        ATG5 Expression Induced by MDMA (Ecstasy), Interferes with Neuronal Differentiation of Neuroblastoma Cells

        The amphetamine derivative 3, 4-methylenedioxymetham- phetamine (MDMA) has become a popular recreational drug, and has also been shown to cause serotonergic neurotoxicity. This report shows that MDMA impairs brain development in a whole mouse embryo culture. The results of quantitative real-time PCR analysis showed that autophagy-related protein 5 (Atg5) expression is elevated in mouse embryo and neuroblastoma cells after MDMA treatment. This elevated Atg5 expression inter-feres with the neuronal differentiation of neuroblastoma cells such as SH-SY5Y and PC12 cells. Thus, our results suggest that the use of MDMA during pregnancy may impair neuronal development via an induction of Atg5 expression.

      • Synthesis and characterization of TRITON™ X-based surfactants with carboxylic or amino groups in the oxyethylene chain end

        <P>This paper describes the synthesis and characterization of a series of TRITON™ X-based surfactants with a predominantly alkyl phenol ethoxylate (APE) backbone and carboxylic or amino chain ends. Three carboxylic acid-containing TRITON X (or APE)-derivatives, [OPE2-COOH], [OPE5-COOH], and [OPE10-COOH], were prepared from commercially available octyl phenol ethoxylate (OPE) of different oxyethylene units (i.e., n = 2, 5, and 10) and ethyl bromoacetate via a simple etherification reaction followed by saponification. Two amine-containing TRITON X -derivatives, [NPE10-NH<SUB>2</SUB>] and [NPE10-imidazole], were prepared based on modifications of mesylate of nonyl phenol ethoxylate (NPE) of 10 oxyethylene units reacted with ammonia and imidazole, respectively. Depending on their composition and chain length of oxyethylene units used in the reaction, the surfactants show different thermal degradation behaviors.       • Dkk3, downregulated in cervical cancer, functions as a negative regulator of β-catenin

        <P>The Wnt/β-catenin signaling pathway is activated during the malignant transformation of keratinocytes that originate from the human uterine cervix. Dkk1, 2 and 4 have been shown to modulate the Wnt-induced stabilization of the β-catenin signaling pathway. However, the function of Dkk3 in this pathway is unknown. Comparison of the Dkk3 gene expression profiles in cervical cancer and normal cervical tissue by cDNA microarray and subsequent real-time PCR revealed that the Dkk3 gene is frequently downregulated in the cancer. Methylation studies showed that the promoter of Dkk3 was methylated in cervical cancer cell lines and 22 (31.4%) of 70 cervical cancer tissue specimens. This promoter methylation was associated with reduced expression of Dkk3 mRNA in the paired normal and tumor tissue samples. Further, the reintroduction of Dkk3 into HeLa cervical cancer cells resulted in reduced colony formation and retarded cell growth. The forced expression of Dkk3 markedly attenuated β-catenin-responsive luciferase activity in a dose-dependent manner and decreased the β-catenin levels. By utilizing a yeast two-hybrid screen, βTrCP, a negative regulator of β-catenin was identified as a novel Dkk3-interacting partner. Coexpression with βTrCP synergistically enhanced the inhibitory function of Dkk3 on β-catenin. The stable expression of Dkk3 blocks the nuclear translocation of β-catenin, resulting in downregulation of its downstream targets (VEGF and cylcin D), whereas knockdown of Dkk3 abrogates this blocking. We conclude from our finding that Dkk3 is a negative regulator of β-catenin and its downregulation contribute to an activation of the β-catenin signaling pathway. © 2008 Wiley-Liss, Inc.</P>

      • Clinical outcomes of patients with hepatorenal syndrome after living donor liver transplantation

        <P><B>Abstract</B></P><P>Liver transplantation (LT) is the treatment of choice for hepatorenal syndrome (HRS). However, the clinical benefits of living donor liver transplantation (LDLT) are not yet well established. We, therefore, investigated the outcomes of patients with HRS who underwent LDLT and patients with HRS who received transplants from deceased donors. This study focused on 71 patients with HRS out of a total of 726 consecutive adult Korean patients who underwent LT at a single Asian center. We compared 48 patients who underwent LDLT with 23 patients who underwent deceased donor liver transplantation (DDLT). Patients with HRS showed poorer survival than patients without HRS (<I>P</I> = 0.01). Poorer survival was associated with higher in‐hospital mortality for patients with HRS (18.3% versus 5.2%, <I>P</I> < 0.001). In comparison with DDLT, LDLT was associated with younger donors and shorter ischemic times. The survival rate with LDLT was significantly higher than the survival rate with DDLT (<I>P</I> = 0.02). Among patients with high Model for End‐Stage Liver Disease scores (≥30) or type 1 HRS, the survival rates for the LDLT group were not inferior to those for the DDLT group. LDLT significantly improved recipient survival after adjustments for several risk factors (hazard ratio = 0.20, 95% confidence interval = 0.05‐0.85, <I>P</I> = 0.03). Kidney function was significantly improved after LT, and there was no difference between LDLT and DDLT. No patients in the HRS cohort required maintenance renal replacement therapy.         글루탐산 생산을 위한 Lactococcus lactis HY7803 균주의 대두 발효 적용

        The glutamic acid producing strain for development savory taste enhancing foods was identified the possibility through application into soybean fermentation. To check the effects on glutamic acid production during soybean fermentation, Lactococcus lactis HY7803 was introduced as a starter. The soybean samples were analyzed on days 0, 7, 14 and 21. The numbers of bacteria decreased gradually, while the content of amino-type nitrogen increased during fermentation in the soybean with L. lactis HY7803. Glutamic acid content in soybeans with L. lactis HY7803 increased from 114.99 ± 9.37 pmol/ul on day 0 to 138.14 ± 1.76 pmol/ul on day 21, showing an overall higher amino acid content than soybeans without L. lactis HY7803 and similar content to soybeans with Aspergillus oryzae SNU-G. It was clearly distinguished through principal component analysis. Consequently, our results indicate that L. lactis HY7803 is available as a fungus replacement and may be a good starter strain for enhancing savory taste in vitro as well as soybean fermentation.

        Selection of Lactococcus lactis HY7803 for Glutamic Acid Production Based on Comparative Genomic Analysis

        Comparative genomic analysis was performed on eight species of lactic acid bacteria (LAB)― Lactococcus (L.) lactis, Lactobacillus (Lb.) plantarum, Lb. casei, Lb. brevis, Leuconostoc (Leu.) mesenteroides, Lb. fermentum, Lb. buchneri, and Lb. curvatus―to assess their glutamic acid production pathways. Glutamic acid is important for umami taste in foods. The only genes for glutamic acid production identified in the eight LAB were for conversion from glutamine in L. lactis and Leu. mesenteroides, and from glucose via citrate in L. lactis. Thus, L. lactis was considered to be potentially the best of the species for glutamic acid production. By biochemical analyses, L. lactis HY7803 was selected for glutamic acid production from among 17 L. lactis strains. Strain HY7803 produced 83.16 pmol/μl glutamic acid from glucose, and exogenous supplementation of citrate increased this to 108.42 pmol/μl. Including glutamic acid, strain HY7803 produced more of 10 free amino acids than L. lactis reference strains IL1403 and ATCC 7962 in the presence of exogenous citrate. The differences in the amino acid profiles of the strains were illuminated by principal component analysis. Our results indicate that L. lactis HY7803 may be a good starter strain for glutamic acid production.

        다문화예비학교의 역할과 개선과제 - 한국어 강사의 교육 경험을 중심으로 -

        본 연구는 중도입국 청소년들의 교육지원 정책 일환으로 실시되고 있는 다문화예비학교가 어떻게 운영되고 있는지 탐색하고, 이를 바탕으로 발전 방향을 모색하고자 하였다. 이를 위하여 경기도 다문화예비학교에 배치되어 한국어교육을 담당하고 있는 한국어 강사 6명을 대상으로 개별심층면담을 실시하였다. 연구결과 첫째, 다문화예비학교는 중도입국 청소년에게 공교육 진입기회를 부여 할뿐만 아니라 한국사회와의 소통 통로로써 중요한 기능을 하고 있었다. 둘째, 다문화예비학교에서 이루어지는 교육과정에서 중도입국 청소년들의 다양한 국적과 학습격차, 상이한 입학시기로 인해 개별 특성에 맞는 교육기회 제공이 필요하다. 마지막으로 다문화예비학교의 주요 변화 방안으로 중도입국 청소년들에게 학생으로서의 지위와 소속감을 부여하는 것과 유관기관과의 네트워크 형성으로 통합적 서비스 제공 구축이 필요한 것으로 나타났다. 다문화예비학교에 대한 이해를 제공하고 나아가 향후 중도입국 청소년에 대한 교육정책 수립과 실천 방안을 모색하기 위한 사회적 담론화에 기여하고자 한다. The present study aims to investigate the operation of multicultural preparatory schools, implemented as a part of education support policy for immigrant teenagers, and to seek improvement measures based on the study findings. To this end, an in-depth interview was conducted individually targeting six Korean language instructors in charge of Korean language education at a multicultural preparatory school in Gyeonggi Province. The study results are as follows. First, multicultural preparatory schools not only provide the opportunity for immigrant teenagers to enter public education but also perform a crucial function as a channel of communication with the Korean society. Second, the curriculum at multicultural preparatory schools needs to offer education opportunities tailored to individual needs based on diverse nationalities of immigrant teenagers, gaps in academic performance, and different time of enrollment. Finally, it was found that as a main strategy for improvement, multicultural preparatory schools need to provide a comprehensive service by forming a network with affiliated organizations while encouraging immigrant teenagers to develop a sense of belonging and status as students. This study intends to promote understanding of multicultural preparatory schools and contribute to initiation of a social discourse that can lead to establishment of education policy and implementation strategies for future immigrant teenagers.

      • VR 및 Desktop 기반 운전 시뮬레이터 사이의 실재감과 생리적 각성 반응에 관한 비교연구

        가상현실의 자연스러운 응용분야는 작업 훈련(Task Training) 분야이다. 이러한 작업 훈련 분야에서 가장 중요한 요소 중 하나는 각성 수준(Arousal Level)으로, 적절한 각성 수준은 과제 훈련의 효과를 결정 짓는다. 본 연구는 VR 환경과 Desktop 환경의 자동차 운전 시뮬레이터에서 피험자가 실험을 위하여 설계된 2 개의 시나리오를 수행하는 동안 피험자의 생리 신호를 측정하고, 실험 후 각 환경에 대한 주관적인 실재감을 측정하여, 두 실험 환경 사이에 피험자의 주관적인 실재감이 유의한 차이가 있는지 알아보고, 피험자가 수행한 시나리오와 시나리오에 종속된 실험 태스크에 대하여, 각성 수준을 평가할 때 유용한 지표로 활용되는 피부전도도(GSR) 반응이 피험자의 주관적인 실재감의 결과와 어떠한 관계를 보이는지 살펴 보았다. 실험 결과 두 실험 환경 사이의 피험자의 주관적인 실재감은 유의한 차이가 있었고, VR 환경에서 더 높게 나왔다. 그러나 두 실험 환경 사이의 피험자의 생리적 각성 반응은 실재감의 결과처럼 유의한 차이를 보여주지 못했으며, 오히려 시나리오에 종속된 실험 태스크에 따라서 유의한 차이를 보여주었다. 따라서 실재감과 피부전도도(GSR)측정에 의한 각성도 는 직접적인 관계가 적은 것으로 생각되며, 이는 피부전도도(GSR)를 실재감의 한 척도로 쓰기에는 적합하지 않을 수 있다는 것을 의미한다.
