        Attenuation of Experimental Autoimmune Hepatitis in Mice with Bone Mesenchymal Stem Cell-Derived Exosomes Carrying MicroRNA-223-3p

        MicroRNA-223-3p (miR-223-3p) is one of the potential microRNAs that have been shown to alleviate inflammatory responses in pre-clinical investigations and is highly encased in exosomes derived from bone mesenchymal stem cells (MSC-exosomes). MSC-exosomes are able to function as carriers to deliver microRNAs into cells. Autoimmune hepatitis is one of the challenging liver diseases with no effective treatment other than steroid hormones. Here, we examined whether MSC-exosomes can transfer miR-223-3p to treat autoimmune hepatitis in an experimental model. We found that MSC-exosomes were successfully incorporated with miR-223-3p and delivered miR-223-3p into macrophages. Moreover, there was no toxic effect of exosomes on the macrophages. Furthermore, treatments of either exosomes or exosomes with miR-223-3p successfully attenuated inflammatory responses in the liver of autoimmune hepatitis and inflammatory cytokine release in both the liver and macrophages. The mechanism may be related to the regulation of miR-223-3p level and STAT3 expression in the liver and macrophages. These results suggest that MSC-exosomes can be used to deliver miR-223-3p for the treatment of autoimmune hepatitis.

        Interaction between Barium Oxide and Carbon Film in BaO/C/Mo System under High Temperature

        Carbon ¯lm, used as the coating of the Mo grid in pulsed-controlled grid traveling wave tubes, can eectively suppress electron emission from the Mo grid contaminated by the emission material of the hot cathode, i.e. BaO or Ba, so that the lifetime of the tubes can be prolonged signi¯cantly but the reasons for it have not been well understood. To study the eect of it on the BaO/Mo system under high temperature, carbon ¯lms were prepared on Mo substrate at room temperature by a dual-ion-beam sputtering deposition system, and post-annealing was conducted to know their microstructure under high temperature. In our experiments, BaO layers were coated on Mo and carbon-coated Mo substrates by the chemical method to compare with each other, and the prepared BaO/Mo and BaO/C/Mo samples were annealed at two dierent temperatures of 973 K and 1223 K in order to investigate the interaction between barium oxide and carbon ¯lm under high temperature. The results show that the BaO/C/Mo changes into C/Mo after the exhaustion of BaO at 1223 K, nevertheless, This does not happen at 973 K. In this paper, the mechanism whereby the addition of carbon ¯lm can suppress the grid emission under its operating condi-tion is discussed according to the experimental results and the calculation of the reaction free energy.

        Heterologous expression of Solanum tuberosum NAC1 gene confers enhanced tolerance to salt stress in transgenic Nicotiana benthamiana

        The plant-specific transcription factors NACs (NAM, ATAF1, 2, CUC), one major transcription factor family, play significant roles in various physiological processes including abiotic stresses. In our study, an NAC gene from potato (Solanum tuberosum L.) was cloned and named as StNAC1 for a high-sequence similarity to SlNAC1, a well-known tomato NAC gene regulating multiple stress responses and fruit ripening. StNAC1 gene was significantly induced under salt stress. Then, we constructed StNAC1-overexpressing transgenic Nicotiana benthamiana plants and obtained three homozygous transgenic lines. The phenotypic analysis results showed that StNAC1-overexpressing transgenic plants had not only higher seed germination and green leaf rates, but also accumulated less ROS and more proline than wide-type plants under salt stress, which resulted in improving transgenic plants salt tolerance. These suggested that StNAC1 gene might function as a positive regulator in plant response to salt stress.

        WDRP, a DWD protein component of CUL4-based E3 ligases, acts as a receptor of CDPK-related protein kinase 5 to mediate kinase degradation in Arabidopsis

        CRK5 is a member of the Arabidopsis thaliana Ca2+-dependent protein kinase-related kinase family. Here, a yeast two-hybrid screen was performed with a truncated form of AtCRK5 as bait to identify interacting proteins and determine its physiological roles. One gene encoding the DWD protein WDRP was isolated. Furthermore, in vitro and in vivo co-immunoprecipitation results strongly supported that these two proteins interact with each other. Using a cell-free degradation assay, we also established that CRK5 was an unstable protein that was degraded through the proteasome pathway. The rate of CRK5 degradation was delayed in a WDRP knockout line. On the other hand, the degradation of CRK5 mediated by WDRP might not affect the phosphorylation of PIN2 by CRK5. Overall, we demonstrated that AtCRK5 interacted with a DWD protein, AtWDRP; the protein AtWDRP targets the kinase for ubiquitin-dependent degradation. Therefore, this report describes a new kinase regulation pathway for CRK family proteins in Arabidopsis.

      • GOLPH3, a Good Prognostic Indicator in Early-stage NSCLC Related to Tumor Angiogenesis

        Background: Golgi phosphoprotein-3 (GOLPH3) is implicated in cancer development and progression. The aim of this study was to evaluate the prognostic significance of GOLPH3 protein and its association with tumor angiogenesis in patients with early-stage NSCLC. Materials and Methods: Immunohistochemistry was performed to determine GOLPH3 protein expression and allow assessment of intratumoral microvessel density (MVD) by counting CD-34 positive immunostained endothelial cells. Correlations of expression with MVD, clinicopathologic features and clinical prognosis were analyzed. Results: A notably higher level of GOLPH3 expression was found in early-stage NSCC tissues at the protein level. However, we do not find any correlation between GOLPH3 expression and clinicopathologic features (p>0.05), although higher MVD was positively associated with GOLPH3 overexpression (p<0.001). Expression of GOLPH3 was found to be an independent prognostic factor in early-stage NSCLC patients, those expressing high levels of GOLPH3 exhibiting a substantially lower 5-year overall survival than GOLPH3-negative patients (adjusted HR =1.899, 95% CI: 1.021-3.532, p=0.043). Conclusions: High expression of the GOLPH3 protein is common in early-stage NSCC, and is closely associated with tumor progression, increased tumor angiogenesis, and poor survival. We conclude a possibility of its use as a diagnostic and prognostic marker in early-stage NSCC patients.

        A Review on Microbial Decaffeination

        Exclusive exposure of caffeine might bring some adverse effects to human health, and cause some risks to environment. Microbial decaffeination might be a new alternative approach because of its high specificity and efficiency. Many studies revealed that several microorganisms can degrade caffeine, including genera Alcaligenes,Rhodococcus, Klebsiella, Acinetobacter, Enterobacter, Stenotrophomonas and Pseudomonas. Biodegradation of caffeine can be significantly influenced by temperature, pH, aeration rate and initial caffeine concentration of the incubation. There are two main pathways of caffeine biodegradation: C-8 oxidation and N-demethylation, while N-demethylation is the general catabolism pathway in bacteria. Application of microbial decaffeination has also been discussed in this review.

        The Identification of Six Novel Proteins with Fibronectin or Collagen Type І Binding Activity from Streptococcus suis Serotype 2

        Streptococcus suis, a major swine pathogen, is an emergingzoonotic agent that causes meningitis and septic shock. Bacterialcell wall and secreted proteins are often involved ininteractions with extracellular matrix proteins (ECMs), whichplay important roles in the initial steps of pathogenesis. Inthis study, 2D SDS-PAGE, western blotting-based bindingaffinity measurements, and microtiter plate binding assayswere used to identify cell wall and secreted proteins from S. suis that interact with fibronectin and collagen type І. Weidentified six proteins from S. suis, including three proteins(translation elongation factor G, oligopeptide-binding proteinOppA precursor, and phosphoglycerate mutase) thatshow both fibronectin and collagen type І binding activity. To the best of our knowledge, these three newly identifiedproteins had no previously reported fibronectin or collagentype І binding activity. Overall, the aim in this study was toidentify proteins with ECM binding activity from S. suis andit represents the first report of six new proteins from S. suisthat interact with fibronectin or collagen type І.

        Biological characterization of mesenchymal stem cells from bovine umbilical cord

        Mesenchymal stem cells (MSCs) are multi-potential cells that are able to proliferate and differentiate into othercell types. Much research has been done on the MSCs from the umbilical cord (UCMSCs) in human, mice, andavian, but little literature has been published about these cells in big livestock. Here, we choose Luxi cattle asthe experimental animal, we describe an external culture of the UCMSCs from it and summarize the biologicalcharacteristics of these cells, e.g., morphologic appearance, surface antigens, colony-forming ability, geneexpression, and differentiation potential were detected via using immunofluorescence and reverse transcriptionpolymerase chain reaction (RT-PCR). The induced cells, osteoblast, lipoblast, hepatocyte, islet cells, andneurocyte were identified by Alizarin red staining, Oil-red-O staining, Periodic acid-schiff staining, andDithizone staining and RT-PCR detection for specific genes. Results suggest that biological characteristics ofthe UCMSCs were similar to those of MSCs previously analyzed. The primary UCMSCs were sub-cultured topassage 32, the UCMSCs expressed gene CD29, CD44, CD73, CD90, and CD166, induced cells illustratedtypical staining, and expressed specific genes, which indicate that the UCMSCs could be a novel alternativesource of MSCs for experimental and clinical applications.

        Research on the Cutting Performances and Wear Mechanisms of TiAlCrN Coated Tools during Dry Turning

        Physical vapour deposition technique was used to deposit TiAlCrN coatings on the YT15 tungsten carbide inserts. The dry turning tests of 20CrMo steel were carried out to evaluate performances of TiAlCrN coated and uncoated tools on the CA6140A lathe. The effect of the two kinds of tools on cutting forces, cutting temperature, surface roughness and tool wear had been investigated to assess the performance of TiAlCrN coated tools. The results showed that the cutting force and cutting temperature obtained by TiAlCrN coated tools were decreased and the TiACrN coated tools produced a better surface finish in comparison with the uncoated tools. The TiAlCrN coated tools yield working life about 45 min, which was two times of that for uncoated tools. The wear mechanisms of the TiAlCrN coated tools were mainly oxidation and boundary wear, accompanied with diffusion wear.

        Isolation of L-theanine from Tea Solution by Cation Exchange Resin in Batch and Fixed Bed Column

        L-Theanine, a bioactive compound in tea, was isolated from tea solution using cation exchange resin no.732. The adsorption of L-theanine by cation exchange resin no.732 fit the Langmuir isotherm model and was a monolayer molecular interaction process. Thermodynamic studies revealed that the adsorption of L-theanine by resin no.732 was an exothermic and spontaneous physically driven process. The adsorption capacity was influenced by temperature, initial concentration, and pH. The L-theanine adsorption capacity under conditions at room temperature,pH 4.73, and initial L-theanine concentration 18 g/L was 241.731 ± 3.679 mg/g. The Thomas model was fit to describe the column adsorption data at different flow rates and initial concentrations. The L-theanine adsorbed by resin no.732 could be desorbed by 0.134 mol/L Na2HPO4aqueous solution with a recovery rate of 84.96%. These findings indicate that resin no.732 was a promising material for isolating L-theanine from tea solution.
