      • Exploring Adolescent-parent Relationships in Asian American Immigrant Families: An Ecological Perspective

        The relationship between an adolescent and his/her parents is one of the most important relationships that can have a significant effect on adolescents' well-being and functioning. While there has been an increase in research on Asian American families in recent years, still much less is known about adolescent-parent relationships in these families. Asian American adolescents face some of the challenges that mainstream European American adolescents face, but their experiences are complicated by the cultural and immigration-related factors that have unique contribution to their relationships with their parents. As such, there is urgent need for research that identifies and provides a comprehensive understanding of factors that contribute to the experiences of Asian American immigrant families. The current paper provides a systematic look at adolescent-parent relationships in Asian American immigrant families using the Bronfenbrenner's ecological model. More specifically, this paper provides a succinct review of the literature on developmental issues, immigration, and culture-related factors that affect Asian American adolescent-parent relationships, and guided by Bronfenbrenner's ecological theory, an ecological framework of Asian American adolescent-parent relationships is proposed.

      • The Iterated Ritz Method: Basis, implementation and further development

        The Ritz method is known as very successful strategy for discretizing continuous problems, but it has never been used for solving systems of algebraic equations. The Iterated Ritz Method (IRM) is a novel iterative solver based on the discretized Ritz procedure applied at each iteration step. With an appropriate choice of coordinate vectors, the method may be efficient in linear, nonlinear and optimization problems. Additionally, some iterative methods can be explained as special cases of this approach, which helps to understand advantages and limitations of these methods and gives motivation for their improvement in sense of IRM. In this paper, some ideas for generation of efficient coordinate vectors are presented. The algorithm was developed and tested independently and then implemented into the open source program FEAP. Method has been successfully applied to displacement based (even ill-conditioned) models of structural engineering practice. With this original approach, a new iterative solution strategy has been opened.

        Flight Simulation Training Devices: Application, Classification, and Research

        Safe and efficient training using flight simulation training devices (FSTD) is one of the fundamental components of training in the commercial, military, and general aviation. When compared with the live training, the most significant benefits of ground trainers include improved safety and the reduced cost of a pilot training process. Flight simulation is a multidisciplinary subject that relies on several research disciplines which have a tendency to be investigated separately and in parallel with each other. This paper presents a comprehensive overview of the research within the FSTD domain with a motivation to highlight contributions from separate research topics from a general aspect, which is necessary as FSTD is a complex man–machine system. Application areas of FSTD usage are addressed, and the terminology used in the literature is discussed. Identification, classification, and overview of major research fields in the FSTD domain are presented. Specific characteristics of FSTD for fighter aircraft are discussed separately.

      • Crystal structure of the DNA binding domain of the transcription factor T-bet suggests simultaneous recognition of distant genome sites

        <P>The transcription factor T-bet (Tbox protein expressed in T cells) is one of the master regulators of both the innate and adaptive immune responses. It plays a central role in T-cell lineage commitment, where it controls the T(H)1 response, and in gene regulation in plasma B-cells and dendritic cells. T-bet is a member of the Tbox family of transcription factors; however, T-bet coordinately regulates the expression of many more genes than other Tbox proteins. A central unresolved question is how T-bet is able to simultaneously recognize distant Tbox binding sites, which may be located thousands of base pairs away. We have determined the crystal structure of the Tbox DNA binding domain (DBD) of T-bet in complex with a palindromic DNA. The structure shows a quaternary structure in which the T-bet dimer has its DNA binding regions splayed far apart, making it impossible for a single dimer to bind both sites of the DNA palindrome. In contrast to most other Tbox proteins, a single T-bet DBD dimer binds simultaneously to identical half-sites on two independent DNA. A fluorescence-based assay confirms that T-bet dimers are able to bring two independent DNA molecules into close juxtaposition. Furthermore, chromosome conformation capture assays confirm that T-bet functions in the direct formation of chromatin loops in vitro and in vivo. The data are consistent with a looping/synapsing model for transcriptional regulation by T-bet in which a single dimer of the transcription factor can recognize and coalesce distinct genetic elements, either a promoter plus a distant regulatory element, or promoters on two different genes.</P>

      • CD4 effector T cell differentiation is controlled by IL-15 that is expressed and presented <i>in trans</i>

        <P><B>Abstract</B></P> <P>T cells are both producers and consumers of cytokines, and autocrine cytokine signaling plays a critical role in T cell immunity. IL-15 is a homeostatic cytokine for T cells that also controls inflammatory immune responses. An autocrine role of T cell-derived IL-15, however, remains unclear. Here we examined IL-15 expression and signaling upon effector T cell differentiation in mice, and, surprisingly, found that CD4 T cells did not express IL-15. CD4 T cells lacked <I>Il15</I> gene reporter activity, did not contain IL-15 transcripts, and did not produce IL-15Rα, the proprietary IL-15 receptor required for IL-15 <I>trans</I>-presentation. Moreover, IL-15 failed to inhibit Th17 cell differentiation and failed to generate Foxp3<SUP>+</SUP> Treg cells <I>in vitro</I>. IL-2, which utilizes the same IL-2Rβ/γc receptor complex, however, successfully did so. Exogenous IL-15 only exerted bioactivity and controlled T cell differentiation when it was <I>trans</I>-presented by IL-15Rα. Consequently, IL-15Rα-bound IL-15, but not free IL-15, suppressed Th17 cell differentiation and induced Treg cell generation. Collectively, these results reveal the absence of an IL-15 autocrine loop in CD4 T cells and strongly suggest that IL-15 <I>trans</I>-presentation by non-CD4 T cells is the primary mechanism via which IL-15 controls CD4 effector T cell differentiation.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Unlike most other γc cytokines, IL-15 is not produced by activated CD4 T cells. </LI> <LI> Free IL-15 phosphorylates STAT5 but does not affect CD4 T cell differentiation. </LI> <LI> IL-15Rα-bound IL-15, but not free IL-15, suppresses Th17 cell generation. </LI> <LI> IL-15 <I>trans</I>-presentation is required to control CD4 effector T cell differentiation. </LI> </UL> </P>

      • Activated T Cells Secrete an Alternatively Spliced Form of Common γ-Chain that Inhibits Cytokine Signaling and Exacerbates Inflammation

        The common γ-chain (γc) plays a central role in signaling by IL-2 and other γc-dependent cytokines. Here we report that activated T cells produce an alternatively spliced form of γc mRNA that results in protein expression and secretion of the γc extracellular domain. The soluble form of γc (sγc) is present in serum and directly binds to IL-2Rβ and IL-7Rα proteins on T cells to inhibit cytokine signaling and promote inflammation. sγc suppressed IL-7 signaling to impair naive T cell survival during homeostasis and exacerbated Th17-cell-mediated inflammation by inhibiting IL-2 signaling upon T cell activation. Reciprocally, the severity of Th17-cell-mediated inflammatory diseases was markedly diminished in mice lacking sγc. Thus, sγc expression is a naturally occurring immunomodulator that regulates γc cytokine signaling and controls T cell activation and differentiation.

      • Breast Cancer in Bedouin-Arab Patients in Southern Israel: Epidemiologic and Biologic Features in Comparison with Jewish Patients

        Background: Breast cancer (BC) is the most frequent cancer type, and the leading cause of death from cancer among women in Israel. The Bedouin-Arab (BA) population in southern Israel is characterized by a high rate of consanguinity, common hereditary disorders, and transition from a semi-nomadic, traditional society to a more sedentary and urbanized society. In this hospital-based study, the demographic and the clinicopathological characteristics of BC in BA were compared with Jewish patients. Materials and Methods: 85 BA patients treated at the Soroka Medical Center, Beer Sheba, during the years 2004-2012, were studied and compared with 180 consecutive Jewish patients treated during the year 2007. Clinicopathological features compared included age, menopausal state, number of births, a history of BC in first-degree relatives, tumor size (T), extent of lymph-node involvement (N), distant metastases (M), stage, grade, estrogen and progesterone receptor (ER/PR), and Her2 status. Types of treatment, relapse rate and site, as well as outcome were also studied. Cox's regression models were applied for studying disease-free, and overall survival. Results: Compared with Jewish patients, BA patients were younger (average age $49{\pm}12$ yrs vs $59{\pm}13$, p<0.001), had a lower rate of BC in first-degree relatives (p<0.001), and a larger number of births ($6{\pm}4.2$ vs $2.5{\pm}1.9$, p<0.001). BA patients had larger tumors (p=0.02), more extensive lymph-node involvement (p=0.002), and more advanced stage (p=0.003). Grade, ER, PR, and Her2 status were similar in the two ethnic groups. Relapse type was most commonly systemic in BA patients (p=0.05), and loco-regional in Jewish patients (p=0.02). Median survival was 63, and 35 months for Jewish and BA patients, respectively (log-rank test, p=0.02). In Cox multivariate analysis, stage and PR status (HR-0.14, p<0.0001; HR-3.11, p=0.046), but not ethnicity, influenced overall survival. Conclusions: BC presents a decade earlier, and with more advanced disease in BA compared with Jewish patients. Biologic parameters including grade, ER, PR, and Her2 status were similar in both groups. Although prognosis was worse in BA than in Jewish patients, it was affected only by stage and PR status, but not by ethnicity.
