        LINC00319 promotes cancer stem cell-like properties in laryngeal squamous cell carcinoma via E2F1-mediated upregulation of HMGB3

        Laryngeal squamous cell carcinoma (LSCC) is one of the most common subtypes of head and neck malignancies worldwide. Long intervening/intergenic noncoding RNAs (LINCRNAs) have been recently implicated in various biological processes that take place in the setting of laryngeal cancer, but the regulatory role of LINC00319 in LSCC remains largely unknown. The current study aimed to elucidate the regulatory effect of LINC00319 on the development and progression of LSCC via high-mobility group box 3 (HMGB3). Microarray-based analysis was initially conducted to identify differentially expressed long noncoding RNAs, after which the expression of LINC00319 and HMGB3 in LSCC tissues and cells was determined accordingly. CD133 + CD144 + TU177 cells were subsequently isolated and transfected with LINC00319 overexpression vector (oe-LINC00319), short hairpin RNA (sh)-LINC00319, sh-HMGB3, sh-E2F transcription factor 1 (E2F1), and oe-E2F1, as well as their corresponding controls. The proliferative, invasion, self-renewal, and tumorigenic abilities of CD133 + CD144 + TU177 cells were then evaluated. Our in vitro findings were further confirmed following subcutaneous injection of cells expressing the corresponding plasmids into nude mice. LINC00319 and HMGB3 expressions were elevated in LSCC cells and tissues. LINC00319 increased HMGB3 expression by recruiting E2F1. Furthermore, the stimulatory role of LINC00319 on the proliferation, invasion, self-renewal ability, and tumorigenicity of CD133 + CD144 + TU177 cells was achieved by upregulating HMGB3 via recruitment of E2F1. The in vitro findings were also confirmed by in vivo experiments. Taken together, these data show that downregulating LINC00319 in CD133 + CD144 + TU177 cells may serve as a potential anticancer regimen by inhibiting the proliferation and invasion of cancer stem cells in LSCC.

        Numerical modeling and global performance analysis of a 15-MW Semisubmersible Floating Offshore Wind Turbine (FOWT)

        The global performance of a 15 MW floating offshore wind turbine, a newly designed semisubmersible floating foundation with multiple heave plates by CNOOC, is investigated with two independent turbine-floater-mooring coupled dynamic analysis programs CHARM3D-FAST and OrcaFlex. The semisubmersible platform hosts IEA 15 MW reference wind turbine modulated for VolturnUS-S and hybrid type (chain-wire-chain with clumps) 3×2 mooring lines targeting the water depth of 100 m. The numerical free-decay simulation results are compared with physical experiments with 1:64 scaled model in 3D wave basin, from which appropriate drag coefficients for heave plates were estimated. The tuned numerical simulation tools were then used for the feasibility and global performance analysis of the FOWT considering the 50-yr-storm condition and maximum operational condition. The effect of tower flexibility was investigated by comparing tower-base fore-aft bending moment and nacelle translational accelerations. It is found that the tower-base bending moment and nacelle accelerations can be appreciably increased due to the tower flexibility.

      • Effect of Raw Fly Ash on Water Holding Capacity of Different Sizes Distributed Soil and Cellulose Treatment of Fly Ash

        Environmental pollution caused by huge amount of generated coal fly ash (FA) without utilization and the deterioration of desertification in arid areas are big social problems. This study investigated the effect of raw coal fly ash amending on water holding capacity (WHC) of decomposed granite soil (DGS) and Akatama soil (AS) with different size distribution. Cellulose treated FA was also tested in the experiment, as an investigation on the polymer grafting method of FA modification, to try to increase the positive effect on WHC of soil. The test soil was dried at room temperature and 40 ºC respectively, and its moisture loss was monitored per hour. The particle size has an effect on WHC, and the relationship between the particle size and WHC is not a leaner relationship. It shows there WHC has the dependency on soil particle size and there are peak points of the WHC curve at certain ranges of particle size. In general, raw FA has a positive effect on the WHC of DGS and AS. However, cellulose-treated didn’t have a significant positive effect on WHC of DGS. This study shows that the impact of FA on WHC is complex, and many factors can influence WHC such as particle sizes, drying temperature, soil type, mixing ratio of FA, and FA treatment type.
