      • Preparation and Characterization of Monosized Germanium Particles by Pulsated Orifice Ejection Method

        Monosized germanium micro particles are prepared by a newly developed Pulsated Orifice Ejection Method. The obtained particles are categorized into two kinds of the microstructures as refined and coarse ones. The morphological difference is estimated to be determined by the undercooling level during nucleation. Actually, the increase in the temperature of the melt was effective in coarsening the microstructure, because the temperature of the melt intensely relates to the undercooling level. The transition temperature of coarse and refined microstructures is found to be 1300-1350K. Furthermore, a triggered nucleation could improve the crystallinity of the particles in the short separation.

        Mono-sized silicon particles were effectively fabricated by a novel way named pulsated orifice ejection method (POEM). The particles are with very narrow particles size distribution and very small standard deviation of mean particle size. There are two different types spherical silicon particles were found. One consists of many grains mainly in random boundaries. The other consists of two or three grains with only twin orientation relationships, even single crystal in cross-section was also found within this type of spherical silicon particles.

        TP53 variants in p53 signatures and the clonality of STICs in RRSO samples

        Objective: Precursor lesions may be identified in fallopian tube tissue after risk-reducing salpingo-oophorectomy (RRSO) in patients with pathogenic variants of . Serous tubal intraepithelial carcinoma (STIC) is considered a precursor of high-grade serous carcinoma, whereas the significance of the p53 signature remains unclear. In this study, we investigated the relationship between the p53 signature and the risk of ovarian cancer. Methods: We analyzed the clinicopathological findings and conducted DNA sequencing for variants of p53 signatures and STIC lesions isolated using laser capture microdissectionin 13 patients with pathogenic variants of who underwent RRSO and 17 control patients with the benign gynecologic disease. Results: pathogenic variants were detected significantly higher in RRSO group than control (p<0.001). No difference in the frequency of p53 signatures were observed between groups (53.8% vs 29.4%; p=0.17). sequencing and next-generation sequencing analysis in a patient with STIC and occult cancer revealed 2 mutations causing different p53 staining for STICs and another mutation shared between STIC and occult cancer. Conclusion: The sequence analysis for revealed 2 types of p53 signatures, one with a risk of progression to STIC and ovarian cancer with pathological variants in and the other with a low risk of progression without pathological variants in as seen in control.

        Current status of hereditary breast and ovarian cancer practice among gynecologic oncologists in Japan: a nationwide survey by the Japan Society of Gynecologic Oncology (JSGO)

        Objective: The practices pertaining to hereditary breast and ovarian cancer (HBOC) in Japan have been rapidly changing owing to the clinical development of poly(ADP-ribose) polymerase inhibitors, the increasing availability of companion diagnostics, and the broadened insurance coverage of HBOC management from April 2020. A questionnaire of gynecologic oncologists was conducted to understand the current status and to promote the widespread standardization of future HBOC management. Methods: A Google Form questionnaire was administered to the members of the Japan Society of Gynecologic Oncology. The survey consisted of 25 questions in 4 categories: respondent demographics, HBOC management experience, insurance coverage of HBOC management, and educational opportunities related to HBOC. Results: A total of 666 valid responses were received. Regarding the prevalence of HBOC practice, the majority of physicians responded in the negative and required human resources, information sharing and educational opportunities, and expanded insurance coverage to adopt and improve HBOC practice. Most physicians were not satisfied with the educational opportunities provided so far, and further expansion was desired. They remarked on the psychological burdens of many HBOC managements. Physicians reported these burdens could be alleviated by securing sufficient time to engage in HBOC management, creating easy-to-understand explanatory material for patients, collaboration with specialists in genetic medicine, and educational opportunities. Conclusion: Gynecologic oncologists in Japan are struggling to deal with psychological burdens in HBOC practice. To promote the clinical practice of HBOC management, there is an urgent need to strengthen human resources and improve educational opportunities, and expand insurance coverage for HBOC management.

      • LAMC1 expression patterns and associated clinical features of endometrial cancer

        Objective: Laminins are one of the major components of the extracellular matrix. Recent studies have indicated the relationship between specific laminin expression profiles and cancer progression. Upregulation of LAMC1, which encodes laminin-γ1, is of particular note because this is related to aggressive behavior of cancer cells of various origins. This study aimed to analyze the influence of LAMC1 expression on the clinical features of endometrial cancer. Methods: The subjects were patients with endometrial cancer (EC), atypical endometrial hyperplasia complex (AEHC) or normal endometrium (NE) who underwent hysterectomy during January 2008 and December 2011 at our institute. The study included 100 cases of EC, 9 of AEHC, and 16 NE. LAMC1 immunohistochemical staining was evaluated as negative or positive by two independent evaluators blinded from clinical information. Patient characteristics and clinical outcomes were obtained retrospectively from clinical records. This study was approved by our institutional review board. Results: LAMC1 expression was significantly elevated in EC compared to AEHC and NE (p<0.05). Endometrioid (EM) grade 3, serous and clear cell adenocarcinoma indicated frequent LAMC1 expression compared to EM grade 1 and 2 (85.2% vs. 51.3%, p<0.05). Patients with advanced FIGO surgical stage (stage III and IV) and positive lymphovascular space invasion had a significantly higher LAMC1 expression rate compared to early stage cases (83.7% vs. 63.2%, p<0.05) and cases without lymphovascular space invasion (86.3% vs. 61.0%, p<0.05), respectively. Kaplan-Meier analysis revealed that LAMC1 positive cases had a significantly shorter progression-free survival (p<0.05) and a tendency to have a reduced overall survival (p=0.052). Conclusion: In endometrial cancer, LAMC1 expression is related to aggressive tumor behavior and reduced progression free survival.
