700, 2,250-2,699, 1,800-2,249 and <1,800 litres, respectively. The EBVs for lactation milk yield were estimated for all the animals using an individual animal model having fixed effect of herd-year and season of calving and random effect of animal. Fixed effect of parity and random effect of permanent environment were incorporated when multiple lactation were used. There were 396 young bulls used for semen collection and A.I during 1973-2004. However, progeny with lactation yields recorded, were available only for 91 bulls and dams could be traced for only 63 bulls. Overall lactation milk yield averaged 1,440.8 kg. Milk yield was 10% heritable with repeatability of 39%. Ranking bulls on highest lactation milk yield of their dams, the in-vogue criteria of selecting bulls, had a rank correlation of 0.167 (p<0.190) with ranking based on EBVs from animal model analysis. Bulls' EBVs for all lactations had rank correlation of 0.716 (p<0.001) with EBVs based on first lactation milk yield and 0.766 (p<0.001) with average EBVs of dam and sire (pedigree index). Ranking of bulls on highest lactation yield of their dams has no association with their ranking based on animal model evaluation. Young Sahiwal bulls should be selected on the basis of pedigree index instead of highest lactation yield of dams. This can help improve the genetic potential of the breed accruing to conservation and development efforts.

        Inducing Potential Mutants in Bread Wheat Using Different Doses of Certain Physical and Chemical Mutagens

        Mutation is an effective strategy not only for creating novel variation into crop genome but also for direct releasing adapted and high-yielding genotypes. The current work explores inducing genetic variability in bread wheat using physical and chemical mutagens. Three wheat cultivars were treated by three mutagens; gamma irradiation (five doses; 250, 300, 350, 400 and 450 Gray); laser ray (three treatments; 1, 1.5, and 2 hour exposure) and EMS (three concentrations; 0.2, 0.3 and 0.4%). Besides, a combination of physical (laser) and chemical (EMS) mutagens using middle range of each treatment (1.5 hour laser and 0.3% EMS) was attempted to be applied. The treated seeds were sown in the first season and 4050 M1 plants were harvested. The harvested seeds were sown in the second season, and 78750 M2 plants were obtained. The selection was performed in second season (M2) based on morpho-physiological and yield traits; flag leaf area, flag leaf chlorophyll content, plant height, spike length, grain yield per plant and its components. Based on evaluated traits fourteen mutants were selected to be evaluated in the third generation (M3). The results indicated that the used mutagens had direct impact and significantly improved agronomic traits in derivative mutants compared to their parent cultivars. Moreover, the maximum increment in yield related traits were obtained by 0.4% EMS, 1 and 2 hour-laser, 350-Gy, 1.5 hour × 0.3% EMS and 250-Gy. The obtained results highlighted the importance of these doses of applied mutagens to induce useful genetic variability in bread wheat for improving grain yield and contributing traits.

        Identification of Arcanobacterium pyogenes isolated by post mortem examinations of a bearded dragon and a gecko by phenotypic and genotypic properties

        The present study was designed to identify phenotypically and genotypically two Arcanobacterium (A.) pyogenes strains isolated by post mortem examinations of a bearded dragon and a gecko. The A. pyogenes strains showed the typical biochemical properties and displayed CAMP-like synergistic hemolytic activities with various indicator strains. The species identity could be confirmed genotypically by amplification and sequencing of the 16S rDNA gene and, as novel target gene, by sequencing of the beta subunit of RNA polymerase encoding gene rpoB, of both strains and of reference strains representing nine species of the genus Arcanobacterium. The species identity of the two A. pyogenes strains could additionally be confirmed by PCR mediated amplification of species specific parts of the 16S-23S rDNA intergenic spacer region, the pyolysin encoding gene plo and by amplification of the collagen-binding protein encoding gene cbpA. All these molecular targets might help to improve the future identification and further characterization of A. pyogenes which, as demonstrated in the present study, could also be isolated from reptile specimens.

        NEW CCD OBSERVATIONS AND THE FIRST PHOTOMETRIC STUDY OF THE CONTACT BINARY AP UMI

        We obtain the first complete CCD light curves (LCs) of the contact binary AP UMi in the VRI bands and analyzed them by means of the PHOEBE code. A spotted model is applied to treat the asymmetry in the LCs. The LC morphology clearly shows the O'Connell effect and the solution shows an influence of star spots on both components. Such effect of star spots is common between the RS CVn and W UMa chromospherically active stars. Based on the obtained solution of the LCs we investigate the evolutionary state of the components and conclude that the system is a pre-intermediate contact binary ($f=0.29$) with mass ratio $q=0.38$, and it is an A-type W UMa system where the less massive secondary component is cooler than the more massive primary one.} We obtain the first complete CCD light curves (LCs) of the contact binary AP UMi in the VRI bands and analyzed them by means of the PHOEBE code.         Analgesic and antipyretic actions of Muntingia calabura leaves chloroform extractin animal models

        The present study was carried out to elucidate the potential of Muntingia (M.) calabura leaves chloroform extract (MCCE) as antinociceptive, anti-inflammatory and antipyretic agents using various animal models. The dried powdered leaves of M. calabura (20 g) were soaked in chloroform for 72 h and the supernatant obtained was then evaporated to dryness. The crude dried extract (0.912 g), dissolved in dimethyl sulfoxide (1 : 20; w/v) and considered as a stock solution (100% concentration/strength), was then diluted to the concentrations of 10 and 50% and used together in all experimental models. The MCCE was found to show significant (P < 0.05) antinociceptive and antipyretic activities, but less remarkable anti-inflammatory activity. Only the antinociceptive activity of MCCE measured using the abdominal constriction test and in the first phase of the formalin test occurred in a concentration-dependent manner. The anti-inflammatory activity of 50 and 100% concentrations MCCE was observed only at the range of time interval of 60 - 120 and 60 min, respectively. Based on the results, we conclude that the M. calabura leaves chloroform extract possessed remarkable antinociceptive and antipyretic, but less effective anti-inflammatory, activities and thus justifies the Peruvian folklore claims of its medicinal values.         Removal of deltamethrin insecticide over highly porous activated carbon prepared from pistachio nutshells

        Potassium hydroxide-activated carbons (CK21, CK11, and CK12) were prepared from pis-tachio nutshells. Physicochemical properties of activated carbons were characterized by TGA, pHpzc, Fourier transform infrared spectroscopy, scanning electron microscopy, and N2-adsorption at -196°C. The examinations showed that activated carbons have high sur-face area ranging between 695-1218 m2/g, total pore volume ranging between 0.527-0.772 mL/g, and a pore radius around 1.4 nm. The presence of acidic and basic surface C-O groups was confirmed.Batch adsorption experiments were carried out to study the effects of adsorbent dosage, temperature, initial concentration of adsorbate, and contact time on deltamethrin adsorption by activated carbons. The kinetic studies showed that the adsorp-tion data followed a pseudo-second order kinetic model. The Langmuir model showed a maximum adsorption capacity of 162.6 mg/g at 35°C on CK12. Thermodynamic studies indicated that adsorption was spontaneous and increased with temperature, suggesting an endothermic process.

        Synthesis, molecular docking studies of coumarinyl-pyrazolinyl substituted thiazoles as non-competitive inhibitors of mushroom tyrosinase

        A series of coumarinyl-pyrazolinyl substituted thiazoles derivatives were synthesized and their inhibitory effects on the DPPH and mushroom tyrosinase were evaluated. The results showed that all of the synthesized compounds exhibited significant mushroom tyrosinase inhibitory activities. In particular, 3-(5-(4-(benzyloxy)-3-methoxyphenyl)-1-(4-(4-bromophenyl)thiazol-2-yl)-4,5-dihydro-1H-pyrazol-3-yl)-2H-chromen-2-one (7j) exhibited the most potent tyrosinase inhibitory activity with IC<SUB>50</SUB> value 0.00458+/-0.00022μM compared with the IC<SUB>50</SUB> value of kojic acid is 16.84+/-0.052μM. The inhibition mechanism analyzed by Lineweaver-Burk plots revealed that the type of inhibition of compound 7j on tyrosinase was noncompetitive. The docking study against tyrosinase enzyme was also performed to determine the binding affinity of the compounds. The compound 7a showed the highest binding affinity (-10.20kcal/mol) with active binding site of tyrosinase. The initial structure activity relationships (SARs) analysis suggested that further development of such compounds might be of interest. The statistics of our results endorses that compound 7j may serve asa structural template for the design and development of novel tyrosinase inhibitors.

        Management of Charcoal Rot of Sesame by Seed Soaking in Medicinal Plant Extracts and Hot Water

        Macrophomina phaseolina causing charcoal rot was isolated from sesame seeds (cvs. Giza 32 and Shandawel-3)collected from different localities of Assiut, Sohage and El-Minia Governorates. The fungus was found in the highest frequently in samples collected from Assiut Governorate followed by Sohag and finally EL Minia Governorate. The obtained isolates were different in their virulence on the tested sesame cvs. Also, they differed in their growth nature including colony color and sclerotial production. The color of colonies of the pathogen seem to be correlated with density of sclerotial formation. Aqueous extracts of Majorna, Wild chamomile,Geranium oil and Nees plants were highly toxic to tested isolates of M. phaseolina, in vitro. On the other hand the rest of the tested aqueous extracts had no effect. Under greenhouse conditions in 2005 and 2006seasons, soaking seeds of sesame before sowing in aqueous extracts of Eucalyptus, Nerium, Ocimum and Roesmary plants decreased the disease incidence. Aqueous extracts of Eucalyptus and Ocimum were the most effective treatment. Dipping sesame seeds in hot water at 60oC for 5 minutes increased seed germination of Giza 32 and Shandawel-3 cvs. followed by 55 oC, 50oC and 45oC, while 40oC treatment resulted the lowest seed germination rate. Dipping sesame seeds in hot water at different temperature before planting decreased seed,seedling and charcoal rots. Soaking seeds in hot water at 60oC increased greatly plant height and decreased seed,seedling rot and charcoal rot followed by 55oC and 50oC, under greenhouse condition.
