The aim of this study was to isolate microorganisms from half milk samples of dairy goats by California mastitis test (CMT) during the lactation period and to further investigate the susceptibility of isolated organisms to antimicrobial drugs. From a total of 235 half milk samples with CMT scores of 2 or above from 366 dairy goats distributed throughout Jeonnam province, microorganisms were isolated from 198 (83.5%) samples either singly (99.0%) or in combination (1.0%). The most prevalent microorganism was the coagulase-negative Staphylococcus spp., (44.4%, n=88) followed by Staphylococcus aureus (24.2%, n=48), Escherichia coli (11.1%, n=22) and Streptococcus spp. (7.6%, n=15). Isolated bacteria also included Bacillus spp. (2.5%, n=5), Pseudomonas spp. (2.5%, n=5), Micrococcus spp. (1.5%, n=3), Corynebacterium spp. (1.5%, n=3), Enterococcus facium (1.0%, n=2), Morganella morganii (0.5%, n=1) and Streptococcus agalactiae (0.5%, n=1). During the summer season, a high prevalence of all micro-organisms were observed in which Staphylococcus spp. (30.8%), Escherichia coli (8.6%), and Streptococcus spp. (5.6%) were among the most prevalent bacteria isolated. Staphylococcus spp. was also shown to be high in the winter (21.7%). In most samples, the presence of bacterial pathogens in goat milk led to the increase in the total somatic cell count (SCC). Most of the half milk samples of dairy goats with bacterial contamination showed SCC of ≥1×10(6) cells/ml (90.4%). Minor pathogens (11.4%) were more detected from milk samples with SCC of <1×10(6) cells/ml than major pathogens (4.1%), while the major pathogens tended to be higher from samples with SCC of ≥3×10(6) cells/ml. Susceptibility of these bacteria to 12 antimicrobial agents was tested by the Kirby-Bauer disc diffusion method. Results indicated that more than 90% of bacteria isolated from CMT 2+ dairy goat half milk samples were susceptible to trimethoprim/sulfamethoxazole, amoxicillin/clavulanic, enrofloxacin and cephalothin while they were resistant to tetracycline (44.7%).
Fabrication of Bi-2212/SrSO<sub>4</sub> Composite Superconductors by Melting Powder Mixtures

<P>We fabricated Bi-2212/SrSO4 composite superconductors by the melt casting process and evaluated the effects of the powder mixing method and melting temperature on their microstructure and superconducting properties. In the melt casting process, the Bi-2212 powders were mixed with SrSO4 by hand-mixing (HM) and planetary ball milling (PBM) and then the powder mixtures were melted at 1100°C~1200°C, solidified, and annealed. We found that the powder mixture prepared by PBM was finer and more homogeneously mixed than that prepared by HM, resulting in more homogeneous microstructure and smaller SrSO4 and second phases after annealing. The critical current (Ic) also varied significantly with the powder mixing method and the melting temperature. The Ic of the annealed rod prepared by PBM was 193 A at 77 K when melted at 1100°C, which is higher than that of the annealed rod prepared by HM (132 A). This enhancement in the Ic value for the former is considered to be due to its more uniform microstructure.</P>

<P><B>Abstract</B></P> <P>The tripartite motif-containing (TRIM) proteins are involved in a wide range of cellular processes, and the role of TRIM1 in immunity has been explored. However, fundamental studies on fish TRIM1 are lacking. In this study, we cloned and characterized TRIM1 cDNA from the Korean rose bitterling, <I>Rhodeus uyekii</I> (RuTRIM1). Two RuTRIM1 isoforms (RuTRIM1-X1 and RuTRIM1-X2) were identified. The coding sequence (CDS) of RuTRIM1-X1 comprised 2157 bp encoding a 718-aa protein, and the CDS of RuTRIM1-X2 comprised 1929 bp encoding a 642-aa protein. Both RuTRIM1 isoforms contained a RING finger domain, B-box 1, B-box 2, coiled-coil domain, COS box, FN3 motif, and PRY/SPRY domain. The deduced RuTRIM1-X1 and RuTRIM1-X2 proteins had high amino acid identity (76.27–98.89%) with orthologs from various other species, and a phylogenetic tree was constructed. RuTRIM1-X1 and RuTRIM1-X2 mRNA were expressed in all tissues examined, with the highest expression levels detected in the hepatopancreas. During early development, RuTRIM1-X1 and RuTRIM1-X2 mRNA levels changed differently from the gastrula period to the first feeding stage. An <I>in vivo</I> ubiquitination assay showed that RuTRIM1 exhibited RING-dependent E3 ubiquitin ligase activity, mainly by comparing RuTRIM1-X2 to RuTRIM1-X1. The subcellular localization of the two RuTRIM1 protein isoforms was characterized, revealing that they formed aggregates in cytoplasmic bodies in Raw264.7 cells. Interferon-γ/lipopolysaccharide-induced nuclear factor-κB signaling was negatively regulated by RuTRIM1-X1 and RuTRIM1-X2, and the negative effect was reversed in RING deletion mutants. To our knowledge, this is the first study to characterize fish TRIM1, which may play a role in the inflammatory response.</P> <P><B>Highlights</B></P> <P> <UL> <LI> We isolated two TRIM1 isoforms (RuTRIM1-X1 and RuTRIM1-X2) from <I>Rhodeus uyekii.</I> </LI> <LI> RuTRIM1 cDNAs encode polypeptides with the RBCC motif. </LI> <LI> RuTRIM1 proteins share 76.27–98.89% homology with orthologs from other species. </LI> <LI> The subcellular localization and Ub ligase activity of the two RuTRIM1 protein isoforms were characterized. </LI> <LI> RuTRIM1-X1 and RuTRIM1-X2 negatively regulate the IFN-γ/LPS-induced NF-κB signaling. </LI> </UL> </P>

Malignant pleural effusion (MPE) and blood samples can be used as a practical source for detection of epidermal growth factor receptor (EGFR) mutations in patients with advanced non-small cell lung cancer. We compared EGFR mutation status of cell blocks, cell-free fluid of MPE, and plasma from patients with lung adenocarcinoma. We obtained paired samples of MPE and plasma from 14 pathologically-confirmed lung adenocarcinoma patients. Peptide nucleic acid (PNA)-mediated real-time polymerase chain reaction (RT-PCR) clamping was performed for determination of EGFR mutation status. EGFR mutations were detected in five (35.7%) cell blocks of MPE, which showed results identical to those of the corresponding cell-free fluid, whereas mutations were detected in the plasma of only two (40.0%) of the five patients. Of seven patients treated with EGFR tyrosine kinase inhibitors (TKIs), EGFR mutations were detected in cell blocks, cell-free fluid of MPE, and plasma for only one of the four patients who responded to EGFR TKIs, while mutations were detected only in cell blocks of MPE and cell-free fluid of the three remaining patients. Our results suggest that detection of EGFR mutations in cell-free pleural fluid from lung adenocarcinoma patients using highly sensitive methods may be feasible, but that analysis of free plasma may lead to undetected mutations and misdiagnosis. Purpose: The objective of the present study was to investigate and analyze the influence of physical activities in the zero hour class, a class preceding regular classes, on physical fitness and attitude toward learning in high school students. Method: Sixty high school students were divided into two groups: exercise group (n=30) and non-exercise group (n=30). Subjects in exercise group had done an exercise program 42 min/day, 3 days/week during 12 weeks (40-50 %VO2max for 1-6 weeks and 50-60% %VO2max for 7-12 weeks). The exercise program applied to exercise group consisted of warm up, main exercise, and cool down. And the main exercise was comprised of a combination of circuit-based resistance training and aerobic training. We measured physical fitness (muscular strength, flexibility, power, and cardiorespiratory endurance) and attitude toward learning (attention concentration, learning methods, and self learning) three times (pre, during, post exercise program). Repeated ANOVA analysis was used for the data analysis with the significant level α=.05. Result: As the results, all elements of physical fitness showed the significant improvement only in exercise group (p<.01). While attention concentration showed significant increase (p<.01), other two elements of attitude toward learning showed no significant difference. Conclusion: In summary, physical activity program in the zero hour class showed positive effects on physical fitness and attitude toward learning in high school students. Therefore it is considered that the application of physical activities to the zero class could help to educe potential learning ability of high school students.

Serine-arginine-rich nuclear protein LUC7L plays an important role in the regulation of myogenesis in mice. In the present study, we isolated and characterized the Korean rose bitterling Rhodeus uyekii Luc7l cDNA, designated RuLuc7l. The RuLuc7l cDNA is 1,688 bp long and encodes a 364-amino-acid polypeptide containing serine/arginine-rich region at the C-terminus. The deduced RuLuc7l protein has high amino acid identity (71-97%) with those of other species including human. Phylogenetic analysis revealed that RuLUC7L clustered with fish LUC7L proteins. The expression of RuLuc7l mRNA was high in the brain, kidney, and stomach of Korean rose bitterling. Expression of the RuLuc7l mRNA was detected from 1 day post-fertilization (dpf) and moderately increased until 21 dpf during the early development. Further investigations are required to elucidate the functional role of RuLUC7L in myogenesis in R. uyekii.
Mitochondrial Complexes I and II Are More Susceptible to Autophagy Deficiency in Mouse β-Cells

<P><B>Background</B></P><P>Damaged mitochondria are removed by autophagy. Therefore, impairment of autophagy induces the accumulation of damaged mitochondria and mitochondrial dysfunction in most mammalian cells. Here, we investigated mitochondrial function and the expression of mitochondrial complexes in autophagy-related 7 (<I>Atg7</I>)-deficient β-cells.</P><P><B>Methods</B></P><P>To evaluate the effect of autophagy deficiency on mitochondrial function in pancreatic β-cells, we isolated islets from <I>Atg7</I><SUP>F/F</SUP>:RIP-<I>Cre</I>+ mice and wild-type littermates. Oxygen consumption rate and intracellular adenosine 5'-triphosphate (ATP) content were measured. The expression of mitochondrial complex genes in <I>Atg7</I>-deficient islets and in β-TC6 cells transfected with si<I>Atg7</I> was measured by quantitative real-time polymerase chain reaction.</P><P><B>Results</B></P><P>Baseline oxygen consumption rate of <I>Atg7</I>-deficient islets was significantly lower than that of control islets (<I>P</I><0.05). Intracellular ATP content of <I>Atg7</I>-deficient islets during glucose stimulation was also significantly lower than that of control islets (<I>P</I><0.05). By Oxygraph-2k analysis, mitochondrial respiration in <I>Atg7</I>-deficient islets was significantly decreased overall, although state 3 respiration and responses to antimycin A were unaffected. The mRNA levels of mitochondrial complexes I, II, III, and V in <I>Atg7</I>-deficient islets were significantly lower than in control islets (<I>P</I><0.05). Down-regulation of <I>Atg7</I> in β-TC6 cells also reduced the expression of complexes I and II, with marginal significance (<I>P</I><0.1).</P><P><B>Conclusion</B></P><P>Impairment of autophagy in pancreatic β-cells suppressed the expression of some mitochondrial respiratory complexes, and may contribute to mitochondrial dysfunction. Among the complexes, I and II seem to be most vulnerable to autophagy deficiency.</P>
